    Site-specific mutagenesis of human interleukin-6 and its biological activity

    AbstractAmino acid substitutions of human interleukin-6 (IL-6) were performed. Single substitution Met162 → Ala and double substitutions Leu159, 166 → Val resulted in a significant decrease of IL-6 activity in the production of immunoglobulin (lg) from B-cells. Single substitution Leu166→Val or Leu159→Val gave a slight or no significant decrease in the Ig-induction activity, respectively. The receptor-binding activity of each IL-6 mutant was also examined. It was observed that the decrease of the receptor-binding activity was generally in parallel with that of the Ig-induction activity. We therefore suggest that hydrophobic side-chains existing in Met162, Leu159, and Leu164 are significantly involved in the receptor-binding of IL-6

    Operational status of the superconducting system for LHD

    Large Helical Device (LHD) is a heliotron-type experimental fusion device which has the capability of confining current-less and steady-state plasma. The primary feature on the engineering aspect of LHD is using superconducting (SC) coils for magnetic confinement: two pool boiling helical coils (H1, H2) and three pairs of forced-flow poloidal coils (IV, IS, OV). These coils are connected to the power supplies by SC bus-lines. Five plasma experimental campaigns have been performed successfully in four years from 1998. The fifth operation cycle started in August 2001 and finished in March 2002. We have succeeded to obtain high plasma parameters such as 10 keV of electron temperature, 5 keV of ion temperature and beta value of 3.2%. The operational histories of the SC coils, the SC bus-lines and the cryogenic system have been demonstrating high reliability of the large scale SC system. The operational status and the results of device engineering experiments are summarized

    Solitary Cardiac Metastasis of Hepatocellular Carcinoma

    Cardiac metastasis originating from hepatocellular carcinoma (HCC) is a rare condition with a poor prognosis. No therapeutic standards for cardiac metastasis originating from HCC have been established. At 19 months after a curative hepatectomy, a 64-year-old Japanese hepatitis B virus-positive male patient experienced solitary cardiac metastasis originating from HCC. The cardiac tumor was discovered in the right ventricle. The patient received three courses of radiotherapy and chemotherapy and survived > 3 years after the initial diagnosis of cardiac metastasis. His case demonstrates that radiotherapy combined with chemotherapy can be an effective treatment for cardiac metastasis

    Analysis on the cryogenic stability and mechanical properties of the LHD helical coils

    Transient normal-transitions have been observed in the superconducting helical coils of LHD. Propagation of a normal-zone is analyzed with a numerical simulation code that deals with the magnetic diffusion process in a pure aluminum stabilizer. During excitation tests, a number of spike signals are observed in the balance voltage of the helical coils, which seem to be caused by mechanical disturbances. The spike signals are analyzed by applying pulse height analysis and the mechanical properties of the coil windings are investigated

    Stability and safety estimates and tests of a superconducting bus-line for large-scale superconducting coils

    We have been developing a flexible superconducting bus-line as a unit electrical feeder between large-scale superconducting coils and their power supplies away from the coils. The designed superconducting bus-line consists of a pair of +/- aluminum stabilized NbTi/Cu compacted strand cables and a coaxial four-channel transfer line. A full-scale model of the SC bus-line (20 m long) has been constructed and tested successfully up to 40 kA without a quench under the short-circuit condition. Stability tests were also done by inducing a forced quench with heaters. A minimum propagation current larger than 32.5 kA was confirmed. Thus, the bus-line was cryogenically stabilized at the rated current of 30 kA. We have examined the test results and evaluated the stability and safety margins of this bus-line. The design criteria for a superconducting bus-line are also shown for large-scale superconducting coils with operating current as a parameter

    Asymmetrical normal-zone propagation observed in the aluminum-stabilized superconductor for the LHD helical coils

    Transient normal-transitions have been observed in the superconducting helical coils of the Large Helical Device (LHD). Stability tests have been performed for an R&D coil as an upgrading program of LHD, and we observed asymmetrical propagation of an initiated normal-zone. In some conditions, a normal-zone propagates only in one direction along the conductor and it hence forms a traveling normal-zone. The Hall electric field generated in the longitudinal direction in the aluminum stabilizer is a plausible candidate to explain the observed asymmetrical normal-zone propagation

    Normal transitions and a subsequent quench were experienced with the pool-cooled helical coils of the Large Helical Device (LHD) during its excitation test. Although the initiated normal zone once started to recover, a disruptive transverse propagation followed and triggered an emergency discharging program. The cryogenic stability of the composite-type superconductor has been studied by sample experiments as well as by numerical calculations. Due to the rather long magnetic diffusion time constant in the pure Al stabilizer, transient stability of the conductor seems to play an important role for driving finite propagation of a normal zone. The cause of the final quench is also discussed from the viewpoint of cooling deterioration due to a possible accumulation of He bubble

    Study on development policy for new cryogenic structural material for superconducting magnet of fusion reactor

    A fusion DEMO will require large-scale cryogenic structure including TF coil cases. Because of huge electromagnetic forces, extra thick plates and/or wrought products will be supplied. Since the midsection of the huge block is weaker than the block surface region, the design yield stress must be determined taking account of this lower strength part. To search the manufacturing process to improve the midsection strength, the crystal refinement strengthening and the precipitation strengthening are considered together with the carbon and nitrogen solid solution strengthening. XM-19 was focused based on the variation of the yield stress and the fracture toughness, a 100 mm thick block and a 30 mm thick plate were trial produced, and strength and the fracture toughness at the midsection were evaluated. This study will present the experimental data and discuss the development policy for a new cryogenic structural material for a fusion reactor
