    Evidence of Attenuation of VHE Blazar Spectra by Extragalactic Background Light

    The spectrum of two blazar objects, 1ES 1959+650 and 1ES 2344+514, are analyzed for evidence of interactions with the Extragalactic Background Light (EBL), using combined data from the Very Energetic Radiation Imaging Telescope Array System and the Fermi Gamma-ray Space Telescope, By analyzing the distinct curvature across the combined spectrum, we infer that the very-high energy (VHE) gamma-rays must be attenuated by interactions with the EBL. We also find that the measured 1ES 1959+650 spectrum is sensitive to the intrinsic blazar model, with a preference for a power-law with an exponential cutoff (EPWL). The measured curvature is a combination of the EBL attenuation and the exponential cut-off, as determined by statistical tests assuming that there is no EBL. Finally, a test of the optical depth due to the EBL indicates that the two blazars are consistent with the amount of EBL given by the Franceschini model of intergalactic optical depth

    Visual Responses in Mice Lacking Critical Components of All Known Retinal Phototransduction Cascades

    The mammalian visual system relies upon light detection by outer-retinal rod/cone photoreceptors and melanopsin-expressing retinal ganglion cells. Gnat1(-/-); Cnga3(-/-); Opn4(-/-) mice lack critical elements of each of these photoreceptive mechanisms via targeted disruption of genes encoding rod alpha transducin (Gnat1); the cone-specific alpha 3 cyclic nucleotide gated channel subunit (Cnga3); and melanopsin (Opn4). Although assumed blind, we show here that these mice retain sufficiently widespread retinal photoreception to drive a reproducible flash electroretinogram (ERG). The threshold sensitivity of this ERG is similar to that of cone-based responses, however it is lost under light adapted conditions. Its spectral efficiency is consistent with that of rod opsin, but not cone opsins or melanopsin, indicating that it originates with light absorption by the rod pigment. The TKO light response survives intravitreal injection of U73122 (a phospholipase C antagonist), but is inhibited by a missense mutation of cone alpha transducin (Gnat2(cpfl3)), suggesting Gnat2-dependence. Visual responses in TKO mice extend beyond the retina to encompass the lateral margins of the lateral geniculate nucleus and components of the visual cortex. Our data thus suggest that a Gnat1-independent phototransduction mechanism downstream of rod opsin can support relatively widespread responses in the mammalian visual system. This anomalous rod opsin-based vision should be considered in experiments relying upon Gnat1 knockout to silence rod phototransduction

    Exploration for volcanogenic sulphide mineralisation at Benglog, north Wales

    Exploration for volcanogenic sulphide mineralisation around Benglog is one of three investigations designed to assess the metallogenic potential of the Ordovician Aran Volcanic Group. Detailed geological mapping in the Benglog area enabled an interpretation of the volcanic environment, critical to such an assessment, to be made. The eruptive rocks are acid and basic in composition; the acid rocks are mostly ash-flow tuffs derived from outside the area, whereas the basic rocks have a local derivation. They are all interbedded with dark grey or black silty mudstone and were probably erupted in a submarine environment. Contemporaneous dolerite sills were intruded into wet sediment. This environment was suitable for volcanogenic exhalative sulphide deposits to form and indications of a metallogenic horizon were found at the top of the Y Fron Formation in the form of abundant pyrite, minor pyrrhotite and minor base metal enrichment. Soil samples, analysed for copper, lead and zinc, were collected and geophysical surveys were carried out along eleven east-west trending traverse lines 300 m apart across the volcanic succession. Indications were found of minor vein mineralisation at dolerite intrusion margins and locally along faults. Very high chargeability and low resistivity anomalies over mudstones did not spatially coincide with geochemical anomalies in soil, but the secondary redistribution of metals in soils and variable thickness of overburden precluded confident interpretation of the source of many soil anomalies. Geochemical drainage data, in conjunction with rock analyses, show strong barium enrichment in mudstones which could be volcanogenic in origin but related to two separate eruptive episodes. The findings of the survey were inconclusive. An environment suitable for the formation of volcanogenic exhalative sulphide deposits was established, but the geochemical and geophysical surveys located only minor vein mineralisation and tenuous indications of other styles of mineralisation. Recommendations are made for further work

    Niclosamide enhances abiraterone treatment via inhibition of androgen receptor variants in castration resistant prostate cancer.

    Considerable evidence from both clinical and experimental studies suggests that androgen receptor variants, particularly androgen receptor variant 7 (AR-V7), are critical in the induction of resistance to enzalutamide and abiraterone. In this study, we investigated the role of AR-V7 in the cross-resistance of enzalutamide and abiraterone and examined if inhibition of AR-V7 can improve abiraterone treatment response. We found that enzalutamide-resistant cells are cross-resistant to abiraterone, and that AR-V7 confers resistance to abiraterone. Knock down of AR-V7 by siRNA in abiraterone resistant CWR22Rv1 and C4-2B MDVR cells restored their sensitivity to abiraterone, indicating that AR-V7 is involved in abiraterone resistance. Abiraterone resistant prostate cancer cells generated by chronic treatment with abiraterone showed enhanced AR-V7 protein expression. Niclosamide, an FDA-approved antihelminthic drug that has been previously identified as a potent inhibitor of AR-V7, re-sensitizes resistant cells to abiraterone treatment in vitro and in vivo. In summary, this preclinical study suggests that overexpression of AR-V7 contributes to resistance to abiraterone, and supports the development of combination of abiraterone with niclosamide as a potential treatment for advanced castration resistant prostate cancer

    The azimuthal component of Poynting's vector and the angular momentum of light

    The usual description in basic electromagnetic theory of the linear and angular momenta of light is centred upon the identification of Poynting's vector as the linear momentum density and its cross product with position, or azimuthal component, as the angular momentum density. This seemingly reasonable approach brings with it peculiarities, however, in particular with regards to the separation of angular momentum into orbital and spin contributions, which has sometimes been regarded as contrived. In the present paper, we observe that densities are not unique, which leads us to ask whether the usual description is, in fact, the most natural choice. To answer this, we adopt a fundamental rather than heuristic approach by first identifying appropriate symmetries of Maxwell's equations and subsequently applying Noether's theorem to obtain associated conservation laws. We do not arrive at the usual description. Rather, an equally acceptable one in which the relationship between linear and angular momenta is nevertheless more subtle and in which orbital and spin contributions emerge separately and with transparent forms
