    The goal of this thesis is to elucidate the assembly mechanism of macromolecular complexes in the microtubule-based axoneme in flagella. The axoneme is comprised of more than 400 distinct polypeptides. While all axonemal proteins are synthesized in the cell body, they need to be imported to the tip of the growing flagella for the final assembly. They are then assembled into distinctive structural complexes and integrated into precise positions relative to each other. The assembled macromolecular complexes operate in concert enabling the axoneme to beat rhythmically as a biological machine. This arduous process involves multiple reactions and meticulous regulation. However, the assembly process in the cell body and at the tip of flagella remains largely unknown. This thesis took advantage of a flagella model organism Chlamydomonas and a rather simple and a well-characterized axonemal complex, the radial spoke, to identify the key assembly mechanisms. Genetic studies have revealed two spoke components, RSP3 and LC8, that are particularly important for the assembly of the entire complex. Mutants defective in either gene generate paralyzed flagella devoid of the RS. Using a variety of approaches that are possible for this model organism, this study revealed that a stack of LC8 dimers directly binds to RSP3 at the tip of the flagella to promote a series of events, including phosphorylation of RSP3, formation of the base of the RS and docking of the spoke to the axoneme. These findings have shed light on the series of events that occur between transport and assembly. These findings also have broad implications, as LC8 is a promiscuous molecule that interacts with many flagellar and non-flagellar proteins in the cell body. Many of them play vital roles in the normal functioning of cells or in the pathogenesis of microorganisms. Some LC8 target proteins exist in the molecular complexes, as the radial spoke. However, the LC8-involved reactions remain poorly defined. The findings from this thesis show that LC8 bindings can trigger pleiotropic effects on a single target protein.     Effect of Trifluoromethyl Substitution on C-3 Position in 1H NMR of Quinolones / Coumarins

    The structural assignment of differently substituted quinolones / coumarins was reviewed using 1H NMR spectral data. In case of quinolones / coumarins, with varied substitutions at C-4 and different positions of the benzenoid ring, there is a great variation in the 1H NMR value of the proton only attached with C-3. It was observed that when different substitutions (methyl / amino / hydroxy / acetoxy / trifluoromethyl) were carried out at C-4 position, maximum deshielding with the proton attached with C-3 was observed with trifluoromethyl substitution. The observation is same even with the variation in different groups at the benzenoid ring.Â

    A case of perforation of rectum due to self-administered enema in a pregnant woman

    A 23 year old primigravida at 35 weeks gestation presented with pain abdomen, fever and dissension of abdomen. Initially patient was managed conservatively at peripheral centre for two days and then reported to our hospital. An ultrasound done which shows collection inside peritoneal cavity and perforation was suspected. Decision of laparotomy was done. First caesarean section was done and a single live male baby born weighing 2.4 kg born. Then on exploration a small perforation in rectum was found and it was repaired by surgeon. But they found two more perforations and colostomy was done. On taking detailed history patient told that she-herself administered enema because of constipation. In post op period patient expired because of septicemia and ARDS

    Supervariable approach to particle on a torus knot: A model for Hodge theory

    We analyze a particle constrained to move on a (p,q)(p,q)-torus knot within the framework of supervariable approach and deduce the BRST as well as anti-BRST symmetries. We also capture the nilpotency and absolute anti-commutativity of (anti-)BRST symmetries in this framework. Further, we show the existence of some novel symmetries in the system such as (anti-)co-BRST, bosonic, and ghost scale symmetries. We demonstrate that the conserved charges (corresponding to these symmetries) adhere to an algebra which is analogous to that of de Rham cohomological operators of differential geometry. As the charges (and the symmetries) find a physical realization with the differential geometrical operators, at the algebraic level, the present model presents a prototype for Hodge theory.Comment: 15 pages, no figure
