    Influencia de un injerto en el perfil de ácidos grasos y algunas propiedades fisicoquímicas de la semilla y el aceite de semillas de sandía

    This study aimed to investigate the effects of grafting on the fatty acid profile and some physicochemical properties of watermelon seed and seed oil. The ‘Crimson Tide’ cultivar was used as the scion while two wild watermelon (Citrullus lanatus var. citroides (A1 and A2)), one Lagenaria siceraria (A3) and one Cucurbita maxima Duchesne x Cucurbita moschata Duchesne (A4) were used as rootstocks. The use of rootstock significantly influenced the fatty acid profile and the physical parameters of seeds and seed oils. The highest linoleic acid ratio was found in the seed oil from A1 and A2, the oil from A3 had the highest oleic acid ratio. The results showed that the content and acid value in seed oils were improved, and that total phenolic compounds and antioxidant activity of both seed and oil were decreased by grafting. Wild rootstocks can be used in watermelon cultivation to obtain a watermelon seed which is rich in linoleic acid.El objetivo de este estudio fue investigar los efectos del injerto en el perfil de ácidos grasos y algunas propiedades fisicoquímicas de la semilla y el aceite de semillas de sandía. El cultivar ‘Crimson Tide’ se utilizó como vástago, mientras que dos sandías silvestres (Citrullus lanatus var. Citroides (A1 y A2)), una Lagenaria siceraria (A3) y una Cucurbita maxima Duchesne x Cucurbita moschata Duchesne (A4) se utilizaron como portainjertos. El uso de portainjertos influyó significativamente en el perfil de ácidos grasos y los parámetros físicos de semillas y aceites de semillas. La proporción de ácido linoleico más alta se encontró en el aceite de semillas de A1 y A2, el aceite de A3 tuvo la proporción de ácido oleico más alta. Los resultados mostraron que el contenido de aceite y el índice de acidez mejoró y los compuestos fenólicos totales y la actividad antioxidante tanto de la semilla como del aceite se redujeron mediante el injerto. Para obtener un aceite de semillas de sandía rico en ácido linoleico, se pueden utilizar portainjertos silvestres en el cultivo de sandía

    Cellphone-Based Hand-Held Microplate Reader for Point-of-Care Testing of Enzyme-Linked Immunosorbent Assays

    Standard microplate based enzyme-linked immunosorbent assays (ELISA) are widely utilized for various nanomedicine, molecular sensing, and disease screening applications, and this multiwell plate batched analysis dramatically reduces diagnosis costs per patient compared to nonbatched or nonstandard tests. However, their use in resource-limited and field-settings is inhibited by the necessity for relatively large and expensive readout instruments. To mitigate this problem, we created a hand-held and cost-effective cellphone-based colorimetric microplate reader, which uses a 3D-printed optomechanical attachment to hold and illuminate a 96-well plate using a light-emitting-diode (LED) array. This LED light is transmitted through each well, and is then collected via 96 individual optical fibers. Captured images of this fiber-bundle are transmitted to our servers through a custom-designed app for processing using a machine learning algorithm, yielding diagnostic results, which are delivered to the user within ∼1 min per 96-well plate, and are visualized using the same app. We successfully tested this mobile platform in a clinical microbiology laboratory using FDA-approved mumps IgG, measles IgG, and herpes simplex virus IgG (HSV-1 and HSV-2) ELISA tests using a total of 567 and 571 patient samples for training and blind testing, respectively, and achieved an accuracy of 99.6%, 98.6%, 99.4%, and 99.4% for mumps, measles, HSV-1, and HSV-2 tests, respectively. This cost-effective and hand-held platform could assist health-care professionals to perform high-throughput disease screening or tracking of vaccination campaigns at the point-of-care, even in resource-poor and field-settings. Also, its intrinsic wireless connectivity can serve epidemiological studies, generating spatiotemporal maps of disease prevalence and immunity

    Fire behaviour assessment of rigid polyurethane foams containing nanoclay and intumescent flame retardant based on cone calorimeter tests

    In this study, nanoclay was incorporated into rigid polyurethane foams at different loadings, namely 5 wt. %, 10 wt. % and 15 wt. %, to enhance the fire resistance of the foams. In addition, an intumescent flame retardant composed of ammonium polyphosphate/pentaerythritol was additionally introduced to generate a synergistic positive effect on the fire resistance of the foams. The fire behaviour of the foams containing nanoclay with and without the intumescent flame retardant was investigated by using a cone calorimeter. The experimental results showed that the fire resistance of the rigid polyurethane foams could be enhanced with increasing the nanoclay loading in the foam. Furthermore, it was found that the nanoclay had a good synergistic effect with the intumescent flame retardant in the rigid polyurethane foams in terms of fire resistance. © 2019 University of Chemical Technology and Metallurgy

    Confirmed Exposure to Tick-Borne Encephalitis Virus and Probable Human Cases of Tick-Borne Encephalitis in Central/Northern Anatolia, Turkey

    Tick-borne encephalitis virus (TBEV) is the aetiological agent of tick-borne encephalitis (TBE), a potentially fatal central nervous system infection of humans. TBE is endemic in many areas of Europe and Asia; however, very scarce data on TBEV activity are available from Turkey. We aimed to identify TBEV exposure in healthy blood donors and the impact of TBEV in central nervous system infections in Central/Northern Anatolia. Two-thousand four hundred and fifty four sera, collected from blood donors at Ankara, Konya, Eskişehir and Zonguldak branches of the Turkish Red Crescent Middle Anatolia Regional Blood Center, were analysed for TBEV serosurveillance. Paired serum and cerebrospinal fluid samples from 108 patients with the diagnosis of aseptic meningitis/encephalitis of unknown aetiology were also evaluated to identify TBE and neuroborreliosis cases. Commercial enzyme-linked immunosorbent assays and indirect immunofluorescence tests were employed for antibody detection. Forty-seven donor samples (1.9%) were reactive for TBEV IgG. In 25 persons with IgG reactivity (53.1%), risk factors for tick-borne infections were revealed. One sample from Zonguldak province (1/198; 0.5%) in the Black Sea region of Turkey was confirmed to possess neutralizing antibodies via plaque reduction neutralization test. TBEV IgM was detected in 9.2% (8/108) of the patients. IgM was accompanied by IgG reactivity in two persons where, in one, recent history of a tick bite was also identified. Intrathecal antibody production for TBEV could not be demonstrated. No evidence for Borrelia infections could be found. Confirmed exposure to TBEV and/or an antigenically similar tick-borne flavivirus is documented for the first time in blood donors in Zonguldak in Northern Anatolia. Probable cases of TBE have also been identified from Central Anatolia. The epidemiology of TBEV activity in Turkey needs to be assessed and benefits of vaccination for general population, risk groups or travellers must be considered
