    Intra-Laboratory Calibration Exercise for Quantification of Microplastic Particles in Fine-Grained Sediment Samples: Special Focus on the Influence of User Experience

    An intra-laboratory calibration to quantify microplastic in fine-grained marine sediments was performed with two objectives: (a) to determine the recovery rate of self-produced microplastics characterized by a size ranging from 220 µm to 5 mm and differing in color (pink, orange, gray, yellow, silver), shape (fragments, filaments, spheres, films), and chemical composition (polystyrene, polyethylene, polyvinyl chloride, acrylonitrile-butadiene-styrene, polypropylene, poly(methyl methacrylate)) artificially introduced into real samples; and (b) to analyze whether operator experience can be a key factor in the quality of the results. To answer this question, the same protocol was assigned to an experienced and an inexperienced operator. The results of this comparison are detailed in terms of root mean square and percent error.     Litter in alien species of possible commercial interest: The blue crab (Callinectes sapidus Rathbun, 1896) as case study

    Marine litter levels were measured in the stomach contents, hepatopancreas, and gonad tissues of crustacea decapod (Callinectes sapidus Rathbun, 1896; n\ua0=\ua06), a widespread alien species affecting the Lesina lagoon. Results highlight a mean of 2.5 (SD\ua0=\ua01.6) items/animal and indicate the presence of metals fragments (13%) and plastics (13% PE; 6.7% PET) in the stomach contents of collected individuals. No microplastic particles were detected in the hepatopancreas or in muscle tissue, while microplastic fibres (nylon, rayon, polyester) were found present in female specimen gonads. The presence of synthetic fibres in the investigated species reflects the relative contamination level in this habitat type and suggests that the blue crab could be considered a model organism for evaluating the contamination status of the study area

    Potential Use of Tea Tree Oil as a Disinfectant Agent against Coronaviruses: A Combined Experimental and Simulation Study

    : The COVID-19 pandemic has highlighted the relevance of proper disinfection procedures and renewed interest in developing novel disinfectant materials as a preventive strategy to limit SARS-CoV-2 contamination. Given its widely known antibacterial, antifungal, and antiviral properties, Melaleuca alternifolia essential oil, also named Tea tree oil (TTO), is recognized as a potential effective and safe natural disinfectant agent. In particular, the proposed antiviral activity of TTO involves the inhibition of viral entry and fusion, interfering with the structural dynamics of the membrane and with the protein envelope components. In this study, for the first time, we demonstrated the virucidal effects of TTO against the feline coronavirus (FCoVII) and the human coronavirus OC43 (HCoV-OC43), both used as surrogate models for SARS-CoV-2. Then, to atomistically uncover the possible effects exerted by TTO compounds on the outer surface of the SARS-CoV-2 virion, we performed Gaussian accelerated Molecular Dynamics simulations of a SARS-CoV-2 envelope portion, including a complete model of the Spike glycoprotein in the absence or presence of the three main TTO compounds (terpinen-4-ol, Îł-terpinene, and 1,8-cineole).     Transcription factors operate in developmental processes to mediate inductive events and cell competence, and perturbation of their function or regulation can dramatically affect morphogenesis, organogenesis, and growth. We report that a narrow spectrum of amino-acid substitutions within the transactivation domain of the v-maf avian musculoaponeurotic fibrosarcoma oncogene homolog (MAF), a leucine zipper-containing transcription factor of the AP1 superfamily, profoundly affect development. Seven different de novo missense mutations involving conserved residues of the four GSK3 phosphorylation motifs were identified in eight unrelated individuals. The distinctive clinical phenotype, for which we propose the eponym Aymé-Gripp syndrome, is not limited to lens and eye defects as previously reported for MAF/Maf loss of function but includes sensorineural deafness, intellectual disability, seizures, brachycephaly, distinctive flat facial appearance, skeletal anomalies, mammary gland hypoplasia, and reduced growth. Disease-causing mutations were demonstrated to impair proper MAF phosphorylation, ubiquitination and proteasomal degradation, perturbed gene expression in primary skin fibroblasts, and induced neurodevelopmental defects in an in vivo model. Our findings nosologically and clinically delineate a previously poorly understood recognizable multisystem disorder, provide evidence for MAF governing a wider range of developmental programs than previously appreciated, and describe a novel instance of protein dosage effect severely perturbing developmen

    Microplastics Occurrence in the European Common Frog (Rana temporaria) from Cottian Alps (Northwest Italy)

    Microplastics (MPs) pollution is arousing growing attention, yet knowledge about its occurrence in amphibians is scant to date. With this study, we aimed to determine whether plastic (>5000 μm) and MPs (10–5000 μm) could be detected in adult Rana temporaria from a high-mountain ecosystem (the Cottian Alps, northwest Italy). To do this, aquatic compartments and the digestive tract of adult R. temporaria were analyzed. Water, sediment, periphyton, aquatic macroinvertebrates, and tadpoles tested negative for plastic and MPs. Microplastics were detected in all the adult frogs (n = 5); all the identified items (one per specimen) were fibers (size range: 550.91–2355.51 µm). A statistically significant positive correlation between the particle length and frog size was recorded. The predominant fiber color was blue. The chemical composition was polyamide (60%), polyethylene (20%), and polyethylene terephthalate (20%). Since both the biotic and the abiotic freshwater compartments (tadpoles included) revealed the absence of MPs, it can be assumed that adult frogs ingest MPs from the surrounding terrestrial environment.This research was partially funded by Fondazione CRT, the ALPLA II project, grant number 21D03.Peer reviewe

The general thought associates the use of glitter with extraordinary and rare events, in reality, its presence in everyday life is much more ubiquitous than one might think. The main objective of this preliminary study was to create a mini product survey with the aim of analyzing the distribution and composition of glitter in the local market of the city of Trieste (Italy), thus creating a starting point for larger future studies. The analysis of a real context served as a snapshot for the description of the phenomenon, from which useful information was obtained: glitter is everywhere, cheap, disperses easily in the environment, is made of different materials, often has sharp-edged shapes, and mainly affects sensitive audiences, such as children. It is time to put glitter in the spotlight

    A Multidisciplinary Approach to Evaluate the Effects of Contaminants of Emerging Concern on Natural Freshwater and Brackish Water Phytoplankton Communities

    7noEcotoxicological assays on monospecific phytoplankton have limited application for detecting the effects of environmental pollutants on multiple species communities. With this study, we took an ecotoxicological, ecological, and biochemical approach to evaluate the effects of two contaminants of emerging concern (zinc oxide nanoparticles, ZnO NPs, and potassium dichromate, K2Cr2O7) at different concentrations (K2Cr2O7 5.6–18–50 mg/L; ZnO NPs 10–100–300 mg/L) on natural freshwater and brackish water phytoplankton communities. Cell density and absorbance values decreased in freshwater and brackish water phytoplankton communities after exposure to ZnO NPs (100 mg/L and 300 mg/L only for freshwater), whereas growth rate was increased in both freshwater and brackish water phytoplankton communities after exposure to ZnO NPs 10 mg/L. Differently, there was no clear relationship between concentration and inhibition growth after exposure to K2Cr2O7: the lowest cell density was recorded after exposure to 18 mg/L. Moreover, the evenness index value was lower compared to the other concentrations, indicating the growth of a few, albeit resistant species to higher K2Cr2O7 concentrations. Generally, Bacillariophyceae and Dinoficee were prevalent in phytoplankton cultures after exposure to ZnO NPs and K2Cr2O7. The Shannon-Wiener index was slightly higher in the negative than the positive controls, but diversity was low after all treatments in both ecotoxicological assays. The evenness index was always very close to zero, indicating the numerical predominance of one or very few species. Finally, the decrease in chlorophyll-a and pheophytin-a in both ecotoxicological assays indicated a change in photosynthetic activity.
