    Telomere length predicts for outcome to FCR chemotherapy in CLL

    We have previously shown that dividing patients with CLL into those with telomeres inside the fusogenic range (TL-IFR) and outside the fusogenic range (TL-OFR) is powerful prognostic tool. Here, we used a high-throughput version of the assay (HT-STELA) to establish whether telomere length could predict for outcome to fludarabine, cyclophosphamide, rituximab (FCR)-based treatment using samples collected from two concurrent phase II studies, ARCTIC and ADMIRE (n = 260). In univariate analysis, patients with TL-IFR had reduced progression-free survival (PFS) (P < 0.0001; HR = 2.17) and shorter overall survival (OS) (P = 0.0002; HR = 2.44). Bifurcation of the IGHV-mutated and unmutated subsets according to telomere length revealed that patients with TL-IFR in each subset had shorter PFS (HR = 4.35 and HR = 1.48, respectively) and shorter OS (HR = 3.81 and HR = 2.18, respectively). In addition, the OS of the TL-OFR and TL-IFR subsets were not significantly altered by IGHV mutation status (P = 0.61; HR = 1.24 and P = 0.41; HR = 1.47, respectively). In multivariate modeling, telomere length was the dominant co-variable for PFS (P = 0.0002; HR = 1.85) and OS (P = 0.05; HR = 1.61). Taken together, our data suggest that HT-STELA is a powerful predictor of outcome to FCR-based treatment and could be used to inform the design of future risk-adapted clinical trials

    Movements and spatial use of odontocetes in the western main Hawaiian Islands: results from satellite-tagging and photo-identification off Kaua‘i and Ni‘ihau in July/August 2011

    Although considerable information is available on residency patterns and spatial use of odontocetes in the eastern half of the Hawai‘i Range Complex (HRC), much less is known about odontocetes in the western half of the HRC. In the second year of a three-year effort in the western main Hawaiian Islands we undertook surveys off Kaua‘i and Ni‘ihau in July/August 2011, to examine spatial use and residency patterns using satellite tags, to provide visual verification of acoustically-detected odontocetes on the Pacific Missile Range Facility (PMRF), and to obtain individual identification photographs and biopsy samples for assessment of population identity and structure. During 18 days of field effort we covered 1,972 km of trackline and had 65 encounters with five species of odontocetes. Twenty-four of the encounters, of three species, were cued by acoustic detections from the Marine Mammal Monitoring on Navy Ranges (M3R) system, thus providing species verifications for future use of the M3R system on the PMRF range. During the 65 encounters we obtained 22,645 photos for individual and species identification, and collected 48 biopsy samples for genetic analyses. One encounter with a group of four killer whales was only the second encounter with this species in 12 years of directed field surveys in Hawaiian waters. Photos from that encounter were compared to our photo-identification catalog but no matches were found, further suggesting that there is no population of this species resident to the Hawaiian Islands. There were three encounters with a lone pantropical spotted dolphin, each time in association with a group of spinner dolphins. Photos of this individual matched to a spotted dolphin identified off Kaua‘i in 2004 and in 2005, both times with spinner dolphins, suggesting this individual may be part of a long-term association with spinner dolphins. Four satellite tags were deployed; three on rough-toothed dolphins and one on a bottlenose dolphin. These are the first tag deployments on either species in Hawaiian waters and the first deployments of satellite tags on free-ranging rough-toothed dolphins anywhere in the world. Rough-toothed dolphin tag data were obtained over periods from 7.6 to 18.5 days. Over these periods the three rough-toothed dolphins moved cumulative horizontal distances ranging from 573 to 1,295 km, yet remained an average distance from the tagging locations of from 10.4 to 13.9 km. Median depths used by the three rough-toothed dolphins ranged from 816 to 1,107 m, with median distance from shore ranging from 11.6 to 12.2 km. Two of the three individuals had been previously photo-identified off Kaua‘i (in 2007 or 2008), and all link by association with the resident population from Kaua‘i and Ni‘ihau. Movement and habitat use data were obtained over a 34-day period for the satellite-tagged bottlenose dolphin. During this time the individual remained associated with the island of Kaua‘i using waters with a median depth of 82 m. Although this individual had not been previously photo-identified, others from the group it was in had been previously documented off Kaua‘i and/or Ni‘ihau in 2003-2005, suggesting it is part of the island-resident population.     Telomere dysfunction accurately predicts clinical outcome in chronic lymphocytic leukaemia, even in patients with early stage disease

    © 2014 John Wiley & Sons Ltd. Defining the prognosis of individual cancer sufferers remains a significant clinical challenge. Here we assessed the ability of high-resolution single telomere length analysis (STELA), combined with an experimentally derived definition of telomere dysfunction, to predict the clinical outcome of patients with chronic lymphocytic leukaemia (CLL). We defined the upper telomere length threshold at which telomere fusions occur and then used the mean of the telomere 'fusogenic' range as a prognostic tool. Patients with telomeres within the fusogenic range had a significantly shorter overall survival (P  <  0·0001; Hazard ratio [HR] = 13·2, 95% confidence interval [CI]  = 11·6-106·4) and this was preserved in early-stage disease patients (P  <  0·0001, HR=19·3, 95% CI = 17·8-802·5). Indeed, our assay allowed the accurate stratification of Binet stage A patients into those with indolent disease (91% survival at 10 years) and those with poor prognosis (13% survival at 10 years). Furthermore, patients with telomeres above the fusogenic mean showed superior prognosis regardless of their IGHV mutation status or cytogenetic risk group. In keeping with this finding, telomere dysfunction was the dominant variable in multivariate analysis. Taken together, this study provides compelling evidence for the use of high-resolution telomere length analysis coupled with a definition of telomere dysfunction in the prognostic assessment of CLL

Telomeres can become shorter due to cell division and specific lifestyle factors. Critically shortened telomeres are linked to cellular dysfunction, senescence and aging. A number of studies have used low resolution techniques to assess telomere length in the placenta. In this study, we applied Single Telomere Length Analysis (STELA) which provides high-resolution chromosome specific telomere length profiles to ask whether we could obtain more detailed information on the length of individual telomeres in the placenta. Methods Term placentas (37–42 weeks) were collected from women delivering at University Hospital of Wales or Royal Gwent Hospital within 2 h of delivery. Multiple telomere-length distributions were determined using STELA. Intraplacental variation of telomere length was analysed (N = 5). Telomere length distributions were compared between labouring (N = 10) and non-labouring (N = 11) participants. Finally, telomere length was compared between female (N = 17) and male (N = 20) placenta. Results There were no significant influences of sampling site, mode of delivery or foetal sex on the telomere-length distributions obtained. The mean telomere length was 7.7 kb ranging from 5.0 kb to 11.7 kb across all samples (N = 42) and longer compared with other human tissues at birth. STELA also revealed considerable telomere length heterogeneity within samples. Conclusions We have shown that STELA can be used to study telomere length homeostasis in the placenta regardless of sampling site, mode of delivery and foetal sex. Moreover, as each amplicon is derived from a single telomeric molecule, from a single cell, STELA can reveal the full detail of telomere-length distributions, including telomeres within the length ranges observed in senescent cells. STELA thus provides a new tool to interrogate the relationship between telomere length and pregnancy complications linked to placental dysfunction

    Extraordinary Late-Time Infrared Emission of Type IIn Supernovae

    Near-Infrared (NIR) observations are presented for five Type IIn supernovae (SN 1995N, SN 1997ab, SN 1998S, SN 1999Z, and SN 1999el) that exhibit strong infrared excesses at late times (t >= 100 d). H- and K-band emission from these objects is dominated by a continuum that rises toward longer wavelengths. The data are interpreted as thermal emission from dust, probably situated in a pre-existing circumstellar nebula. The IR luminosities implied by single temperature blackbody fits are quite large,> 10^(41 - 42) erg s^-1, and the emission evolves slowly, lasting for years after maximum light. For SN 1995N, the integrated energy release via IR dust emission was 0.5 -- 1 * 10^50 erg. A number of dust heating scenarios are considered, the most likely being an infrared echo poweredby X-ray and UV emissions from the shock interaction with a dense circumstellar medium.Comment: 14 Pages, 3 Figures, Accecpted for publication in The Astrophysical Journa
