Assessing emission reduction targets with dynamic models: deriving target load functions for use in integrated assessment
International audienceInternational agreements to reduce the emission of acidifying sulphur (S) and nitrogen (N) compounds have been negotiated on the basis of an understanding of the link between acidification related changes in soil and surface water chemistry and terrestrial and aquatic biota. The quantification of this link is incorporated within the concept of critical loads. Critical loads are calculated using steady state models and give no indication of the time within which acidified ecosystems might be expected to recover. Dynamic models provide an opportunity to assess the timescale of recovery and can go further to provide outputs which can be used in future emission reduction strategies. In this respect, the Target Load Function (TLF) is proposed as a means of assessing the deposition load necessary to restore a damaged ecosystem to some pre-defined acceptable state by a certain time in the future. A target load represents the deposition of S and N in a defined year (implementation year) for which the critical limit is achieved in a defined time (target year). A TLF is constructed using an appropriate dynamic model to determine the value of a chemical criterion at a given point in time given a temporal pattern of S and N deposition loads. A TLF requires information regarding: (i) the chemical criterion required to protect the chosen biological receptor (i.e. the critical limit); (ii) the year in which the critical limit is required to be achieved; and (iii) time pattern of future emission reductions. In addition, the TLF can be assessed for whole regions to incorporate the effect of these three essentially ecosystem management decisions. FAIRMODE: A FORUM FOR AIR QUALITY MODELLING IN EUROPE
FAIRMODE (Forum for AIR quality MODelling in Europe) is an air quality modelling network that was established as a
joint initiative of the European Environment Agency (EEA) and European Commission’s Joint Research Centre (JRC). In a
common effort EEA and JRC aim at responding to the requirements of the new Air Quality Directive, with particular focus on the
introduction of modelling as a necessary tool for air quality assessment and air quality management. The main aim of the modelling
network is to bring together air quality modellers and model users in order to promote and support harmonised use of modelling for
the assessment of air quality by EU and EEA member countries. The network will thus encourage synergy – at a local, national and
European level - through the development and implementation of a common infrastructure based on best practices for reporting and
storing information relevant to air quality modelling. A major objective of the FAIRMODE initiative is to provide guidance to
present and future air quality model users in EEA’s EIONET partnership network. FAIRMODE also aims to enhance awareness of
model usefulness, reliability and accuracy through model validation and intercomparison exercises at a national or European level.
The JRC has taken on a leading role in the co-ordination of the latter activities gaining from its experience in leading the
“Eurodelta” and “CityDelta” intercomparison exercises.
A centralised web portal has been created in support of FAIRMODE, which is currently being used for internal communication
purposes of the network participants, but will also provide the means for exchange of relevant material and experiences between all
interested modellers and model users. The initial activities of the network will be organised by two main Work Groups, focusing on
the preparation of a Guidance Document for model use and on model QA/QC procedures (input data, other uncertainties)
respectively. The progress of the preparation of these documents as well as of the rest of the regular activities of the network will be
reviewed and discussed within the frame of annual Plenary meetings and Steering Committee meetings
Dynamic modeling and target loads of sulfur and nitrogen for surface waters in Finland, Norway, Sweden, and the United Kingdom
The target load concept is an extension of the critical load concept of air pollution inputs to ecosystems. The advantage of target loads over critical loads is that one can define the deposition and the point in time (target year) when the critical (chemical) limit is no longer violated. This information on the timing of recovery requires dynamic modelling. Using a well-documented dynamic model, target loads for acidic deposition were determined for 848 surface waters across Finland, Norway, Sweden and the United Kingdom for the target year 2050. In the majority of sites (n = 675), the critical ANC-limit was predicted to be achieved by 2050; however, for 127 sites target loads were determined. In addition, 46 sites were infeasible, i.e., even a deposition reduction to zero would not achieve the limit by 2050. The average maximum target load for sulphur was 38% lower than the respective critical load across the study lakes (n = 127). Target loads on a large regional scale can inform effects-based emission reduction policies; the current assessment suggests that reductions beyond the Gothenburg Protocol are required to ensure surface water recovery from acidification by 2050

A full scale comparative study of methods for generation of functional Dendritic cells for use as cancer vaccines
<p/> <p>Background</p> <p>Dendritic cells (DCs) are professional antigen-presenting cells with the ability to induce primary T-cell responses and are commonly produced by culturing monocytes in the presence of IL-4 and GM-CSF for 5–7 days (Standard DC). Recently, Dauer and co-workers presented a modified protocol for differentiation of human monocytes into mature DCs within 48 hours (Fast DC). Here we report a functional comparison of the two strategies for generation of DCs from human monocytes with adaptions for large-scale clinical use.</p> <p>Methods</p> <p>The Elutra Cell Selection System was used to isolate monocytes after collection of leukapheresis product. The enriched monocytes were cultured in gas permeable Teflon bags with IL-4 and GM-CSF for 24 hours (Fast DC) or 5 days (Standard DC) to obtain immature DCs. The cells were then transfected with mRNA from the leukemia cell line Jurkat E6 by electroporation and incubated for additional 24 h or 2 days in the presence of pro-inflammatory cytokines (TNFα, IL-1β, IL-6 and PGE<sub>2</sub>) to obtain mature DCs.</p> <p>Results</p> <p>Mature Fast DC and Standard DC displayed comparable levels of many markers expressed on DC, including HLA-DR, CD83, CD86, CD208 and CCR7. However, compared to Standard DC, mature Fast DC was CD14<sup>high </sup>CD209<sup>low</sup>. Fast DC and Standard DC transfected with Jurkat E6-cell mRNA were equally able to elicit T cell specifically recognizing transfected DCs in vitro. IFNγ-secreting T cells were observed in both the CD4+ and CD8+ subsets.</p> <p>Conclusion</p> <p>Our results indicate that mature Fast DC are functional antigen presenting cells (APCs) capable of inducing primary T-cell responses, and suggest that these cells may be valuable for generation of anti-tumor vaccines.</p
γ-Secretase modulators show selectivity for γ-secretase–mediated amyloid precursor protein intramembrane processing
The aggregation of β-amyloid peptide 42 results in the formation of toxic oligomers and plaques, which plays a pivotal role in Alzheimer's disease pathogenesis. Aβ42 is one of several Aβ peptides, all of Aβ30 to Aβ43 that are produced as a result of γ-secretase–mediated regulated intramembrane proteolysis of the amyloid precursor protein. γ-Secretase modulators (GSMs) represent a promising class of Aβ42-lowering anti-amyloidogenic compounds for the treatment of AD. Gamma-secretase modulators change the relative proportion of secreted Aβ peptides, while sparing the γ-secretase–mediated processing event resulting in the release of the cytoplasmic APP intracellular domain. In this study, we have characterized how GSMs affect the γ-secretase cleavage of three γ-secretase substrates, E-cadherin, ephrin type A receptor 4 (EphA4) and ephrin type B receptor 2 (EphB2), which all are implicated in important contexts of cell signalling. By using a reporter gene assay, we demonstrate that the γ-secretase–dependent generation of EphA4 and EphB2 intracellular domains is unaffected by GSMs. We also show that γ-secretase processing of EphA4 and EphB2 results in the release of several Aβ-like peptides, but that only the production of Aβ-like proteins from EphA4 is modulated by GSMs, but with an order of magnitude lower potency as compared to Aβ modulation. Collectively, these results suggest that GSMs are selective for γ-secretase–mediated Aβ production
Aspiration–sclerotherapy Results in Effective Control of Liver Volume in Patients with Liver Cysts
Purpose To study the extent to which aspiration–sclerotherapy reduces liver volume and whether this therapy results in relief of symptoms. Results Four patients, group I, with isolated large liver cysts, and 11 patients, group II, with polycystic livers, underwent aspiration–sclerotherapy. Average volume of aspirated cyst fluid was 1,044 ml (range 225–2,000 ml) in group I and 1,326 ml (range 40–4,200 ml) in group II. Mean liver volume before the procedure was 2,157 ml (range 1,706–2,841 ml) in group I and 4,086 ml (range 1,553–7,085 ml) in group II. This decreased after the procedure to 1,757 ml (range 1,479–2,187 ml) in group I. In group II there was a statistically significant decrease to 3,347 ml (range 1,249–6,930 ml, P = 0.008). Volume reduction was 17.1% (range −34.7% to −4.1%) and 19.2% (range −53.9% to +2.4%) in groups I and II, respectively. Clinical severity of all symptoms decreased, except for involuntary weight loss and pain in group II. Conclusion Aspiration–sclerotherapy is an effective means of achieving liver volume reduction and relief of symptoms
Sequence stratigraphy, basin morphology and sea-level history for the Permian Kapp Starostin Formation of Svalbard, Norway
Based on seven measured sections from Svalbard, the marine strata of the Permian Kapp Starostin Formation are arranged into seven transgressive–regressive sequences (TR1–TR7) of c. 4–5 Ma average duration, each bound by a maximum regressive surface. Facies, including heterozoan-dominated limestones, spiculitic cherts, sandstones, siltstones and shales, record deposition within inner, middle and outer shelf areas. The lowermost sequence, TR1, comprises most of the basal Vøringen Member, which records a transgression across the Gipshuken Formation following a hiatus of unknown duration. Temperate to cold, storm-dominated facies established in inner to middle shelf areas between the latest Artinskian and Kungurian. Prolonged deepening during sequences TR2 and TR3 was succeeded by a long-term shallowing-upward trend that lasted until the latest Permian (TR4–TR7). A major depocentre existed in central and western Spitsbergen while to the north, Dickson Land remained a shallow platform, leading to a shallow homoclinal ramp in NE Spitsbergen and Nordaustlandet. The Middle Permian extinction (late Capitanian) is recorded near the base of TR6 in deeper parts of the basin only; elsewhere this sequence is not recorded. Likewise the youngest sequence, TR7, extending to the upper formational contact of latest Permian age, is found only in the basin depocentre. Comparison with age-equivalent strata in the Sverdrup Basin of Canada reveals a remarkably similar depositional history, with, for example, two (third-order) sea-level cycles recorded in the Late Permian of both regions, in keeping with the global record. Sequence stratigraphy may therefore be a powerful correlative tool for onshore and offshore Permian deposits across NW Pangaea
