    Worldwide, tuberculosis (TB) remains the most frequent and important infectious disease causing morbidity and death. One-third of the world's population is infected with Mycobacterium tuberculosis (Mtb), the etiologic agent of TB. In this context, TB is in the top three, with malaria and HIV being the leading causes of death from a single infectious agent, and about two million deaths are attributable to TB annually. The bacterial enzyme MurA catalyzes the transfer of enolpyruvate from phosphoenolpyruvate (PEP) to uridine diphospho-N-acetylglucosamine (UNAG), which is the first committed step of bacterial cell wall biosynthesis. In this work, 3D structural model of Mtb-MurA enzyme has been developed, for the first time, by homology modeling and molecular dynamics simulation techniques. The model provided clear insight in its structure features, i.e. substrate binding pocket, and common docking site. Multiple sequence alignment and 3D structure model provided the putative substrate binding pocket of Mtb-MurA with respect to E.coli MurA. This analysis was helpful in identifying the binding sites and molecular function of the MurA homologue. Molecular docking study was performed on this 3D structural model, using different classes of inhibitors like fosfomycin, cyclic disulfide analog RWJ-3981, pyrazolopyrimidine analog RWJ-110192, purine analog RWJ-140998, 5-sulfonoxy-anthranilic acid derivatives T6361, T6362 and the results showed that the 5-sulfonoxyanthranilic acid derivatives is showed best interaction compared with other inhibitor, taking in to this we also design a new efficient analogs of T6361 and T6362 which are showed even better interaction with Mtb-MurA than the parental5-sulfonoxy-anthranilic acid derivatives. Further the comparative molecular electrostatic potential and cavity depth analysis of Mtb-MurA suggested several important differences in its substrate and inhibitor binding pocket. Such differences could be exploited in the future for designing of a more specific inhibitor for Mtb-MurA enzym

    Antibacterial Activity of a Mushroom - Stereum ostrea

    The antimicrobial activity of crude culture filtrate and methanol extract of Stereum ostrea was evaluated by disc diffusion method against two Gram-negative bacteria- Klebsiella pneumonia, Pseudomonas aeruginosa and three Gram-positive bacteria such as Bacillus subtilis, Staphylococcus aureus and Micrococcus sps. To determine the minimal inhibitory concentrations (MICs) we studied concentrations of 10-50 µl of crude and methanol extract of Stereum ostrea against selected bacteria. Crude culture filtrate was highly inhibitory when compared to methanol extract, which were evident through the increased zones of inhibition against Gram-negative and Gram-positive bacteria. Crude culture filtrate of Stereum ostrea showed highest and lowest percent of inhibition zone against Bacillus subtilis (15.9 mm) and Klebsiella pneumonia (9.1mm). These results indicate that crude culture filtrate of Stereum ostrea possesses potential broad spectrum antimicrobial activity.     Effect of nano- and micron-sized K0.5Na0.5NbO3 fillers on the dielectric and piezoelectric properties of PVDF composites

    Polymer nanocrystal composites were fabricated by embedding polyvinylidene fluoride (PVDF) with K0.5Na0.5NbO3 (KNN) nanocrystallites of different volume fraction using the hot-pressing technique. For comparison, PVDF-KNN microcrystal composites of the same compositions were also fabricated which facilitated the studies of the crystallite size (wide range) effect on the dielectric and piezoelectric properties. The structural, morphological, dielectric, and piezoelectric properties of these nano and micro crystal composites were investigated. The incorporation of KNN fillers in PVDF at both nanometer and micron scales above 10 vol% resulted in the formation of polar beta-form of PVDF. The room temperature dielectric constant as high as 3273 at 100 Hz was obtained for the PVDF comprising 40 vol% KNN nanocrystallites due to dipole-dipole interactions (as the presence of beta-PVDF is prominent), whereas it was only 236 for the PVDF containing the same amount (40 vol%) of micron-sized crystallites of KNN at the same frequency. Various theoretical models were employed to predict the dielectric constants of the PVDF-KNN nano and micro crystal composites. The PVDF comprising 70 vol% micron-sized crystallites of KNN exhibited a d (33) value of 35 pC/N, while the nanocrystal composites of PVDF-KNN did not exhibit any piezoelectric response perhaps due to the unrelieved internal stress within each grain, besides the fact that they have less domain walls

    A study on various clinical presentations of extradural hemorrhage, factors affecting treatment and early outcome

    Background: In India 11% of deaths are due to trauma and 78% of injury deaths are due to head injury. The aim of this study was to analyse the clinical spectrum and to evaluate the postoperative outcome in patients with head injury with an extradural hematoma.Methods: This was a retrospective observational study which included 100 patients admitted in King George hospital, Andhra medical college, Visakhapatnam, Andhra Pradesh, India over the past two years (01/10/2014 to 30/09/2016) with head injury, diagnosed to have traumatic extradural hemorrhage. A detailed clinical history, physical examination and CT scan was performed in all patients. For patients who were subjected to surgery operative and post-operative findings were noted.Results: The maximum patients suffering from EDH are in the age group of 21-30 years (28%) with male predominance (95%). The most common mode of injury is RTA (58%) under the influence of alcohol. Majority of cases reached hospital within 6 hours from  time of injury (44%). 95% of patients with EDH presented with LOC followed by vomiting in 68% of cases, followed by Headache in 42% of cases. 53% of the cases presented with mass effect over brain parenchyma with frontal EDH as most common location. Out of 100 cases, surgical approach was considered in 57 patients while remaining 47 patients were managed conservatively. Recovery from clinical and functional morbidity was satisfactorily acceptable, following treatment.Conclusions: Early presentation with mild to moderate GCS has good clinical outcome with minimal disability

    Clinical study of acute subdural haematoma – a level I trauma care centre experience

    Background: Acute subdural hematoma is the most common type of traumatic intra cranial hematoma accounting for 24% cases of severe head injuries and caries highest mortality. The aim of this study is to analyse the clinical spectrum, and to evaluate the postoperative outcome.Methods: This is a prospective observational Study which included 150 patients  admitted  in King George hospital, Andhra Medical College, Visakhapatnam, India over the past two years (01st August 2013 to 30th August 2015) with head injury, diagnosed to have Traumatic subdural Hemorrhage. A detailed clinical history, physical examination, CT scan was performed in all patients. Patients who were subjected to surgery, post-operative out-come were compared.Results: The maximum patients suffering from SDH are in the age group of 20-40 years (63%) with male predominance (73%). The most common mode of injury is RTA (73.33%). 40% of cases presented to hospital with gcs 10mm and midline shift of >5mm, Basal cisterns obliteration, post traumatic seizure and delay in interval between the surgery had greatly affected on outcome of patients. Out of 150 cases, surgical approach was considered in 120 patients while remaining 30 patients were managed conservatively.Conclusions: Interval between injury to surgery with interval <4 hours having favourable outcome. Basal cistern obliteration, which is an indicator of increased intra cranial pressure is the single most important factor for unfavourable outcome. Hypotension and post traumatic seizures influence high mortality

    Effect of Chemicals and Growth Regulators on Post-Harvest Shelf-Life and Quality in Papaya (Carica papaya L.) Cv. Red Lady

    The present investigation on papaya (Carica papaya&nbsp;L.) cv. Red Lady was carried out at Horticultural College and Research Institute, Venkataramannagudem, West Godavari district of Andhra Pradesh, during the year 2010-11. The study was carried out using 9 different treatments involving two chemicals calcium nitrate and calcium chloride at 1, 2, 3 and 4% concentration and two growth regulators, viz., GA3&nbsp;at 75, 100, 150 and 200mg/l, and BA at 100, 125, 150 and 175mg/l concentration conducted separately in a factorial concept of completely randomized design (CRD), with three replications, under laboratory conditions. Physical parameters studied were per cent fruit ripening, physiological loss in weight (PLW), fruit firmness, shelf-life, and, physic-chemical properties studied were: total soluble solids (TSS), total sugars, reducing sugars, acidity, ascorbic acid and Brix:acid ratio. Fruits treated with CaCl2&nbsp;@ 4% showed significantly low PLW, per cent fruit ripening, and the highest fruit-firmness, shelf-life, lowest total soluble solids, total sugars, reducing sugars, Brix:acid ratio, and highest acidity and ascorbic acid content, which were on par with CaCl2&nbsp;@ 2% application. Fruits treated with GA3&nbsp;@ 100mg/l also exhibited similar results for these parameters. It was concluded that CaCl2&nbsp;@ 4 % had a beneficial impact on shelf-life of papaya fruits upto 10.67 days without any loss in either physical or physic-chemical properties. Similarly, application of growth regulators such as GA3&nbsp;and BA @100mg/l significantly increased shelf-life of papaya fruits upto 12 days and 11 days, respectively, while showing the best physical and physico-chemical properties

    This focuses on encrypted data search, which is a vital means of protecting the privacy of file encryption before outsourcing in cloud computing or, in general, on almost all network-based information systems in which servers cannot be totally reliable. We officially demonstrate that our proposed plan is selectively secure against the selected keyword attack. We've created a single, authoritative, scalable search in an encrypted data plan that supports multiple data users and multiple data collaborators. We differentiate between features and keywords in our design. Keywords are the actual content of the files, while the attributes refer to the attributes of the user. In addition, using proxy encryption and slow file encryption techniques, the proposed plan is more relevant to the cloud outsourcing model and is efficient at eliminating the user. Unlike the existing public key search scheme, our plan can achieve scalability and system improvement at the same time. Unlike a search plan with native file encryption, our plan lets you search for approved, adaptable keywords in arbitrarily structured data. The appearance of complexity is a straight line to the amount of attributes within the system versus the number of authorized users. Thus, the one-to-many license mechanism is best suited for any huge system, for example, cloud.     Sensitivity of WRF cloud microphysics to simulations of a severe thunderstorm event over southeast India

    In the present study, we have used the Weather Research and Forecasting (WRF) model to simulate the features associated with a severe thunderstorm observed over Gadanki (13.5° N, 79.2° E), over southeast India, on 21 May 2008 and examined its sensitivity to four different microphysical (MP) schemes (Thompson, Lin, WSM6 and Morrison). We have used the WRF model with three nested domains with the innermost domain of 2 km grid spacing with explicit convection. The model was integrated for 36 h with the GFS initial conditions of 00:00 UTC, 21 May 2008. For validating simulated features of the thunderstorm, we have considered the vertical wind measurements made by the Indian MST radar installed at Gadanki, reflectivity profiles by the Doppler Weather Radar at Chennai, and automatic weather station data at Gadanki.There are major differences in the simulations of the thunderstorm among the MP schemes, in spite of using the same initial and boundary conditions and model configuration. First of all, all the four schemes simulated severe convection over Gadanki almost an hour before the observed storm. The DWR data suggested passage of two convective cores over Gadanki on 21 May, which was simulated by the model in all the four MP schemes. Comparatively, the Thompson scheme simulated the observed features of the updraft/downdraft cores reasonably well. However, all the four schemes underestimated strength and vertical extend of the updraft cores. The MP schemes also showed problems in simulating the downdrafts associated with the storm. While the Thompson scheme simulated surface rainfall distribution closer to observations, the other three schemes overestimated observed rainfall. However, all the four MP schemes simulated the surface wind variations associated with the thunderstorm reasonably well. The model simulated reflectivity profiles were consistent with the observed reflectivity profile, showing two convective cores. These features are consistent with the simulated condensate profiles, which peaked around 5-6 km. As the results are dependent on initial conditions, in simulations with different initial conditions, different schemes may become closer to observations. The present study suggests not only large sensitivity but also variability of the microphysical schemes in the simulations of the thunderstorm. The study also emphasizes the need for a comprehensive observational campaign using multi-observational platforms to improve the parameterization of the cloud microphysics and land surface processes over the Indian region
