Crystallization and preliminary X-ray analysis of the ternary complex of procarboxypeptidase A from bovine pancreas
AbstractThe ternary complex of procarboxypeptidase A, chymotrypsinogen C and proproteinase E from bovine pancreas has been crystallized using the sitting drop vapour diffusion method. The success in obtaining crystals has been found to be critically dependent on the prevention of autolysis of the complex. In preliminary stages, crystals twinned by merohedry were obtained from a solution containing MgCl2 and polyethylenglycol 400 as precipitating agent. Later on, untwinned ones could be grown employing CaCl2 instead of MgCl2. These latter crystals belong to the rhombohedral system and to the spacegroup R3 with cell dimensions a = b = 188.5 Å and c = 82.5 Å. Consideration of the possible values of Vm accounts for the presence of one ternary complex molecule-oligomere per asymmetric unit. The crystals diffract beyond 2.6 Å resolution and are suitable for X-ray analysis
Prevalence and risk factors for asthma, rhinitis, eczema, and atopy among preschool children in an Andean city.
BACKGROUND: Limited data are available on prevalence and associated risk factors for atopy and allergic diseases from high-altitude urban settings in Latin America. OBJECTIVE: To estimate the prevalence of atopy, asthma, rhinitis, and eczema, and associations with relevant risk factors in preschool children in the Andean city of Cuenca. METHODS: A cross-sectional study was undertaken using a representative sample of 535 children aged 3-5 years attending 30 nursery schools in the city of Cuenca, Ecuador. Data on allergic diseases and risk factors were collected by parental questionnaire. Atopy was measured by skin prick test (SPT) reactivity to a panel of relevant aeroallergens. Associations between risk factors and the prevalence of atopy and allergic diseases were estimated using multivariable logistic regression. RESULTS: Asthma symptoms were reported for 18% of children, rhinitis for 48%, and eczema for 28%, while SPT reactivity was present in 33%. Population fractions of asthma, rhinitis, and eczema attributable to SPT were 3.4%, 7.9%, and 2.9%, respectively. In multivariable models, an increased risk of asthma was observed among children with a maternal history of rhinitis (OR 1.85); rhinitis was significantly increased in children of high compared to low socioeconomic level (OR 2.09), among children with a maternal history of rhinitis (OR 2.29) or paternal history of eczema (OR 2.07), but reduced among children attending daycare (OR 0.64); eczema was associated with a paternal history of eczema (OR 3.73), and SPT was associated with having a dog inside the house (OR 1.67). CONCLUSIONS: A high prevalence of asthma, rhinitis, and eczema symptoms were observed among preschool children in a high-altitude Andean setting. Despite a high prevalence of atopy, only a small fraction of symptoms was associated with atopy. Parental history of allergic diseases was the most consistent risk factor for symptoms in preschool children
A computerized analysis of the entire retinal ganglion cell population and its spatial distribution in adult rats
AbstractIn adult albino (SD) and pigmented (PVG) rats the entire population of retinal ganglion cells (RGCs) was quantified and their spatial distribution analyzed using a computerized technique. RGCs were back-labelled from the optic nerves (ON) or the superior colliculi (SCi) with Fluorogold (FG). Numbers of RGCs labelled from the ON [SD: 82,818±3,949, n=27; PVG: 89,241±3,576, n=6) were comparable to those labelled from the SCi [SD: 81,486±4,340, n=37; PVG: 87,229±3,199; n=59]. Detailed methodology to provide cell density information at small scales demonstrated the presence of a horizontal region in the dorsal retina with highest densities, resembling a visual streak
Combining stable isotope analysis and conventional techniques to improve knowledge of the diet of the European Roller Coracias garrulus
Diet studies are crucial for understanding the ecology and evolution of species, as well as for establishing appropriate conservation and management strategies. However, they remain methodologically challenging due to variation between seasons, sites, sexes or age groups and even variation between individuals. Due to method-specific characteristics and biases, a combination of existing techniques can overcome the inherent limitations of each technique and provide a more accurate and broad picture of species’ food preferences. Here, we examine diet information obtained using three different assessment methods to better understand the trophic ecology of the European Roller Coracias garrulus, a species targeted by conservation measures in Europe. First, we analysed regurgitated pellets and video recordings to report the diet composition of adult and nestling Rollers, respectively. Secondly, we used stable isotope analysis (SIA) to investigate adult sexual diet segregation as well as to confirm the main findings regarding adult and nestling diets obtained through conventional methods. Based on the analysis of pellets, the diet of adult Rollers was dominated by Coleoptera, while camera images revealed that the diet of nestlings was dominated by Orthoptera, mainly grasshoppers and bush crickets. Blood isotopic signatures of adult and nestling Rollers confirmed the results obtained through pellet and video recording techniques. Of the 45 three methods, pellet analysis contained the most comprehensive trophic information regarding the detectable prey spectrum and prey species contribution, and also provided basic diet information to inform the SIA. Our results also highlight the potential of SIA for assessing intra-specific variation in diet by sampling individuals of known age and sex, which is often unfeasible through conventional approaches. SIA analysis showed no differences in δ13C and δ15N ratios of blood between males and females and a high degree of overlap amongst isotopic niches, suggesting no sex-specific partitioning in resource use. Overall, we showed that the combination of different methods could be used to gain new and clearer insights into avian trophic ecology that are essential for informing habitat management aiming to improve availability of foraging resources
Discovering Spatio-Temporal Patterns in Precision Agriculture Based on Triclustering
Agriculture has undergone some very important changes over
the last few decades. The emergence and evolution of precision agri culture has allowed to move from the uniform site management to the
site-specific management, with both economic and environmental advan tages. However, to be implemented effectively, site-specific management
requires within-field spatial variability to be well-known and character ized. In this paper, an algorithm that delineates within-field management
zones in a maize plantation is introduced. The algorithm, based on tri clustering, mines clusters from temporal remote sensing data. Data from
maize crops in Alentejo, Portugal, have been used to assess the suit ability of applying triclustering to discover patterns over time, that may
eventually help farmers to improve their harvests.Ministerio de Economía y Competitividad TIN2017-88209-C2Fundaçao para a Ciéncia e a Tecnologia (FCT) UIDB/04561/202
