Constitutive activation and accelerated maturation of peripheral blood t cells in healthy adults in burkina faso compared to Germany: The case of malaria?
<p>Abstract</p> <p>Objective</p> <p>It is not exactly known how frequent exposure to <it>Plasmodium falciparum </it>shapes the peripheral blood T-cell population in healthy West Africans.</p> <p>Methods</p> <p>The frequency of peripheral blood CD4<sup>+ </sup>lymphocytes responding to <it>Plasmodium falciparum </it>merozoite surface protein 1 <it>(Pf</it>MSP-1) by production of interferon-gamma (IFN-γ), interleukin-2 (IL-2) or tumor necrosis factor-alpha (TNF-α) was determined using a commercially available flow cytometric activation assay (Fastlmmune) in 17 healthy adults in Nouna, Burkina Faso. T-cell activation and maturation in peripheral blood of healthy adults in Burkina Faso (n = 40) and Germany (n = 20) were compared using immunophenotyping and three-colour flow cytometry.</p> <p>Results</p> <p>Significant numbers of <it>Pf</it>MSV-1 -specific CD4<sup>+ </sup>lymphocytes producing IFN-γ, IL-2 and/or TNF-α were detected in 14 healthy adults in Nouna. Cytokine profiles showed predominant production of IFN-γ and TNF-α. Compared to Germans, Burkinabé showed markedly lower proportions of CCR7<sup>+ </sup>CD45RA<sup>+ </sup>naïve CD4<sup>+ </sup>cells and slightly higher frequencies of CD95<sup>+ </sup>CD4<sup>+ </sup>T-cells and of CD38<sup>+ </sup>CD8<sup>+ </sup>T-cells. The median antibody-binding capacity of CD95<sup>dim </sup>CD4<sup>+ </sup>T-cells in Burkinabé was more than twice the value observed in Germans (263 vs. 108 binding sites per cell, p < 0.0001).</p> <p>Conclusions</p> <p>We hypothesize that an IFN-γ-induced increase in the expression level of CD95 on CD4<sup>+ </sup>lymphocytes may lower the activation threshold of resting naïve CD4<sup>+ </sup>T-cells in healthy adults living in Burkina Faso. Bystander activation of these cells deserves further study as a molecular mechanism linking strong IFN-γ responses against <it>Plasmodium falciparum </it>to decreased susceptibility to parasitemia observed in specific ethnic groups in West Africa.</p
Iterative real-time path integral approach to nonequilibrium quantum transport
We have developed a numerical approach to compute real-time path integral
expressions for quantum transport problems out of equilibrium. The scheme is
based on a deterministic iterative summation of the path integral (ISPI) for
the generating function of the nonequilibrium current. Self-energies due to the
leads, being non-local in time, are fully taken into account within a finite
memory time, thereby including non-Markovian effects, and numerical results are
extrapolated both to vanishing (Trotter) time discretization and to infinite
memory time. This extrapolation scheme converges except at very low
temperatures, and the results are then numerically exact. The method is applied

Dietary cholesterol promotes repair of demyelinated lesions in the adult brain
Multiple Sclerosis (MS) is an inflammatory demyelinating disorder in which remyelination failure contributes to persistent disability. Cholesterol is rate-limiting for myelin biogenesis in the developing CNS; however, whether cholesterol insufficiency contributes to remyelination failure in MS, is unclear. Here, we show the relationship between cholesterol, myelination and neurological parameters in mouse models of demyelination and remyelination. In the cuprizone model, acute disease reduces serum cholesterol levels that can be restored by dietary cholesterol. Concomitant with blood-brain barrier impairment, supplemented cholesterol directly supports oligodendrocyte precursor proliferation and differentiation, and restores the balance of growth factors, creating a permissive environment for repair. This leads to attenuated axon damage, enhanced remyelination and improved motor learning. Remarkably, in experimental autoimmune encephalomyelitis, cholesterol supplementation does not exacerbate disease expression. These findings emphasize the safety of dietary cholesterol in inflammatory diseases and point to a previously unrecognized role of cholesterol in promoting repair after demyelinating episodes
The fate of Böhler's angle in conservatively-treated displaced intra-articular calcaneal fractures
Purpose: Although the predictive value of Böhler's angle on outcome remains subject of debate, the initial angle at the time of trauma still guides treatment. Changes in Böhler's angle during follow-up are frequently reported following surgical treatment of displaced intra-articular calcaneal fractures (DIACF). The aim of the present study was to determine the changes in Böhler's angle as a measure of secondary fracture displacement following conservative management of DIACF. Methods: Thirty-eight patients with a total of 44 displaced intra-articular calcaneal fractures treated conservatively with a minimum of two lateral radiographs during follow-up were analysed. Böhler's angle at different follow-up times was measured by three observers. The change in angle was compared with the angle at trauma, and influence of trauma mechanism and common calcaneal fracture classifications were determined. Results: The results showed a significant decline over time of the Böhler's angle in conservatively-treated patients of more than 11° on average at a mean follow-up of 29.2 weeks. This decrease was not related to gender, the initial angle, or the Essex-Lopresti or Sanders classification. A statistically significantly higher decrease was detected in high energetic trauma compared with low energetic trauma. Conclusion: The conservative treatment of displaced intra-articular calcaneal fractures is still a viable option, yet a significant secondary displacement in time should be taken into account, as reflected in a decrease of Böhler's angle of 11° up to one year following trauma
GRS 1915+105 : High-energy Insights with SPI/INTEGRAL
We report on results of two years of INTEGRAL/SPI monitoring of the Galactic
microquasar GRS 1915+105. From September 2004 to May 2006, the source has been
observed twenty times with long (approx 100 ks) exposures. We present an
analysis of the SPI data and focus on the description of the high-energy (> 20
keV) output of the source. We found that the 20 - 500 keV spectral emission of
GRS 1915+105 was bound between two states. It seems that these high-energy
states are not correlated with the temporal behavior of the source, suggesting
that there is no direct link between the macroscopic characteristics of the
coronal plasma and the the variability of the accretion flow. All spectra are
well fitted by a thermal comptonization component plus an extra high-energy
powerlaw. This confirms the presence of thermal and non-thermal electrons

Image-based Search and Retrieval for Biface Artefacts using Features Capturing Archaeologically Significant Characteristics
Archaeologists are currently producing huge numbers of digitized photographs to record and preserve artefact finds. These images are used to identify and categorize artefacts and reason about connections between artefacts and perform outreach to the public. However, finding specific types of images within collections remains a major challenge. Often, the metadata associated with images is sparse or is inconsistent. This makes keyword-based exploratory search difficult, leaving researchers to rely on serendipity and slowing down the research process. We present an image-based retrieval system that addresses this problem for biface artefacts. In order to identify artefact characteristics that need to be captured by image features, we conducted a contextual inquiry study with experts in bifaces. We then devised several descriptors for matching images of bifaces with similar artefacts. We evaluated the performance of these descriptors using measures that specifically look at the differences between the sets of images returned by the search system using different descriptors. Through this nuanced approach, we have provided a comprehensive analysis of the strengths and weaknesses of the different descriptors and identified implications for design in the search systems for archaeology
Treatment of displaced intra-articular calcaneal fractures by ligamentotaxis: current concepts’ review
Introduction: A large variety of therapeutic modalities for calcaneal fractures have been described in the literature. No single treatment modality for displaced intra-articular calcaneal fractures has proven superior over the other. This review describes and compares the different percutaneous distractional approaches for intra-articular calcaneal fractures. The history, technique, anatomical and fracture considerations, limitations and the results of different distractional approaches reported in the literature are reviewed. Method: Literature review on different percutaneous distractional approaches for displaced intra-articular calcaneal fractures. Results: Eight studies in which application of a distraction technique was used for the treatment of calcaneal fractures were identified. Because of the use of different classification, techniques, and outcome scoring systems, a meta-analysis was not possible. A literature review reveals overall fair to poor result in 10-29% of patients. Ten up to 26% of patients are unable to return to work after percutaneous treatment of their fracture. A secondary arthrodesis has to be performed in 2-15% of the cases. Infectious complications occur in 2-15%. Some loss of reduction is reported in 4-67%. Conclusion: Percutaneous distractional reduction and fixation appears to be a safe technique with overall good results and an acceptable complication rate, compared with other treatment modalities for displaced intra-articular calcaneal fractures. A meta-analysis, based on Cochrane Library criteria is not possible, because of a lack of level 1 and 2 trials on this subject
