    Research Notes : United States : Superoxide dismutase (SOD) and tetrazolium oxidase (TO) zymograms observed in gradient PAGE gels and preliminary inheritance data for type 3 zymograms

    We tested gradient PAGE gels for their ability to resolve isozymes with a specific SOD (EC 1.15.1.1) stain as well as with a nonspecific TO stain. In particular, we wanted to test gradient gels for use in scoring F2 seeds segregating for the cultivar-specific type 1 and type 3 TO zymogram patterns first reported by Gorman and Kiang (1977). TO electrophoretic bands are caused by any number of enzymes (particularly SOD) that prevent the spontaneous reduction of tetrazolium dyes in the electron-transfer staining systems used to detect dehydrogenases

    Scenarios of domain pattern formation in a reaction-diffusion system

    We performed an extensive numerical study of a two-dimensional reaction-diffusion system of the activator-inhibitor type in which domain patterns can form. We showed that both multidomain and labyrinthine patterns may form spontaneously as a result of Turing instability. In the stable homogeneous system with the fast inhibitor one can excite both localized and extended patterns by applying a localized stimulus. Depending on the parameters and the excitation level of the system stripes, spots, wriggled stripes, or labyrinthine patterns form. The labyrinthine patterns may be both connected and disconnected. In the the stable homogeneous system with the slow inhibitor one can excite self-replicating spots, breathing patterns, autowaves and turbulence. The parameter regions in which different types of patterns are realized are explained on the basis of the asymptotic theory of instabilities for patterns with sharp interfaces developed by us in Phys. Rev. E. 53, 3101 (1996). The dynamics of the patterns observed in our simulations is very similar to that of the patterns forming in the ferrocyanide-iodate-sulfite reaction.Comment: 15 pages (REVTeX), 15 figures (postscript and gif), submitted to Phys.     Land-to-sea connectivity: Linking human-derived terrestrial subsidies to subtidal habitat change on open rocky coasts

All rights reserved.Spatial subsidies are considered strong where differences in resource availability between donor and recipient systems are greatest. We tested whether human activities on land can increase subsidies of terrigenous nitrogen to open rocky coasts and whether these differences can predict apparent deforestation of kelp forests. We first identified landscape-scale variation in the human-mediated transfer of dissolved inorganic nitrogen (DIN) from natural, agricultural, and urban catchments to temperate coasts after episodes of rainfall. Compared to natural catchments, subsidies of DIN were on average 8–407 times greater in urban catchments, and 1–63 times greater in agricultural catchments. Urban derived nitrogen was attributed to the release of sewage effluent, as delineated by ή15N isotopic values of transplanted algae. Having made this link, we then assessed whether this catchment-scale variation may account for variation in structure of subtidal habitats, particularly as related to theory of nutrient-driven shifts of habitat from perennial (i.e., canopy-forming algae) to opportunistic species (i.e., turf-forming algae). We not only detected patterns consistent with this theory, but also established that the size and total proportion of patches of turf-forming algae were greater where the ratio of donor : recipient nitrogen loads was greater (i.e., size of subsidy). An important realization was that deforestation may be more strongly related to variation in the size of subsidy rather than size of human populations, particularly among urban catchments. These data directly link the type of human activity within catchments to the modification of land-to-sea subsidies and indirectly support theory that predicts terrestrial inputs to have greater ecological effects where the disparity in resource availability between donor and recipient is exacerbated. Our evidence has been used by coastal managers to reconsider their management of coastal systems and has subsequently contributed to new water-recycling policy and initiatives.Daniel Gorman, Bayden D.     Rituximab monitoring and redosing in pediatric neuromyelitis optica spectrum disorder.

    Abstract OBJECTIVE: To study rituximab in pediatric neuromyelitis optica (NMO)/NMO spectrum disorders (NMOSD) and the relationship between rituximab, B cell repopulation, and relapses in order to improve rituximab monitoring and redosing. METHODS: Multicenter retrospective study of 16 children with NMO/NMOSD receiving 652 rituximab courses. According to CD19 counts, events during rituximab were categorized as "repopulation," "depletion," or "depletion failure" relapses (repopulation threshold CD19 6510 7 10(6) cells/L). RESULTS: The 16 patients (14 girls; mean age 9.6 years, range 1.8-15.3) had a mean of 6.1 events (range 1-11) during a mean follow-up of 6.1 years (range 1.6-13.6) and received a total of 76 rituximab courses (mean 4.7, range 2-9) in 42.6-year cohort treatment. Before rituximab, 62.5% had received azathioprine, mycophenolate mofetil, or cyclophosphamide. Mean time from rituximab to last documented B cell depletion and first repopulation was 4.5 and 6.8 months, respectively, with large interpatient variability. Earliest repopulations occurred with the lowest doses. Significant reduction between pre- and post-rituximab annualized relapse rate (ARR) was observed (p = 0.003). During rituximab, 6 patients were relapse-free, although 21 relapses occurred in 10 patients, including 13 "repopulation," 3 "depletion," and 4 "depletion failure" relapses. Of the 13 "repopulation" relapses, 4 had CD19 10-50 7 10(6) cells/L, 10 had inadequate monitoring ( 641 CD19 in the 4 months before relapses), and 5 had delayed redosing after repopulation detection. CONCLUSION: Rituximab is effective in relapse prevention, but B cell repopulation creates a risk of relapse. Redosing before B cell repopulation could reduce the relapse risk further. CLASSIFICATION OF EVIDENCE: This study provides Class IV evidence that rituximab significantly reduces ARR in pediatric NMO/NMOSD. This study also demonstrates a relationship between B cell repopulation and relapses

    The complications of ‘hiring a hubby’: gender relations and the commoditisation of home maintenance in New Zealand

    This paper examines the commoditization of traditionally male domestic tasks through interviews with handymen who own franchises in the company ‘Hire a Hubby’ in New Zealand and homeowners who have paid for home repair tasks to be done. Discussions of the commoditization of traditionally female tasks in the home have revealed the emotional conflicts of paying others to care as well as the exploitative and degrading conditions that often arise when work takes place behind closed doors. By examining the working conditions and relationships involved when traditionally male tasks are paid for, this paper raises important questions about the valuing of reproductive labour and the production of gendered identities. The paper argues that while working conditions and rates of pay for ‘hubbies’ are better than those for people undertaking commoditized forms of traditionally female domestic labour, the negotiation of this work is still complex and implicated in gendered relations and identities. Working on the home was described by interviewees as an expression of care for family and a performance of the ‘right’ way to be a ‘Kiwi bloke’ and a father. Paying others to do this labour can imply a failure in a duty of care and in the performance of masculinity

    Soluble polyacetylenes derived from the ring-opening metathesis polymerization of substituted cyclooctatetraenes: electrochemical characterization and Schottky barrier devices

    Recent developments in ring-opening metathesis polymerization (ROMP) have enabled the synthesis of poly-cyclooctatetraene (poly-COT), a material which is isostructural to polyacetylene. This liquid-phase polymerization method allows facile construction of interfaces, films, and devices with polyacetylene-like materials. The ROMP method also allows the preparation of soluble, yet highly conjugated polyacetylene analogs from substituted cyclooctatetraenes (R-COT). The redox characteristics of R-COT polymers were investigated at electrodes modified with thin polymer films. Voltammetric methods were used to characterize the redox response, band gap, electrochemical doping, and cis-trans isomerization properties of these polyenes. We have applied poly-COT technology to the fabrication of Schottky diodes and photoelectrochemical cells, by forming poly-COT films on semiconductor surfaces. The resultant semiconductor/organic-metal interfaces behave more ideally than semiconductor contacts with conventional metals, in that changes in the work function of the conducting polymer exert a large and predictable effect on the electrical properties of the resulting Schottky diodes. Transparent films of the solution-processible polymer poly- trimethylsilyl-cyclooctatetraene (poly-TMS-COT) have been cast onto n-silicon substrates and doped with iodine to form surface barrier solar cells. These devices produce photovoltages that are much larger than can be obtained from n-silicon contacts with conventional metals

    Low Cost and Compact Quantum Cryptography

    We present the design of a novel free-space quantum cryptography system, complete with purpose-built software, that can operate in daylight conditions. The transmitter and receiver modules are built using inexpensive off-the-shelf components. Both modules are compact allowing the generation of renewed shared secrets on demand over a short range of a few metres. An analysis of the software is shown as well as results of error rates and therefore shared secret yields at varying background light levels. As the system is designed to eventually work in short-range consumer applications, we also present a use scenario where the consumer can regularly 'top up' a store of secrets for use in a variety of one-time-pad and authentication protocols.Comment: 18 pages, 9 figures, to be published in New Journal of Physic

    Madness decolonized?: Madness as transnational identity in Gail Hornstein’s Agnes’s Jacket

    The US psychologist Gail Hornstein’s monograph Agnes’s Jacket: A Psychologist’s Search for the Meanings of Madness (2009) is an important intervention in the identity politics of the mad movement. Hornstein offers a resignified vision of mad identity that embroiders the central trope of an “anti-colonial” struggle to reclaim the experiential world “colonized” by psychiatry. A series of literal and figurative appeals make recourse to the inner world and (corresponding) cultural world of the mad, as well as to the ethno-symbolic cultural materials of dormant nationhood. This rhetoric is augmented by a model in which the mad comprise a diaspora without an origin, coalescing into a single transnational community. The mad are also depicted as persons displaced from their metaphorical homeland, the “inner” world “colonized” by the psychiatric regime. There are a number of difficulties with Hornstein’s rhetoric, however. Her “ethnicity-and-rights” response to the oppression of the mad is symptomatic of Western parochialism, while her proposed transmutation of putative psychopathology from limit upon identity to parameter of successful identity is open to contestation. Moreover, unless one accepts Hornstein’s porous vision of mad identity, her self-ascribed insider status in relation to the mad community may present a problematic “re-colonization” of mad experience

    Extracting inter-dot tunnel couplings between few donor quantum dots in silicon

    The long term scaling prospects for solid-state quantum computing architectures relies heavily on the ability to simply and reliably measure and control the coherent electron interaction strength, known as the tunnel coupling, tc. Here, we describe a method to extract the tc between two quantum dots (QDs) utilising their different tunnel rates to a reservoir. We demonstrate the technique on a few donor triple QD tunnel coupled to a nearby single-electron transistor(SET)in silicon. The device was patterned using scanning tunneling microscopy-hydrogen lithography allowing for a direct measurement of the tunnel coupling for a given inter-dot distance. We extract tc = ± 5.5 1.8 GHz and tc = ± 2.2 1.3 GHz between each of the nearest-neighbour QDs which are separated by 14.5 nm and 14.0 nm, respectively. The technique allows for an accurate measurement of tc for nanoscale devices even when it is smaller than the electron temperature and is an ideal characterisation tool for multi-dot systems with a charge senso
