Workplace Values: Cross-Cultural Insights from the Service Industries
With the globalization of businesses in recent years, managers must attract and retain the right employees. Part of the fit between a worker and employer is in the area of values. Employees bring personal values to the job and they also are asked to display the workplace values of the organization. Managers must be sure that all workers share the values of the organization. Few studies examine workplace values cross-culturally and qualitatively. Using existing theory, we find that service workers in three countries display workplace values that reflect cultural norms. Managerial implications and directions for future research are given
Leadership and Culture in Ghanaâs Tourism and Hospitality Industry: The Impact of Transformational Leadership on Job Satisfaction in an Emerging Economy
The purpose of this study is to assess the relationship between leadership style, organizational culture, and employee job satisfaction. We specifically examine business leaders in Ghana to explain some of the regionâs characteristics regarding its emerging economy and business environment. This study seeks to illustrate a significant relationship between organizational culture and organizational performance within the tourism and hospitality industry in Ghana. Managing job satisfaction within an organization is essential to firm performance. Maintaining high employee satisfaction is also critical to retaining skilled employees and keeping the turnover rate at a minimum. One of the ways that are effective in helping managers increase satisfaction among their followers is employing the right leadership style and culture. We surveyed 102 small business owners and managers in Ghanaâs tourism and hospitality industry. After examining the data collected from the survey instruments, we have a better understanding of whether there is a noteworthy relationship between leadership style and culture as it relates to job satisfaction in the business environments of West African small businesses
The Effect of Soluble Uterine Factors on Porcine Embryo Development Within a Three-Dimensional Alginate Matrix System
Between day 10 and 12 of gestation in the pig, the embryo undergoes a dramatic morphological change, known as elongation. During elongation the embryo produces and secretes estrogen, which serves as a key signal for maternal recognition of pregnancy. The uterine environment prepares for embryo elongation and implantation by releasing nutrients, metabolites, and hormones in a time-dependent manner relative to paracrine signals from the conceptus (i.e., estrogen) and endocrine/paracrine signals within the endometrium (i.e., progesterone). Despite advances in identification of specific uterine factors present throughout pregnancy in the pig, little is known about the exact mechanisms by which porcine embryos elongate normally, or how this elongation is altered during embryonic loss. Previously, our laboratory has established an in vitro culture system using alginate hydrogels as a three-dimensional (3-D) matrix to support the development of pre-implantation porcine embryos in base serum media without the addition of hormones. The objective of the current study was to improve our 3-D system and determine if uterine flushes from day 9 or 10 of gestation were capable of supporting in vitro embryonic development of in vivo-developed day 9 porcine embryos using the 3- D alginate culture system. Embryos collected on day 9 of gestation were assigned to be cultured in pooled day 9 flushes, day 10 flushes, or base serum media for five days within 0.7% alginate gels. Our results demonstrate that uterine flushes from day 9 or 10 of gestation supported embryonic development in our 3-D hydrogel system, similar to that from embryos cultured in base serum media. These results illustrate that soluble factors present in uterine flushings at day 9 or 10 of gestation support in vitro embryonic development within our 3-D alginate culture system. Developing better scientific understanding of the factors that regulate embryo elongation and early embryonic mortality can be useful for developing strategies to improve sow productivity and increase the profitability of swine production
Secreted metabolome of porcine blastocysts encapsulated with in \u3ci\u3ein vitro\u3c/i\u3e 3D alginate hydrogel culture systems under going morphological changes provides insights into specific mechanisms involved in the initiation of porcine conceptus elongation
Context. The exact mechanisms regulating the initiation of porcine conceptus elongation are not known due to the complexity of the uterine environment. Aims. To identify contributing factors for initiation of conceptus elongation in vitro, this study evaluated differential metabolite abundance within media following culture of blastocysts within unmodified alginate (ALG) or Arg-Gly-Asp (RGD)-modified alginate hydrogel culture systems. Methods. Blastocysts were harvested from pregnant gilts, encapsulated within ALG or RGD or as non-encapsulated control blastocysts (CONT), and cultured. At the termination of 96 h culture, media were separated into blastocyst media groups: non-encapsulated control blastocysts (CONT); ALG and RGD blastocysts with no morphological change (ALGâ and RGDâ); ALG and RGD blastocysts with morphological changes (ALG+ and RGD+) and evaluated for non-targeted metabolomic profiling by liquid chromatography (LC)âmass spectrometry (MS) techniques and gas chromatographyâ (GCâMS). Key results. Analysis of variance identified 280 (LCâMS) and 1 (GCâMS) compounds that differed (P \u3c 0.05), of which 134 (LCâMS) and 1 (GCâMS) were annotated. Metabolites abundance between ALG+ vs ALGâ, RGD+ vs RGDâ, and RGD+ vs ALG+ were further investigated to identify potential differences in metabolic processes during the initiation of elongation. Conclusions. This study identified changes in phospholipid, glycosphingolipid, lipid signalling, and amino acid metabolic processes as potential RGD-independent mechanisms of elongation and identified changes in lysophosphatidylcholine and sphingolipid secretions during RGD-mediated elongation. Implications. These results illustrate changes in phospholipid and sphingolipid metabolic processes and secretions may act as mediators of the RGD-integrin adhesion that promotes porcine conceptus elongation
Metabolic compounds within the porcine uterine environment are unique to the type of conceptus present during the early stages of blastocyst elongation
The objective of this study was to identify metabolites within the porcine uterine milieu during the early stages of blastocyst elongation. At Days 9, 10, or 11 of gestation, reproductive tracts of White crossâbred gilts (n = 38) were collected immediately following harvest and flushed with Roswell Park Memorial Instituteâ1640 medium. Conceptus morphologies were assessed from each pregnancy and corresponding uterine flushings were assigned to one of five treatment groups based on these morphologies: (a) uniform spherical (n = 8); (b) heterogeneous spherical and ovoid (n = 8); (c) uniform ovoid (n = 8); (d) heterogeneous ovoid and tubular (n = 8); and (e) uniform tubular (n = 6). Uterine flushings from these pregnancies were submitted for nontargeted profiling by gas chromatographyâmass spectrometry (GCâMS) and ultra performance liquid chromatography (UPLC)âMS techniques. Unsupervised multivariate principal component analysis (PCA) was performed using pcaMethods and univariate analysis of variance was performed in R with false discovery rate (FDR) adjustment. PCA analysis of the GCâMS and UPLCâMS data identified 153 and 104 metabolites, respectively. After FDR adjustment of the GCâMS and UPLCâMS data, 38 and 59 metabolites, respectively, differed (p \u3c .05) in uterine flushings from pregnancies across the five conceptus stages. Some metabolites were greater (p \u3c .05) in abundance for uterine flushings containing earlier stage conceptuses (i.e., spherical), such as uric acid, tryptophan, and tyrosine. Global analysis of differential gene expression within the porcine conceptus transcriptome as it transitions through spherical, ovoid, and tubular morphologies during the initiation of elongation
This study aimed to identify transcriptome differences between distinct or transitional stage spherical, ovoid, and tubular porcine blastocysts throughout the initiation of elongation. We performed a global transcriptome analysis of differential gene expression using RNAâSeq with high temporal resolution between spherical, ovoid, and tubular stage blastocysts at specific sequential stages of development from litters containing conceptus populations of distinct or transitional blastocysts. After RNAâSeq analysis, significant differentially expressed genes (DEGs) and pathways were identified between distinct morphologies or sequential development stages. Overall, 1898 significant DEGs were identified between distinct spherical and ovoid morphologies, with 311 total DEGs between developmental stages throughout this first morphological transition, while 15 were identified between distinct ovoid and tubular, with eight total throughout these second morphological transition developmental stages. The high quantity of DEGs and pathways between conceptus stages throughout the spherical to ovoid transition suggests the importance of gene regulation during this first morphological transition for initiating elongation. Further, extensive DEG coverage of known elongation signaling pathways was illustrated from spherical to ovoid, and regulation of lipid signaling and membrane/ECM remodeling across these early conceptus stages were implicated as essential to this process, providing novel insights into potential mechanisms governing this rapid morphological change
Roles of key active-site residues in flavocytochrome P450 BM3
Abbreviations used: P450, cytochrome P450 mono-oxygenase; ImC12, 12-(imidazolyl)dodecanoic acid; 1-PIM, 1-phenylimidazole.The effects of mutation of key active-site residues (Arg-47, Tyr-51, Phe-42 and Phe-87) in Bacillus megaterium flavocytochrome P450 BM3 were investigated. Kinetic studies on the oxidation of laurate and arachidonate showed that the side chain of Arg-47 contributes more significantly to stabilization of the fatty acid carboxylate than does that of Tyr-51 (kinetic parameters for oxidation of laurate: R47A mutant, Km 859 ”M, kcat 3960 min-1; Y51F mutant, Km 432 ”M, kcat 6140 min-1; wild-type, Km 288 ”M, kcat 5140 min-1). A slightly increased kcat for the Y51F-catalysed oxidation of laurate is probably due to decreased activation energy (DGâĄ) resulting from a smaller DG of substrate binding. The side chain of Phe-42 acts as a phenyl 'cap' over the mouth of the substrate-binding channel. With mutant F42A, Km is massively increased and kcat is decreased for oxidation of both laurate (Km 2.08 mM, kcat 2450 min-1) and arachidonate (Km 34.9 ”M, kcat 14620 min-1; compared with values of 4.7 ”M and 17100 min-1 respectively for wild-type). Amino acid Phe-87 is critical for efficient catalysis. Mutants F87G and F87Y not only exhibit increased Km and decreased kcat values for fatty acid oxidation, but also undergo an irreversible conversion process from a 'fast' to a 'slow' rate of substrate turnover [for F87G (F87Y)-catalysed laurate oxidation: kcat 'fast', 760 (1620) min-1; kcat 'slow', 48.0 (44.6) min-1; kconv (rate of conversion from fast to slow form), 4.9 (23.8) min-1]. All mutants showed less than 10% uncoupling of NADPH oxidation from fatty acid oxidation. The rate of FMN-to-haem electron transfer was shown to become rate-limiting in all mutants analysed. For wild-type P450 BM3, the rate of FMN-to-haem electron transfer (8340 min-1) is twice the steady-state rate of oxidation (4100 min-1), indicating that other steps contribute to rate limitation. Active-site structures of the mutants were probed with the inhibitors 12-(imidazolyl)dodecanoic acid and 1-phenylimidazole. Mutant F87G binds 1-phenylimidazole > 10-fold more tightly than does the wild-type, whereas mutant Y51F binds the haem-co-ordinating fatty acid analogue 12-(imidazolyl)dodecanoic acid > 30-fold more tightly than wild-type
Leptin: A Metabolic Signal for the Differentiation of Pituitary Cells
Pituitary cell function is impacted by metabolic states and therefore must receive signals that inform them about nutritional status or adiposity. A primary signal from adipocytes is leptin, which recent studies have shown regulates most pituitary cell types. Subsets of all pituitary cell types express leptin receptors and leptin has been shown to exert transcriptional control through classical JAK/STAT pathways. Recent studies show that leptin also signals through post-transcriptional pathways that involve the translational regulatory protein Musashi. Mechanistically, post-transcriptional control would permit rapid cellular regulation of critical pre-existing pituitary transcripts as energy states change. The chapter will review evidence for transcriptional and/or post-transcriptional regulation of leptin targets (including Gnrhr, activin, Fshb, Gh, Ghrhr, and Pou11f1) and the consequences of the loss of leptin signaling to gonadotrope and somatotrope functions
Prevalence and correlates of frailty in an older rural African population:findings from the HAALSI cohort study
Background:Â Frailty is a key predictor of death and dependency, yet little is known about frailty in sub-Saharan Africa despite rapid population ageing. We describe the prevalence and correlates of phenotypic frailty using data from the Health and Aging in Africa: Longitudinal Studies of an INDEPTH Community cohort. Methods:Â We analysed data from rural South Africans aged 40 and over. We used low grip strength, slow gait speed, low body mass index, and combinations of self-reported exhaustion, decline in health, low physical activity and high self-reported sedentariness to derive nine variants of a phenotypic frailty score. Each frailty category was compared with self-reported health, subjective wellbeing, impairment in activities of daily living and the presence of multimorbidity. Cox regression analyses were used to compare subsequent all-cause mortality for non-frail (score 0), pre-frail (score 1â2) and frail participants (score 3+). Results:Â Five thousand fifty nine individuals (mean age 61.7Â years, 2714 female) were included in the analyses. The nine frailty score variants yielded a range of frailty prevalences (5.4% to 13.2%). For all variants, rates were higher in women than in men, and rose steeply with age. Frailty was associated with worse subjective wellbeing, and worse self-reported health. Both prefrailty and frailty were associated with a higher risk of death during a mean 17Â month follow up for all score variants (hazard ratios 1.29 to 2.41 for pre-frail vs non-frail; hazard ratios 2.65 to 8.91 for frail vs non-frail). Conclusions:Â Phenotypic frailty could be measured in this older South African population, and was associated with worse health, wellbeing and earlier death
Enhancing Consumer Attitude Toward Culturally Mixed Symbolic Products from Foreign Global Brands in an Emerging-Market Setting: The Role of Cultural Respect
The extant literature has not examined the conditions that govern integrative and exclusionary reactions to cultural hybrid products with sufficient detail. Within an emerging-market setting, this study explores how culturally mixed symbolic products (CMSPs) from foreign global brands can avoid antagonistic consumer attitudes. Building on social categorization theory, the authors argue that foreign global brands are viewed as belonging to an out-group and may thus encounter difficulties in tapping local cultural capital, resulting in a negative relationship between brand globalness and consumer attitude toward CMSPs. However, they contend that product category moderates this relationship such that there is a stronger negative effect for nonfood products than for food products. Moreover, the authors theorize that (1) cultural respect by foreign global companies directly enhances consumer attitudes toward CMSPs and (2) cultural respect attenuates the negative brand globalnessâCMSP attitude link. These hypotheses are tested using a representative consumer sample from eight provinces/municipalities in China (n = 646). Results provide important implications for global companies on how to benefit from local cultural resources in their localization processes
