    Experimental study of a TLP offshore floating wind turbine

    Tank testing in a wind and wave environment is a key part of the design process for the development of an offshore floating wind turbine. The current paper describes an extensive experiment campaign carried out at the Kelvin Hydrodynamics Laboratory at the University of Strathclyde to determine the hydrodynamic performance of the Iberdrola TLPWIND offshore floating wind turbine with the NREL 5MW reference turbine over a range of environmental conditions. Tests were carried out for 70m water depth and the deployment area selected as off Aberdeen, North Sea. The campaign included free oscillation tests, tests in regular waves and irregular waves, and additionally examined failure and accidental load cases

    The effect of spacing on the vortex-induced vibrations of two tandem flexible cylinders

    Vortex-induced vibrations (VIV) of two flexible cylinders arranged in tandem are studied using a two-way fluidstructure interaction (FSI) method with different spacing ratios (Sx/D) at Reynolds number Re = 500 using a twoway fluid-structure interaction (FSI) method. The main objective of this study is to investigate the effect of spacing on the hydrodynamic interactions and the VIV responses of these cylinders. The responses of the two flexible cylinders are found to be similar to the classical VIV responses at small Sx/D. Once Sx/D is large enough for the vortices to become detached from the upstream cylinder, the response of the upstream cylinder is similar to the typical VIV response whereas the downstream cylinder undergoes wake-induced vibration (WIV). The characteristics of the response of the downstream cylinder in the present study are similar to those of the first two response regimes classified by previous researchers. The third regime is not observed for the flexible downstream cylinder with both ends fixed. The two changes in the phase relation between the cross-flow displacements of the two tandem flexible cylinders are discovered to be linked with the initial-upper branch transition and the upperlower branch transition, respectively. The correlation lengths of the two flexible cylinders decrease significantly in the transition range between the upper and lower branches. Three modes of vortex shedding (2S, P + S and 2P) have been identified in the present study. The upper-branch 2P mode is found to be associated with largeamplitude vibration of the upstream cylinder and the P + S mode is observed to be related to large-amplitude vibration of the downstream cylinder for Sx/D = 3.5 and 5. On the other hand, the lower-branch 2P mode leads to small-amplitude vibration of the downstream cylinder in the post-lock-in range at Sx/D = 2.5. The relative phase shifts of the sectional lift coefficients on different spanwise cross sections can be attributed to the variation of the vortex shedding flow along the flexible cylinders, and these phase shifts result in poor phasing between the forces and the displacements which is related to the decrease of the correlation lengths

    Hypoxia induces a transcriptional early primitive streak signature in pluripotent cells enhancing spontaneous elongation and lineage representation in gastruloids

    The cellular microenvironment, together with intrinsic regulators, shapes stem cell identity and differentiation capacity. Mammalian early embryos are exposed to hypoxia in vivo and appear to benefit from hypoxic culture in vitro. Yet, how hypoxia influences stem cell transcriptional networks and lineage choices remain poorly understood. Here, we investigated the molecular effects of acute and prolonged hypoxia on embryonic and extra-embryonic stem cells as well as the functional impact on differentiation potential. We find a temporal and cell type-specific transcriptional response including an early primitive streak signature in hypoxic embryonic stem cells mediated by HIF1α. Using a 3D gastruloid differentiation model, we show that hypoxia-induced T expression enables symmetry breaking and axial elongation in the absence of exogenous WNT activation. When combined with exogenous WNT activation, hypoxia enhances lineage representation in gastruloids, as demonstrated by highly enriched signatures of gut endoderm, notochord, neuromesodermal progenitors and somites. Our findings directly link the microenvironment to stem cell function and provide a rationale supportive of applying physiological conditions in models of embryo development

    Sex and race and/or ethnicity differences in patients undergoing radiofrequency ablation for Barrett\u27s esophagus: results from the U.S. RFA Registry.

    BACKGROUND: Little is known about differences in Barrett\u27s esophagus (BE) characteristics by sex and race and/or ethnicity or these differences in response to radiofrequency ablation (RFA). OBJECTIVE: We compared disease-specific characteristics, treatment efficacy, and safety outcomes by sex and race and/or ethnicity in patients treated with RFA for BE. DESIGN: The U.S. RFA patient registry is a multicenter collaboration reporting processes and outcomes of care for patients treated with RFA for BE. PATIENTS: Patients enrolled with BE. INTERVENTIONS: RFA. MAIN OUTCOME MEASUREMENTS: We assessed safety (stricture, bleeding, perforation, hospitalization), efficacy (complete eradication of intestinal metaplasia [CEIM]), complete eradication of dysplasia, and number of treatments to CEIM by sex and race and/or ethnicity. RESULTS: Among 5521 patients (4052 men; 5126 white, 137 Hispanic, 82 African American, 40 Asian, 136 heritage not identified), women were younger (60.0 vs 62.1 years) and had shorter BE segments (3.2 vs 4.4 cm) and less dysplasia (37% vs 57%) than did men. Women were almost twice as likely to stricture (odds ratio 1.7; 95% confidence interval, 1.2-2.3). Although white patients were predominantly male, about half of African Americans and Asians with BE were female. African Americans and Asians had less dysplasia than white patients. Asians and African Americans had more strictures than did white patients. There were no sex or race differences in efficacy. LIMITATIONS: Observational study with non-mandated paradigms, no central laboratory for reinterpretation of pathology. CONCLUSION: In the U.S. RFA patient registry, women had shorter BE segments and less-aggressive histology. The usual tendency toward BE in men was absent in African Americans and Asians. Posttreatment stricture was more common among women and Asians. RFA efficacy did not differ by sex or race

    HIPK2 and extrachromosomal histone H2B are separately recruited by Aurora-B for cytokinesis

    Cytokinesis, the final phase of cell division, is necessary to form two distinct daughter cells with correct distribution of genomic and cytoplasmic materials. Its failure provokes genetically unstable states, such as tetraploidization and polyploidization, which can contribute to tumorigenesis. Aurora-B kinase controls multiple cytokinetic events, from chromosome condensation to abscission when the midbody is severed. We have previously shown that HIPK2, a kinase involved in DNA damage response and development, localizes at the midbody and contributes to abscission by phosphorylating extrachromosomal histone H2B at Ser14. Of relevance, HIPK2-defective cells do not phosphorylate H2B and do not successfully complete cytokinesis leading to accumulation of binucleated cells, chromosomal instability, and increased tumorigenicity. However, how HIPK2 and H2B are recruited to the midbody during cytokinesis is still unknown. Here, we show that regardless of their direct (H2B) and indirect (HIPK2) binding of chromosomal DNA, both H2B and HIPK2 localize at the midbody independently of nucleic acids. Instead, by using mitotic kinase-specific inhibitors in a spatio-temporal regulated manner, we found that Aurora-B kinase activity is required to recruit both HIPK2 and H2B to the midbody. Molecular characterization showed that Aurora-B directly binds and phosphorylates H2B at Ser32 while indirectly recruits HIPK2 through the central spindle components MgcRacGAP and PRC1. Thus, among different cytokinetic functions, Aurora-B separately recruits HIPK2 and H2B to the midbody and these activities contribute to faithful cytokinesis

    Quantitative trait loci conferring grain mineral nutrient concentrations in durum wheat 3 wild emmer wheat RIL population

    Mineral nutrient malnutrition, and particularly deficiency in zinc and iron, afflicts over 3 billion people worldwide. Wild emmer wheat, Triticum turgidum ssp. dicoccoides, genepool harbors a rich allelic repertoire for mineral nutrients in the grain. The genetic and physiological basis of grain protein, micronutrients (zinc, iron, copper and manganese) and macronutrients (calcium, magnesium, potassium, phosphorus and sulfur) concentration was studied in tetraploid wheat population of 152 recombinant inbred lines (RILs), derived from a cross between durum wheat (cv. Langdon) and wild emmer (accession G18-16). Wide genetic variation was found among the RILs for all grain minerals, with considerable transgressive effect. A total of 82 QTLs were mapped for 10 minerals with LOD score range of 3.2–16.7. Most QTLs were in favor of the wild allele (50 QTLs). Fourteen pairs of QTLs for the same trait were mapped to seemingly homoeologous positions, reflecting synteny between the A and B genomes. Significant positive correlation was found between grain protein concentration (GPC), Zn, Fe and Cu, which was supported by significant overlap between the respective QTLs, suggesting common physiological and/or genetic factors controlling the concentrations of these mineral nutrients. Few genomic regions (chromosomes 2A, 5A, 6B and 7A) were found to harbor clusters of QTLs for GPC and other nutrients. These identified QTLs may facilitate the use of wild alleles for improving grain nutritional quality of elite wheat cultivars, especially in terms of protein, Zn and Fe

    Mooring effect on wave frequency response of round shape FPSO

    This paper presents the motions response of moored Round Shape FPSO model due to the wave effect. The proposed of this research is analyzed the possibility of model motion response affected by different mooring system attach to the model. Model experiment was applied in this research to collect motion data for the study. Besides, the numerical simulation using diffraction potential, diffraction potential with Morison Heave correction and ANSYS frequency domain study were also applied in this research to generate comparative data to the experimental results. To investigate the effect of the mooring system to motion response, the model experiment was firstly conducted by attached model scale catenary mooring lines to Round Shape FPSO model. After that, the experiment was repeated by attached model scale taut mooring lines to the same model. The results obtain from the regular wave experiment tests and numerical simulation test were presented in this paper. This research concluded that the mooring systems would not give significant effect to wave frequency motion response after compared the motion result obtain from model experiment conducted using different mooring system
