Genomic relatedness within five common Finnish Campylobacter jejuni pulsed-field gel electrophoresis genotypes studied by amplified fragment length polymorphism analysis, ribotyping and serotyping
Thirty-five Finnish Campylobacter jejuni strains with five SmaI/SacII pulsed-field gel electrophoresis (PFGE) genotypes selected among human and chicken isolates from 1997 and 1998 were used for comparison of their PFGE patterns, amplified fragment length polymorphism (AFLP) patterns, HaeIII ribotypes, and heat-stable (HS) serotypes. The discriminatory power of PFGE, AFLP, and ribotyping with HaeIII were shown to be at the same level for this selected set of strains, and these methods assigned the strains into the same groups. The PFGE and AFLP patterns within a genotype were highly similar, indicating genetic relatedness. The same HS serotypes were distributed among different genotypes, and different serotypes were identified within one genotype. HS serotype 12 was only associated with the combined genotype G1 (PFGE-AFLP-ribotype). These studies using polyphasic genotyping methods suggested that common Finnish C. jejuni genotypes form genetic lineages which colonize both humans and chickens

implemented to quantify live C. jejuni, C. coli and C. lari on broiler chicken carcasses at selected processing
steps in the slaughterhouse. The samples were enumerated by culture for comparison. The Campylobacter
counts determined with the PMA-qPCR and the culture method were not concordant. We conclude that
the qPCR combined with PMA treatment of the samples did not fully reduce the signal from dead cells
Substantial CT radiation dose reduction does not affect the preference for CT over direct digital radiography to diagnose isolated zygomatic fractures:a study in human cadavers
Introduction: Zygomatic fractures can be diagnosed with either computed tomography (CT) or direct digital radiography (DR). The aim of the present study was to assess the effect of CT dose reduction on the preference for facial CT versus DR for accurate diagnosis of isolated zygomatic fractures. Materials and methods: Eight zygomatic fractures were inflicted on four human cadavers with a free fall impactor technique. The cadavers were scanned using eight CT protocols, which were identical except for a systematic decrease in radiation dose per protocol, and one DR protocol. Single axial CT images were displayed alongside a DR image of the same fracture creating a total of 64 dual images for comparison. A total of 54 observers, including radiologists, radiographers and oral and maxillofacial surgeons, made a forced choice for either CT or DR. Results: Forty out of 54 observers (74%) preferred CT over DR (all with P < 0.05). Preference for CT was maintained even when radiation dose reduced from 147.4 mSv to 46.4 mSv (DR dose was 6.9 mSv). Only a single out of all raters preferred DR (P ¼ 0.0003). The remaining 13 observers had no significant preference. Conclusion: This study demonstrates that preference for axial CT over DR is not affected by substantial (~70%) CT dose reduction for the assessment of zygomatico-orbital fractures
Quantitative exposure to livestock-associated MRSA ST398 of pig slaughterhouse workers
Objectives: To quantify livestock-associated MRSA (LA-MRSA) exposure to workers in pig slaughterhouses and assess associated risk factors for carriage in slaughterhouse workers. Methods: A cross-sectional study in three Dutch pig slaughterhouses was undertaken. Nasal swabs of 341 participants, surface wipes, air, and glove samples were screened for presence of MRSA. MRSA was quantitatively determined on gloves and in air samples by culturing and real-time PCR

Classifications of the Campylobacter fetus subspecies fetus and venerealis were first described in 1959 and were based on the source of isolation (intestinal versus genital) and the ability of the strains to proliferate in the genital tract of cows. Two phenotypic assays (1% glycine tolerance and H2S production) were described to differentiate the subspecies. Multiple molecular assays have been applied to differentiate the C. fetus subspecies, but none of these tests is consistent with the phenotypic identification methods. In this study, we defined the core genome and accessory genes of C. fetus, which are based on the closed genomes of five C. fetus strains. Phylogenetic analysis of the core genomes of 23 C. fetus strains of the two subspecies showed a division into two clusters. The phylogenetic core genome clusters were not consistent with the phenotypic classifications of the C. fetus subspecies. However, they were consistent with the molecular characteristics of the strains, which were determined by multilocus sequence typing, sap typing, and the presence/absence of insertion sequences and a type I restriction modification system. The similarity of the genome characteristics of three of the phenotypically defined C. fetus subsp. fetus strains to C. fetus subsp. venerealis strains, when considering the core genome and accessory genes, requires a critical evaluation of the clinical relevance of C. fetus subspecies identification by phenotypic assays
Tuning the Energetic Landscape of Ruddlesden-Popper Perovskite Films for Efficient Solar Cells
Ruddlesden-Popper perovskite films deposited with different methods show very diverse phase segregation and composition. When DMSO is used as solvent, the conventional method based on spin-coating and annealing produces very poor devices, whereas the vacuum-assisted method proposed here allows obtaining devices with efficiency up to 14.14%. The conventional method gives rise to a three-dimensional (3D)-like phase on the top of the film but dominant n = 2 phase with large domains (∼40 μm) at the bottom of the film. These n = 2 domains are oriented with their inorganic slabs parallel to the substrate and inhibit the charge transport in the vertical direction. Consequently, severe monomolecular and bimolecular charge recombination occurs in the solar cells. Conversely, the vacuum-assisted method yields films with a 3D-like phase dominant throughout their entire thickness and only a small amount of n ≤ 2 domains of limited dimensions (∼3 μm) at the bottom, which facilitate charge transport and reduce charge recombination

Mutational analysis of human CEACAM1: the potential of receptor polymorphism in increasing host susceptibility to bacterial infection
A common overlapping site on the N-terminal IgV-like domain of human carcinoembryonic antigen (CEA)-related cell adhesion molecules (CEACAMs) is targeted by several important human respiratory pathogens. These include Neisseria meningitidis (Nm) and Haemophilus influenzae (Hi) that can cause disseminated or persistent localized infections. To define the precise structural features that determine the binding of distinct pathogens with CEACAMs, we have undertaken molecular modelling and mutation of the receptor molecules at previously implicated key target residues required for bacterial binding. These include Ser-32, Tyr-34, Val-39, Gln-44 and Gln-89, in addition to Ile-91, the primary docking site for the pathogens. Most, but not all, of these residues located adjacent to each other in a previous N-domain model of human CEACAM1, which was based on REI, CD2 and CD4. In the current studies, we have refined this model based on the mouse CEACAM1 crystal structure, and observe that all of the above residues form an exposed continuous binding region on the N-domain. Examination of the model also suggested that substitution of two of these residues 34 and 89 could affect the accessibility of Ile-91 for ligand binding. By introducing selected mutations at the positions 91, 34 and 89, we confirmed the primary importance of Ile-91 in all bacterial binding to CEACAM1 despite the inter- and intraspecies structural differences between the bacterial CEACAM-binding ligands. The studies further indicated that the efficiency of binding was significantly enhanced for specific strains by mutations such as Y34F and Q89N, which also altered the hierarchy of Nm versus Hi strain binding. These studies imply that distinct polymorphisms in human epithelial CEACAMs have the potential to decrease or increase the risk of infection by the receptor-targeting pathogens
Photochromism in Ruddlesden-Popper copper-based perovskites:A light-induced change of coordination number at the surface
Ruddlesden-Popper organic-inorganic hybrid copper-based perovskites have been studied for decades owing to a variety of interesting properties, such as thermochromism and piezochromism, and the mechanisms behind these phenomena have been explained. Another possible property of these materials that has seldomly been investigated is photochromism. In this work, the photochromic properties of bis(phenethylammonium) tetrachlorocuprate (also known as phenethylammonium copper chloride) are reported for the first time. This material has attracted scientific interest owing to the fact that it shows both ferroelectric and ferromagnetic behavior. This work highlights the difference in stability between two Ruddlesden-Popper copper-based perovskites - with phenethylammonium (PEA) or methylammonium (MA) as the cations - during external stimuli. Various techniques, such as Raman and X-ray photoelectron spectroscopy, and grazing-incidence wide-angle X-ray scattering, combined with optical studies, were used to investigate the underlying photochemical processes at a molecular level. It is found that for the PEA compound, ultraviolet illumination causes a color change from yellow to brown. This is the result of two independent events, namely a Cu2+ reduction reaction and a transition from an octahedral copper-chloride structure to square-planar CuCl42-. After illumination, the material (brown color) is unstable in air, which is evident from a color change back to yellow. Interestingly, the similar compound bis(methylammonium) tetrachlorocuprate does not display photochromic behavior, which is attributed to the different nature of the two organic cations
