    Exosomes isolation and characterization in serum is feasible in non-small cell lung cancer patients: critical analysis of evidence and potential role in clinical practice

    Exosomes are nano-sized vesicles of endolysosomal origin, released by several cytotypes in physiological and pathological conditions. Tumor derived exosomes, interacting with other cells of the tumor microenvironment, modulate tumor progression, angiogenic switch, metastasis, and immune escape. Recently, extracellular vesicles were proposed as excellent biomarkers for disease monitoring and prognosis in cancer patients. Non-small cell lung cancer (NSCLC) has a poor 5-year survival rate due to the delay in the detection of the disease. The majority of patients are diagnosed in an advanced disease stage. Exosomes might be promising beneficial tools as biomarker candidates in the scenario of NSCLC, since they contain both, proteins and miRNAs. The clinical case reported in this manuscript is a proof of concept revealing that NSCLC exosomes and sorted miRNAs might constitute, in a near future, novel biomarkers.     Inheritance of DNA Transferred from American Trypanosomes to Human Hosts

    Interspecies DNA transfer is a major biological process leading to the accumulation of mutations inherited by sexual reproduction among eukaryotes. Lateral DNA transfer events and their inheritance has been challenging to document. In this study we modified a thermal asymmetric interlaced PCR by using additional targeted primers, along with Southern blots, fluorescence techniques, and bioinformatics, to identify lateral DNA transfer events from parasite to host. Instances of naturally occurring human infections by Trypanosoma cruzi are documented, where mitochondrial minicircles integrated mainly into retrotransposable LINE-1 of various chromosomes. The founders of five families show minicircle integrations that were transferred vertically to their progeny. Microhomology end-joining of 6 to 22 AC-rich nucleotide repeats in the minicircles and host DNA mediates foreign DNA integration. Heterogeneous minicircle sequences were distributed randomly among families, with diversity increasing due to subsequent rearrangement of inserted fragments. Mosaic recombination and hitchhiking on retrotransposition events to different loci were more prevalent in germ line as compared to somatic cells. Potential new genes, pseudogenes, and knockouts were identified. A pathway of minicircle integration and maintenance in the host genome is suggested. Thus, infection by T. cruzi has the unexpected consequence of increasing human genetic diversity, and Chagas disease may be a fortuitous share of negative selection. This demonstration of contemporary transfer of eukaryotic DNA to the human genome and its subsequent inheritance by descendants introduces a significant change in the scientific concept of evolutionary biology and medicine

    Understanding water and energy fluxes in the Amazonia: Lessons from an observation-model intercomparison

    Tropical forests are an important part of global water and energy cycles, but the mechanisms that drive seasonality of their land-atmosphere exchanges have proven challenging to capture in models. Here, we (1) report the seasonality of fluxes of latent heat (LE), sensible heat (H), and outgoing short and longwave radiation at four diverse tropical forest sites across Amazonia—along the equator from the Caxiuanã and Tapajós National Forests in the eastern Amazon to a forest near Manaus, and from the equatorial zone to the southern forest in Reserva Jaru; (2) investigate how vegetation and climate influence these fluxes; and (3) evaluate land surface model performance by comparing simulations to observations. We found that previously identified failure of models to capture observed dry-season increases in evapotranspiration (ET) was associated with model overestimations of (1) magnitude and seasonality of Bowen ratios (relative to aseasonal observations in which sensible was only 20%–30% of the latent heat flux) indicating model exaggerated water limitation, (2) canopy emissivity and reflectance (albedo was only 10%–15% of incoming solar radiation, compared to 0.15%–0.22% simulated), and (3) vegetation temperatures (due to underestimation of dry-season ET and associated cooling). These partially compensating model-observation discrepancies (e.g., higher temperatures expected from excess Bowen ratios were partially ameliorated by brighter leaves and more interception/evaporation) significantly biased seasonal model estimates of net radiation (Rn), the key driver of water and energy fluxes (LE ~ 0.6 Rn and H ~ 0.15 Rn), though these biases varied among sites and models. A better representation of energy-related parameters associated with dynamic phenology (e.g., leaf optical properties, canopy interception, and skin temperature) could improve simulations and benchmarking of current vegetation–atmosphere exchange and reduce uncertainty of regional and global biogeochemical models

    Evolution and Nucleosynthesis of Zero Metal Intermediate Mass Stars

    New stellar models with mass ranging between 4 and 8 Mo, Z=0 and Y=0.23 are presented. The models have been evolved from the pre Main Sequence up to the Asymptotic Giant Branch (AGB). At variance with previous claims, we find that these updated stellar models do experience thermal pulses in the AGB phase. In particular we show that: a) in models with mass larger than 6 Mo, the second dredge up is able to raise the CNO abundance in the envelope enough to allow a "normal" AGB evolution, in the sense that the thermal pulses and the third dredge up settle on; b) in models of lower mass, the efficiency of the CNO cycle in the H-burning shell is controlled by the carbon produced locally via the 3alpha reactions. Nevertheless the He-burning shell becomes thermally unstable after the early AGB. The expansion of the overlying layers induced by these weak He-shell flashes is not sufficient by itself to allow a deep penetration of the convective envelope. However, immediately after that, the maximum luminosity of the He flash is attained and a convective shell systematically forms at the base of the H-rich envelope. The innermost part of this convective shell probably overlaps the underlying C-rich region left by the inter-shell convection during the thermal pulse, so that fresh carbon is dredged up in a "hot" H-rich environment and a H flash occurs. This flash favours the expansion of the outermost layers already started by the weak thermal pulse and a deeper penetration of the convective envelope takes place. Then, the carbon abundance in the envelope rises to a level high enough that the further evolution of these models closely resembles that of more metal rich AGB stars.     Widespread reduction in sun-induced fluorescence from the Amazon during the 2015/2016 El Nino

    The tropical carbon balance dominates year-to-year variations in the CO2 exchange with the atmosphere through photosynthesis, respiration and fires. Because of its high correlation with gross primary productivity (GPP), observations of sun-induced fluorescence (SIF) are of great interest. We developed a new remotely sensed SIF product with improved signal-to-noise in the tropics, and use it here to quantify the impact of the 2015/2016 El Nino Amazon drought. We find that SIF was strongly suppressed over areas with anomalously high temperatures and decreased levels of water in the soil. SIF went below its climatological range starting from the end of the 2015 dry season (October) and returned to normal levels by February 2016 when atmospheric conditions returned to normal, but well before the end of anomalously low precipitation that persisted through June 2016. Impacts were not uniform across the Amazon basin, with the eastern part experiencing much larger (10-15%) SIF reductions than the western part of the basin (2-5%). We estimate the integrated loss of GPP relative to eight previous years to be 0.34-0.48 PgC in the three-month period October-November-December 2015.     Com o objetivo de avaliar o desenvolvimento de duas esp\ue9cies arb\uf3reas nativas - Anadenanthera peregrina (angico-vermelho) e Schinus terebinthifolius (aroeira-vermelha) - submetidas a doses de fertilizante de libera\ue7\ue3o lenta na formula\ue7\ue3o 13-06-16, conduziu-se um experimento em viveiro utilizando uma mistura de mat\ue9rias-primas com composto org\ue2nico, vermiculita e Plantmax\uae para compor substrato base. O experimento foi conduzido no delineamento experimental inteiramente casualizado com seis tratamentos em quatro repeti\ue7\uf5es. Os tratamentos foram: T1\u20130 kg (testemunha); T2\u20132 kg; T3\u20134 kg; T4\u20136 kg; T5\u20138 kg e T6\u201310 kg de fertilizante de libera\ue7\ue3o lenta por metro c\ufabico de substrato base. Avaliou-se aos 189 dias ap\uf3s a semeadura a altura total, di\ue2metro do colo, biomassa fresca da parte a\ue9rea, biomassa seca da parte a\ue9rea, biomassa seca da raiz, biomassa seca total, rela\ue7\ue3o entre altura e di\ue2metro do colo e o \uedndice de qualidade de Dickson. Os resultados evidenciam que a aplica\ue7\ue3o de fertilizante de libera\ue7\ue3o lenta leva a ganhos significativos de crescimento em ambas as esp\ue9cies, sendo as melhores doses 6,68 a 5,54 kg m-3 para a esp\ue9cie Anadenanthera peregrina, e 8,38 a 14,42 kg m-3 para Schinus terebinthifolius.In order to assess the development of two native species, Anadenanthera peregrina and Schinus terebinthifolius , submitted to doses of slow-release fertilizer formulation 13-06-16, an experiment was conducted in greenhouse using a mixture of raw materials with organic compost, vermiculite and plantmax to compose base substrate. The experiment was conducted in a completely randomized design with six treatments and four replications. The treatments were : T1-0 kg (control), T2-2 kg, 4 kg-T3 , T4-6 kg, T5-8 kg and T6 \u2013 10 kg slow release fertilizer per cubic meter of base substrate . Was evaluated at 189 days after sowing the total height, stem diameter, fresh weight of shoot, dry shoot biomass, root dry biomass, total biomass, the relationship between height and stem diameter and quality index Dickson. The results suggest that application of slow release fertilizer leads to significant gains in growth in both species, with the best doses from 6.68 to 5.54 kg m-3 for Anadenanthera peregrina species, and from 8.38 to 14,42 kg m-3 for Schinus terebinthifolius

    Diversity across major and candidate genes in European local pig breeds

    The aim of this work was to analyse the distribution of causal and candidate mutations associated to relevant productive traits in twenty local European pig breeds. Also, the potential of the SNP panel employed for elucidating the genetic structure and relationships among breeds was evaluated. Most relevant genes and mutations associated with pig morphological, productive, meat quality, reproductive and disease resistance traits were prioritized and analyzed in a maximum of 47 blood samples from each of the breeds (Alentejana, Apulo-Calabrese, Basque, Bísara, Majorcan Black, Black Slavonian (Crna slavonska), Casertana, Cinta Senese, Gascon, Iberian, Krškopolje (Krškopoljski), Lithuanian indigenous wattle, Lithuanian White Old Type, Mora Romagnola, Moravka, Nero Siciliano, Sarda, Schwäbisch-Hällisches Schwein (Swabian Hall pig), Swallow-Bellied Mangalitsa and Turopolje). We successfully analyzed allelic variation in 39 polymorphisms, located in 33 candidate genes. Results provide relevant information regarding genetic diversity and segregation of SNPs associated to production and quality traits. Coat color and morphological trait-genes that show low level of segregation, and fixed SNPs may be useful for traceability. On the other hand, we detected SNPs which may be useful for association studies as well as breeding programs. For instance, we observed predominance of alleles that might be unfavorable for disease resistance and boar taint in most breeds and segregation of many alleles involved in meat quality, fatness and growth traits. Overall, these findings provide a detailed catalogue of segregating candidate SNPs in 20 European local pig breeds that may be useful for traceability purposes, for association studies and for breeding schemes. Population genetic analyses based on these candidate genes are able to uncover some clues regarding the hidden genetic substructure of these populations, as the extreme genetic closeness between Iberian and Alentejana breeds and an uneven admixture of the breeds studied. The results are in agreement with available knowledge regarding breed history and management, although largest panels of neutral markers should be employed to get a deeper understanding of the population's structure and relationships

    Climate control of terrestrial carbon exchange across biomes and continents

    Understanding the relationships between climate and carbon exchange by terrestrial ecosystems is critical to predict future levels of atmospheric carbon dioxide because of the potential accelerating effects of positive climate-carbon cycle feedbacks. However, directly observed relationships between climate and terrestrial CO2exchange with the atmosphere across biomes and continents are lacking. Here we present data describing the relationships between net ecosystem exchange of carbon (NEE) and climate factors as measured using the eddy covariance method at 125 unique sites in various ecosystems over six continents with a total of 559 site-years. We find that NEE observed at eddy covariance sites is (1) a strong function of mean annual temperature at mid- and high-latitudes, (2) a strong function of dryness at mid- and low-latitudes, and (3) a function of both temperature and dryness around the mid-latitudinal belt (45°N). The sensitivity of NEE to mean annual temperature breaks down at ∼16 ®C (a threshold value of mean annual temperature), above which no further increase of CO,.2uptake with temperature was observed and dryness influence overrules temperature influence. © 2010 lOP Publishing Ltd

    Malaria is a leading cause of mortality, particularly in sub-Saharan African children. Prompt and efficacious treatment is important as patients may progress within a few hours to severe and possibly fatal disease. Chlorproguanil-dapsone-artesunate (CDA) was a promising artemisinin-based combination therapy (ACT), but its development was prematurely stopped because of safety concerns secondary to its associated risk of haemolytic anaemia in glucose-6-phosphate dehydrogenase (G6PD)-deficient individuals.
