    Reproductive biology of greasy grouper, Epinephelus tauvina and coral hind grouper Cephalopholis miniata (Family: Serranidae) in the southern Red Sea, Shalatien, Egypt

    Groupers are the main component of commercial fisheries in the Red Sea. The reproductive biology of Epinephelus tauvina and Cephalopholis miniata collected at the Shalatien landing site in the southern Egyptian Red Sea were investigated from January to December 2017. A total of 212 specimens of E. tauvina with a total length range of 23.6 to 70.3 cm and 243 specimens of C. miniata with a total length range of 17.4 to 42.1 cm were examined. Females of E. tauvina and C. miniata accounted for 61.7 and 61.9% of the total fish sampled, showing a 1:1.61 and 1:1.63 male-to-female sex ratio, respectively. The monthly distribution of maturity stages and gonadosomatic index values showed that the spawning season for both sexes was extended from April to October for E. tauvina and from April to September for C. miniata. The lengths at first sexual maturity of the males and females of E. tauvina were estimated at 49 and 48.1 cm, respectively, while those for C. miniata were estimated at 23.2 and 22.7 cm, respectively. All our results can help in the proper management of these valuable resources

    Alarming Eating Behaviours among Adolescents in Egypt

    BACKGROUND: Adolescence is a phase of rapid growth and increased nutritional needs. It includes the stressful burden of pubertal changes, both physically and psychologically. Moreover, it is associated with the utmost need for independence and identity formation. An adolescent finds a great way to practice taking their own decisions by making personal food choices. But unfortunately, wrong dietary choices lead to unsatisfactory nutritional status. AIM: To investigate the prevalence of six eating behaviours among adolescents. SUBJECTS AND METHODS: A case-control study was conducted on ninety Egyptian adolescents from 10 to 18 years old. Anthropometric measurements were taken. Body Mass Index (BMI) was calculated. The cases were forty-five children with body mass index ≥ 85th percentile. The control group involved forty-five of matched peers with body mass index ˂ 85th centile. A questionnaire form was constructed according to local customs in Egypt. RESULTS: Two unhealthy behaviours were mostly found in our study group. The first and the predominant one was multitasking while eating practised by 92.1% of candidates and showing the equal distribution in both groups. The second was skipping breakfast and was adopted by 51.7% of the study group with a significantly higher distribution in the ˂ 85th centile group. CONCLUSION: Faulty eating is a behaviour encountered in adolescence irrespective to BMI category. Thus, a normal BMI does not reflect healthy dietary behaviours

    Functionalized Fe3O4 Magnetic Nanoparticle Potentiometric Detection Strategy versus Classical Potentiometric Strategy for Determination of Chlorpheniramine Maleate and Pseudoephedrine HCl

    Nanosized adsorbents when used in potentiometric methods of analysis usually show better performance rather than the traditional potentiometric approach; this is attributed to the high specific surface area of the nanomaterial used in addition to the lack of internal diffusion resistance, thus improving their adsorption capacity. In the presented work, a rapid and sensitive potentiometric determination of chlorpheniramine maleate (CPM) and pseudoephedrine hydrochloride (PSE) in pure form, in pharmaceutical preparation, and in biological fluid was developed based on functionalized magnetic nanoparticles (Fe3O4). This strategy was compared with the classical potentiometric strategy. Three types of sensors were constructed using phosphotungstic acid (PTA), β-cyclodextrin (β-CD), and β-cyclodextrin-conjugated Fe3O4 magnetic nanoparticles for the potentiometric determination of each of CPM and PSE. The prepared sensors were characterized in regards to their composition, life duration, working pH range, and response time. The sensors have demonstrated promising selectivity to CPM and PSE in the presence of pharmaceutical formulation excipients, plasma matrix, and a diversity of both organic and inorganic interfering materials. The developed sensors have displayed good responses. Statistical comparison of the achieved results with a reported method has revealed no significant difference regarding both accuracy and precision

    Chromatographic methods for the simultaneous determination of binary mixture of Saxagliptin HCl and Metformin HCl

    Two chromatographic methods were suggested for the simultaneous determination of a binary mixture containing Saxagliptin HCl (SAG) and Metformin HCl (MET). First method was RP-HPLC method. Chromatographic separation was done on Kinetex⢠columnâC18 (4.6 Ã 150 mm, 2.6 µm) using mobile phase consisted of acetonitrile:phosphate buffer pH = 4.5 ± 0.1 adjusted with orthophosphoric acid (13:87, v/v). Isocratic elution at a flow rate 1.5 mL/min and UV detection at 220.0 nm was performed. Second method was spectro-densitometric method. Chromatographic separation was done on precoated silica gel aluminium plates 60 F254 as a stationary phase and developing system consisting of chloroform:methanol:formic acid (80:20:0.3, by volume). The density of the separated bands was measured by UV detector at 210.0 nm. The proposed methods were validated as per the ICH guidelines parameters like Linearity, precision, accuracy, selectivity, limit of detection and limit of quantitation. Statistical comparison was done between the obtained results and those obtained by the reported methods, showing no significant difference with respect to accuracy and precision.     Greenness assessment of two chromatographic methods developed for the determination of Mupirocin in two binary mixtures along with its impurity

    Abstract Two simple, accurate and precise chromatographic methods have been developed and validated for estimating Mupirocin (MUP) in two binary mixtures. Mixture (1); with Fluticasone propionate (FLU) together with two of their impurities, namely; Pseudomonic acid-D (Pseud-D) and Fluticasone impurity C (FIC). Mixture (2); with Mometasone furoate (MF) along with Pseud-D impurity. High performance thin layer chromatography (HPTLC-densitometry) and high performance liquid chromatography (RP-HPLC) were the two proposed methods. In the HPTLC method, good separation of both mixtures was achieved by using HPTLC plates pre-coated with silica gel 60 F254 as stationary phase and the mobile phase consisted of toluene: chloroform: ethanol at a ratio of (5: 4: 2, by volume). The detection was carried out at 220 nm for MUP and 254 nm for FLU, MF, Pseud-D and FIC. In the HPLC method, chromatographic separation was carried out using Agilent Eclipse XDB (250 mm×4.6 mm, 5 μm) C18 column. For mixture (1), a mobile phase of methanol: sodium di-hydrogen phosphate (pH 3.0) was applied in stepwise gradient elution starting at ratios of (50: 50, v/v) and then switching to (80: 20, v/v) after 7 min at a flow rate of 1 mL.min− 1. Detection was performed using diode array detector at 220 nm for MUP and Pseud-D and 240 nm for FLU and FIC. For mixture (2), the same mobile phase was used, but in isocratic elution in the ratio (80: 20, v/v) at flow rate of 1 mL.min− 1 and detection at 220 nm for MUP and Pseud-D and 248 nm for MF. The two methods successfully separated the cited drugs and were used to determine the drugs in pure form as well as pharmaceutical dosage forms. Validation was done as per International Council on Harmonization guidelines. Furthermore, the greenness of the proposed methods compared to the reported method, was evaluated as per the National Environmental Method Index, analytical Eco scale, Green Analytical Procedure Index and Analytical Greenness metric approaches

    Chemical Constitution and Antimicrobial Activity of Kefir Fermented Beverage

    Kefir beverage (KB) is a fermented milk initiated by kefir grains rich with starter probiotics. The KB produced in this study seemed to contain many chemical compounds elucidated by gas chromatography–mass spectrometry (GC-MS) and IR spectra. These compounds could be classified into different chemical groups such as alcohols, phenols, esters, fatty esters, unsaturated fatty esters, steroids, polyalkenes, heterocyclic compounds and aromatic aldehydes. Both KB and neutralized kefir beverage (NKB) inhibited some pathogenic bacteria including Escherichia coli ATCC11229 (E. coli), Listeria monocytogenes ATCC 4957 (L. monocytogenes), Bacillus cereus ATCC 14579 (B. cereus), Salmonella typhimurium ATCC 14028 (Sal. typhimurium) as well as some tested fungal strains such as Aspergillus flavus ATCC 16872 (A. flavus) and Aspergillus niger ATCC 20611 (A. niger), but the inhibitory activity of KB was more powerful than that obtained by NKB. It also appeared to contain four lactic acid bacteria species, one acetic acid bacterium and two yeast species. Finally, the KB inhibited distinctively both S. aureus and Sal. typhimurium bacteria in a brain heart infusion broth and in some Egyptian fruit juices, including those made with apples, guava, strawberries and tomatoes

    Karyotyping and neurodevelopmental follow-up of intracytoplasmic sperm injection children up to 4years of age

    Objectives: The aim of this work is to study ICSI babies, singleton and twins and compare them with singleton naturally conceived children from the genetic and neurodevelopmental point of view. Materials and methods: This is a prospective study performed on 120 children born to 100 mothers after the successful treatment of male infertility by intracytoplasmic sperm injection (ICSI). Obstetric history and Neonatal examinations were done. Developmental assessment using Denver Developmental Screening Test was done at 1, 2, 3, and 4years of age. Karyotype was done to all babies. The babies were divided into three study groups; group 1 single ICSI, group 2 twin ICSI, and group 3 normally conceived control group. Results: We found no statistically significant difference when we correlated outcome of DDST to gender, mode of delivery, smoking habits of mother, maternal illnesses, paternal smoking, Apgar score, congenital anomalies or type of feeding. There was significant statistical difference between the results of DDST follow-up and gestational age, birth weight and NICU admission. Our developmental assessment using DDST reported that ICSI children in both groups do not display any significant developmental delay when compared with naturally conceived children. Conclusion: Developmental assessment performed in this study revealed reassuring findings, no important differences between ICSI and NC children were noticed indicating that infertility treatment by ICSI technique does not appear to affect the development of child. It also highlights the need for continuous follow-up to evaluate whether ICSI children continue to show satisfactory development later in life

    Assessing the Antiproliferative Potential of a Novel Combretastatin A4 Derivative via Modulating Apoptosis, MAPK/ERK and PI3K/AKT Pathways in Human Breast Cancer Cells

    Background: Breast cancer is the most predominant tumor in women. Even though current medications for distinct breast cancer subtypes are available, the non-specificity of chemotherapeutics and chemoresistance imposes major obstacles in breast cancer treatment. Although combretastatin A-4 (CA-4) has been well-reported to have potential anticancer activity, in vivo studies of CA-4 reveal a decrease in its activity. In this respect, a series of CA-4 analogues have been designed, from which one analog [(1-(3-chloro-4-fluorophenyl)-N-(2methoxyphenyl)-5-(3,4,5-trimethoxyphenyl)-1H-1,2,4-triazole-3-carboxamide, C25H22ClFN4O5] showed drastic cytotoxicity against breast cancer cells. Therefore, this research focused on investigating the in vitro molecular mechanism underlying the cytotoxicity of the CA-4 analogue, particularly the MAPK/ERK as well as PI3K/AKT pathways as attractive therapeutic targets in breast cancer. Methods: The cell viability of MCF-7, MDA-MB231, and MDA-MB453 was assessed after treatment with the CA-4 analogue, and apoptosis was analyzed via Annexin V-FITC/PI dual staining. MAPK/ERK and PI3K/AKT were thoroughly assessed using western blotting. Real-time PCR was used to estimate apoptosis-related markers, including the P53, Bcl-2-associated X protein (Bax), and B-cell lymphoma 2 (Bcl2) genes. Results: The CA-4 analogue reduced the survival of all cancerous cells in a concentration-dependent manner and induced apoptosis through the mitochondrial pathway (39.89 ± 1.5%, 32.82 ± 0.6%, and 23.77 ± 1.1% in MCF-7, MDA-MB231, and MDA-MB453 cells), respectively. The analogue also attenuated the expression of pMEK1/2/t-MEK1/2, p-ERK1/2/t-ERK1/2, p-PI3K/t-PI3K, and p-AKT/t-AKT proteins in all three cancer cell lines in a time-dependent manner. Furthermore, the CA-4 analogue upregulated the expression of the P53 gene and dramatically increased the ratio of Bax/Bcl2 genes. Conclusions: The enhanced cytotoxicity can be attributed to substituting the hydroxyl group in CA-4 with chlorine in the meta-position of ring B, substituting the para-methoxy group in CA-4 with fluorine in the analogue, and lastly, introducing an extension to the compound’s structure (ring C). Therefore, CA-4 analogue can attenuate the proliferation of human breast cancer cells by inducing apoptosis and simultaneously suppressing the MAPK/ERK and PI3K/AKT pathways

    Determinants of Obtaining COVID-19 Vaccination among Health Care Workers with Access to Free COVID-19 Vaccination: A Cross-Sectional Study

    Introduction: Despite global efforts to contain the illness, COVID-19 continues to have severe health, life, and economic repercussions; thus, maintaining vaccine development is mandatory. Different directions concerning COVID-19 vaccines have emerged as a result of the vaccine’s unpredictability. Aims: To study the determinants of the attitudes of healthcare workers (HCWs) to receiving or refusing to receive the vaccine. Methods: The current study adopted an interviewed questionnaire between June and August 2021. A total of 341 HCWs currently working at Assiut University hospitals offered to receive the vaccine were included. Results: Only half of the HCWs (42%) accepted the COVID-19 vaccine. The most common reason that motivated the HCWs was being more susceptible than others to infection (71.8%). On other hand, the common reasons for refusing included: previously contracted the virus (64.8%); did not have time (58.8%); warned by a doctor not to take it (53.8%). Nearly one-third of nonaccepting HCWs depended on television, the Internet, and friends who refused the vaccine for information (p < 0.05). In the final multivariate regression model, there were six significant predictors: sex, job category, chronic disease, being vaccinated for influenza, and using Assiut University hospital staff and the Ministry of Health as sources of information (p < 0.05). Conclusion: Misinformation and negative conceptions are still barriers against achieving the desired rate of vaccination, especially for vulnerable groups such as HCWs
