Effect of buffer on heparin binding and sensing in competitive aqueous media
Although buffer-specific effects on molecular recognition are known in biological science, they remain rare in supramolecular chemistry. The binding between a cationic dye, Mallard Blue (MalB), and polyanionic heparin in aqueous NaCl (150 mM) is studied in three commonly-used buffers (Tris-HCl, HEPES, Phosphate, each 10 mM). Although MalB has a very similar UV-Vis spectrum in each buffer, the sensory response towards heparin was different in each case. This can be ascribed to differences in the complex formed. In Tris-HCl which has the least competitive chloride counter-anions, MalB exhibits a hypsochromic shift of 25 nm, assigned to strong binding and aggregation of the dye on heparin. In more competitive HEPES, containing a sulfonate anion, there is weaker binding and less aggregation of MalB along the heparin; the hypsochromic shift is only 15 nm. In phosphate buffer, MalB can interact quite strongly with buffer phosphate anions; although heparin binding is still observed, the hypsochromic shift associated with dye aggregation is only 5 nm. As such, specific buffer interactions with the MalB-heparin complex mediate host-guest binding and sensing. Buffer choice must be made carefully in studies of molecular recognition – we would caution against using phosphate and sulfonate containing buffers when studying electrostatic binding
Rare Kaon Decays Revisited
We present an updated discussion of K\ra\pi \bar{l} l decays in a combined
framework of chiral perturbation theory and Large-- QCD, which assumes the
dominance of a minimal narrow resonance structure in the invariant mass
dependence of the pair. The proposed picture reproduces very well,
both the experimental K^+\ra\pi^+ e^+ e^- decay rate and the invariant mass spectrum. The predicted Br(K_S\ra\pi^0 e^+ e^-) is, within errors,
consistent with the recently reported result from the NA48 collaboration.
Predictions for the K\ra\pi \mu^{+}\mu^{-} modes are also obtained. We find
that the resulting interference between the {\it direct} and {\it indirect}
CP--violation amplitudes in K_L\ra\pi^0 e^+ e^- is constructive
Percutaneous aortic valve replacement: valvuloplasty studies in vitro
Objective: Valvuloplasty of the aortic valve is currently used in selected patients for severe calcified aortic valve disease, but clinical effectiveness is low and complication rate remains high. In this study, the total particle load after valvuloplasty and the embolization risk of calcific debris into the coronary arteries was analyzed in an in vitro model. Methods: Three highly calcified human aortic leaflets have been sutured into a porcine annulus (N=9). Both coronary arteries were separated and each was anastomized to a silicon line, which was drained off into a measuring beaker. Then valvuloplasty was performed (Thyshak II, 20mm, 1.5atm). After removal of the balloon, 100ml of sodium chloride solution irrigated the ascending aorta. After passing through the separated coronary arteries, the solution was filtered (filter size 0.45μm), dried, and the total amount of particles was analyzed microscopically. Results: Nine experiments were analyzed. After valvuloplasty, all hearts showed a median of 18 particles larger than 1mm in the coronary arteries (range 0-307). The amount of particles smaller than 1mm was 6574 (median, range 2207-14200). In five cases, coronary arteries were completely occluded by bulky particles. Conclusion: This study demonstrated a large amount of calcific particles after valvuloplasty with a consequently high risk for coronary embolic events in case of highly calcified aortic valves. In times of valvuloplasty rediscovering as part of transcatheter valve implantation, the risk of embolization should be taken into consideration and filtering techniques have to be develope
Differential Regulation of Bcl-2 and Bax Expression in Cells Infected with Virulent and Nonvirulent Strains of Sindbis Virus
AbstractSindbis virus is an alphavirus that infects cells in either lytic or persistent infection. In this study we examined the effects of Sindbis virus on cell apoptosis and on the expression of Bcl-2 and Bax. Of the two strains studied, SVA and SVNI, only the neurovirulent strain, SVNI, induced apoptosis of astrocytes and PC-12 cells. SVA, which infects cells in a persistent manner, induced up-regulation of bcl-2 mRNA and Bcl-2 protein, whereas SVNI induced an increase in Bax levels. Our results indicate a differential regulation of Bcl2 and Bax expression by SVA and SVNI, which may be associated with the apoptotic potential of the viruses
Influence of race/ethnicity on response to lupus nephritis treatment: the ALMS study.
OBJECTIVE: To compare the efficacy and safety of mycophenolate mofetil (MMF) and intravenous cyclophosphamide (IVC) as induction treatment for lupus nephritis (LN), by race, ethnicity and geographical region. METHODS: A total of 370 patients with active Class III-V LN received MMF (target dose 3.0 g/day) or IVC (0.5-1.0 g/m(2)/month), plus tapered prednisone, for 24 weeks. Renal function, global disease activity, immunological complement (C3 and C4) and anti-dsDNA levels are the outcomes that were assessed in this study. RESULTS: MMF was not superior to IVC as induction treatment (primary objective). There were important pre-specified interactions between treatment and race (P = 0.047) and treatment and region (P = 0.069) (primary endpoint). MMF and IVC response rates were similar for Asians (53.2 vs 63.9%; P = 0.24) and Whites (56.0 vs 54.2%; P = 0.83), but differed in the combined Other and Black group (60.4 vs 38.5%; P = 0.03). Fewer patients in the Black (40 vs 53.9%; P = 0.39) and Hispanic (38.8 vs 60.9%; P = 0.011) groups responded to IVC. Latin American patients had lower response to IVC (32 vs 60.7%; P = 0.003). Baseline disease characteristics were not predictive of response. The incidence of adverse events (AEs) was similar across groups. Serious AEs were slightly more prevalent among Asians. CONCLUSIONS: MMF and IVC have similar efficacy overall to short-term induction therapy for LN. However, race, ethnicity and geographical region may affect treatment response; more Black and Hispanic patients responded to MMF than IVC. As these factors are inter-related, it is difficult to draw firm conclusions about their importance
Finite-Function-Encoding Quantum States
We investigate the encoding of higher-dimensional logic into quantum states.
To that end we introduce finite-function-encoding (FFE) states which encode
arbitrary -valued logic functions and investigate their structure as an
algebra over the ring of integers modulo . We point out that the
polynomiality of the function is the deciding property for associating
hypergraphs to states. Given a polynomial, we map it to a tensor-edge
hypergraph, where each edge of the hypergraph is associated with a tensor. We
observe how these states generalize the previously defined qudit hypergraph
states, especially through the study of a group of finite-function-encoding
Pauli stabilizers. Finally, we investigate the structure of FFE states under
local unitary operations, with a focus on the bipartite scenario and its
connections to the theory of complex Hadamard matrices.Comment: Comments welcom
