    Character of adsorbed bovine serum albumin from adsorption enthalpies

    Heats of adsorption of BSA onto polystyrene and polycarbonate were determined microcalorimetrically. The polymers were coated onto 50 nm alumina particles which were immersed in buffer for the measurements. Experimental variations to isolate the various component reactions of the integral heats included adsorption of native and denatured BSA from solution, adsorption onto substrates precoated with BSA, varying the thicknesses of adsorbed BSA by varying concentrations of the adsorbing solutions, and determining the denaturation energy by solution of the solid native and denatured BSA into a denaturing solvent.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/26061/1/0000135.pd

    Effect of low-dose irradiation on the surface properties of poly[(trifluoroethoxy)(telomer fluoro alkoxy)phosphazene]

    The effect of low-dose ultraviolet and 3, irradiation on the surface and bulk properties of poly[(trifluoroethoxy)(telomer fluoro alkoxy)phosphazene], was found to initially result in an increased side chain mobility followed by a decreased mobility. The initial increase is attributed to partial cleavage of selected side chains followed by a decreased mobility as a result of interside chain crosslinking. These above effects and the solid state characterization of the polymers were documented by dynamic mechanical analysis, difference dielectric spectroscopy, electron microscopy, electron spectroscopy for chemical analysis, attenuated total internal reflection spectroscopy, contact angle measurements, differential scanning calorimetry, gel permeation chromatography, tensile stress strain, and mass spectroscopy.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/24691/1/0000110.pd

    Neurological complications are common in COVID-19. Although SARS-CoV-2 has been detected in patients' brain tissues, its entry routes and resulting consequences are not well understood. Here, we show a pronounced upregulation of interferon signaling pathways of the neurovascular unit in fatal COVID-19. By investigating the susceptibility of human induced pluripotent stem cell (hiPSC)-derived brain capillary endothelial-like cells (BCECs) to SARS-CoV-2 infection, we found that BCECs were infected and recapitulated transcriptional changes detected in vivo. While BCECs were not compromised in their paracellular tightness, we found SARS-CoV-2 in the basolateral compartment in transwell assays after apical infection, suggesting active replication and transcellular transport of virus across the blood-brain barrier (BBB) in vitro. Moreover, entry of SARS-CoV-2 into BCECs could be reduced by anti-spike-, anti-angiotensin-converting enzyme 2 (ACE2)-, and anti-neuropilin-1 (NRP1)-specific antibodies or the transmembrane protease serine subtype 2 (TMPRSS2) inhibitor nafamostat. Together, our data provide strong support for SARS-CoV-2 brain entry across the BBB resulting in increased interferon signaling

    Thrombogenesis: An epitaxial phenomena. I

    A semiempirical epitaxial model has been derived which correlates and interrelates the surface free energy, ultrastructural morphology, surface charge, surface chemistry, and surface molecular motions of a model triblock copolymer to thrombogenesis. This paper addresses the aspect of: (1) ultrastructure order versus disorder, (2) primary and secondary molecular motions, (3) surface and side chain chemistry, (4) thrombogenesis, and (5) the derived epitaxial model based on the above observations. This model can be extrapolated to include and predict the relative thrombogenic responses of various crystalline and semicrystalline polymeric substrates, i.e., aliphatic polyolefins, polyurethanes, and polypeptides.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/37995/1/820150210_ftp.pd
