    Prevalence and Antibiotic Sensitivity Testing of Bacteria Isolated from Open Wounds of Hospitalized Patients

    Background: Substantial infectivity of open wounds by microorganisms not only precipitates in terms of complexity, morbidity and mortality because of encouraging other fatal comorbidities but also becomes challenging to handle by medical practitioners.Methods: To ascertain the bioburden associated particularly with bacteria in open wounds, this research work was conducted at the Microbiology and Molecular Genetics department of Women University, Multan in association with Pathology Department of Nishtar Hospital and College, Multan from January to June 2018. 65 different wound samples were collected from different wards and analyzed by standard procedures for bacterial isolation and characterization, employing biochemical tests including catalase, oxidase, coagulase, motility, triple-sugar iron (TSI), citrate and indole tests. The isolated bacterial strains were also evaluated for sensitivity or resistance against twelve different antibiotics.Results: All the 65 samples were found to be positive for bacterial presence. Gram-positive cocci constituted 63% of the isolates and gram-negative rods comprised 37%. Biochemical tests revealed that the predominant pathogen was Staphylococcus aureus (63.1%) followed by Proteus spp., (15.4%), Pseudomonas aeruginosa (12.3%) and Escherichia coli (9.2%), respectively. Antibiotic sensitivity testing disclosed that the most effective antibiotics against these isolates were Tigecycline and Polymyxin B while the least effective antibiotics were Ceftazidime and Ampicillin.Conclusion: These findings can prove beneficial in understanding the prevalence of various bacteria in wound infections. In addition, they indicate the need to develop and implement antibiotic stewardship programs so as to combat drug resistance among pathogens.Keywords: Gram-positive cocci; Drug resistance; Staphylococcus aureus   

    Assessment of fish oil to check the stability and meat quality of some commercially available tin packed fish in Islamabad, Pakistan

    Fish contains all the essential nutrients which is important for human health so it is important to analyse all the nutrients present in tin packed fish meat. Keeping in view its importance present study is conducted on the assessment of fish oil to check the stability and quality of fish meat by proximate analysis of commercially available tin packed fish. Four tin packed fish species i.e., Sardine (Sardinella longiceps), Red salmon (Oncorhynchus nerka), Pink salmon (Oncorhynchus gorbuscha) and Skipjack tuna (Katsuwonus pelamis) were collected from different supermarkets of Islamabad, Pakistan. Proximate analysis viz., crude protein, crude fat, moisture and ash contents of the fish meat has been done to evaluate the meat quality. The antioxidant activity in oil was also analysed by FRAP assay. The result indicated that maximum percentage of moisture i.e., 78.61% present in Skipjack Tuna meat, Pink Salmon contain highest percentage of crude protein i.e., 70.00%, Red Salmon contain highest percentage of crude fat i.e., 30.00% while Sardine and Skipjack Tuna contains highest percentage of ash contents i.e., 8.00% and the total antioxidant capacity (uM) is higher in oil of Red Salmon (24.35%) followed by Sardines (14.78%), Skipjack Tuna (9.86%) and Pink Salmon (9.48%).     Comparative Antibacterial Analysis of Four Different Medicinal Plants Against Human Skin Flora

    Background: As we are facing continuously increasing antibiotic resistance, increased infections, diseases and pandemics, the researching community is turning to find the solutions from nature and plants play a vital role in this scenario. Pakistan due to its unique geography and a variety of climatic zones harbors a huge floral diversity.Methods: In this study extracts of four plants of Pakistan namely Azadirachta indica, Curcuma longa, Moringa oleifera and Eucalyptus camaldulensis prepared by Aqueous. Ethanol and Methanol extract preparation methods were examined for their phytochemical content by employing various reagents and methods like foam test for saponins, xanthoprotein test for proteins, Braemar’s test for tannins etc. Antibacterial activity against stored human skin flora samples was performed using the agar well diffusion assay and statistical analysis was performed using ANOVA one way analysis on Microsoft Excel 2010.Result: Each plant extract exhibited antibacterial activity and statistical analysis performed for extracts of each plant showed p value for A. indica as 0.36 (P > 0.05), for C. longa as 0.09 (P > 0.05), for M. oleifera as 0.67 (P > 0.05) and for E. camaldulensis as 0.02 (P < 0.05), which indicates significant antibacterial activity of E. camaldulensis depending on the size of zones of inhibition produced by it. Conclusion: All the extracts prepared from selected plants showed some degree of antibacterial activity against the human skin flora that can also act as opportunistic pathogen. This supports the use of traditionally used plants and herbs for fighting present day soaring health related issues as antimicrobial resistance. Study also suggests further investigations for estimating exact concentrations of extracts to be used that work efficiently in relevant cases and molecular mechanism of action of these extracts.Keywords: Skin Flora; Antibiotic Resistant; Plant Extract; Antibacterial Activity, Medicinal plants   

    Estimation of Genetic Divergence in 40 Elite Cotton Germplasm

    Background: Identification and development of superior cotton genotypes and their further improvement has been one of the primitive aims of plant breeding programmers. Therefore, necessity of analyzing the agro-morphological and yield attributes of advanced lines of cotton is doubtless.Methods: Mean performance and correlation between ten different morphological, yield and fiber related attributes of forty cotton genotypes from all over the Pakistan under National Coordinated Varietal Trial were studied at Central Cotton Research Institute of Multan. Selected traits included several sympodial and monopodial branches, plant height, number of bolls per plant, staple length, seed cotton yield, boll weight, fiber strength, percentage ginning out turn and micronaire value.Results: Statistical analysis of variance disclosed highly significant (p<0.01) differences among all cotton genotypes for majority of the characteristics. Basic descriptive statistical analysis of selected agronomic traits revealed the presence of substantial genetic variation among 40 genotypes of cotton for 10 selected traits. The correlation coefficient was determined both at p<0.05 and p<0.01 levels and the observations demonstrated that some of the characteristics exhibited positive correlation, while others displayed negative correlation with each other. Micronaire showed highly significant positive association with percentage ginning out turn (0.3412) and boll weight (0.2421 g) as well as highly significant negative association with fiber strength (-0.5973).Conclusion: Convincingly, mean performances and correlation of different traits with one another can be utilized in cotton breeding programs in future to improve the yield of cotton seed and to select fiber related attributes with desired characteristics.Keywords: Gossypium hirsutum; Morphological agronomic traits; Statistical analysi

    A Review on Psychosocial Distress among General Population of Pakistan During Covid-19 Pandemic

    The COVID-19 crisis in 2019 significantly impacted human life all over the globe. Psychological health of general population has been affected by the unpredictable circumstances and impulsion of living in isolation, if suspected positive for coronavirus. During COVID-19 outbreak, escalation of various types of psychosocial distresses such as anxiety, sleep disorders and depression has been observed in the general population globally. Thus, there is an urgent need to combat these issues through well planned strategies. The focus of this review is to conduct a thorough evaluation of the available research on the effects of COVID-19 pandemic on the mental status of the general population of Pakistan. We also focused on the psychosocial impact of lockdown on the general community and the significance of protective and risk variables in the development of mental conditions in susceptible groups of population. The review further evaluates the strategies which can be employed to minimize the negative impact of COVID-19-related stress and improve the mental wellbeing of general community in Pakistan by highlighting the significant repercussions of existing research studies

    Development and quality evaluation of dried vegetable soup added with some legumes

    Fruits and vegetables play an important role in the diet due to their high amounts of fiber minerals and vitamin content. Different vegetables i.e., potatoes, carrot, onion, garlic, tomato with different legumes such as lentils and green peas with addition of some seasonings like salt black pepper coriander and cumin was used to prepare dried vegetable soup. Rehydration ratio, physiochemical analysis, sensory evaluation and net number of calories obtain per serving of soup was accomplished. pH of vegetable soup decreased slightly during storage i.e. (6.40±0.32, 6.31±0.08 6.27±0.26)&nbsp;&nbsp; and for acidity they increased slightly i.e. (2.28-2.28) that is good for soup shelf life. Color analysis gave précised data about soup sensory evaluation where mean values of L*a*b* are increasing gradually i.e. (81.72, 81.87±0.21, 81.94±0.03±0.20), (5.61±0.09, 5.57±0.18, 5.59±0.22) and (29.54±0.12, 29.43±0.10, 29.53±0.05). The purpose of adding legumes is to increase the nutritional content up to a mark able extent, where the soup mixes will provide noticeable amounts of the desired nutrients particularly, from vegetables and protein (12.72±0.22, 12.54±0.16, 12.63±0.15) from legumes, with better protein digestibility and mineral accessibility. Water activity and amount of moisture (9.14±0.09, 9.23±0.03, 9.45±0.18) of final product increased stability of the dried soup mixes.&nbsp

    Efficacy of cell-free DNA as a diagnostic biomarker in breast cancer patients

    Abstract Breast cancer is the most prevalent and leading cause of mortality worldwide among women. Cell-free DNA (cfDNA) analysis is an alternative quantitative approach to conventional methods for cancer diagnosis. The current research project aimed to determine the efficacy of cfDNA as a diagnostic biomarker in breast cancer patients in Pakistan. Eighty-four female breast cancer patients were selected as cases, and 152 healthy females as controls. Immunohistochemistry was performed to identify tumor biomarkers along with clinical profiling. cfDNA was extracted from serum using the phenol–chloroform method. The cfDNA level in the serum was estimated using Agarose Gel Electrophoresis and Nanodrop. SPPS version 25.0 was used to perform statistical analyses. The results showed that the cancer biomarkers were significantly associated with breast cancer. The changes in hematological parameters were insignificant, whereas the biochemical parameter variations between the cases and controls were statistically significant. A significant association of cfDNA level with breast cancer was observed. Further cfDNA levels and cancer biomarkers were not statistically significant. A significant correlation was observed between cfDNA and biochemical parameters, except for creatinine, whereas hematological parameters showed no significant correlation.ROC analysis declared cfDNA as an authentic diagnostic marker for breast cancer.
