    Effect of melatonin on male offspring testis and sperm parameters in BALB/c mice after exposing their mother to METHamphetamine during pregnancy and lactation

    Objective(s): Methamphetamine (METH) is a psychostimulant that has harmful effects on all organs,  the nervous system, cardiovascular system, and reproductive system. Since many METH consumers are young people of reproductive age, it poses a risk to the next generation of METH consumers. METH can pass through the placenta and is also secreted into breast milk. Melatonin (MLT) is the primary hormone of the pineal gland that regulates the circadian cycle, and it is also an antioxidant that can mitigate the effects of toxic substances. This study aims to investigate the protective effect of melatonin against the detrimental effects that METH has on the reproductive system of male newborns, whose mothers consumed METH during pregnancy and lactation.Materials and Methods: In the current study, 30 female adult balb/c mice were divided into three groups: control group, vehicle group that received normal saline, and the experimental group that received 5 mg/kg METH intraperitoneally during gestation and lactation. After lactation, the male offspring of each group were randomly divided into two subgroups, one of which received 10 mg/kg melatonin intragastrically for 21 days (corresponding to the lactation period of the mice) (METH-MLT) and the other did not (METH -D.W). After treatment, the mice were sacrificed and testicular tissue and epididymis were obtained for the following tests.Results: The diameter of seminiferous tubules, SOD activity, total Thiol groups concentration, catalase activity, sperm count, and PCNA and CCND gene expression were significantly increased in the METH-MLT group compared with the METH-DW. Apoptotic cells and MDA level ameliorated in the METH-MLT group compared with METH-D.W, and testicular weight had no notable change.Conclusion: The current study represents that consumption of METH during pregnancy and lactation can have adverse effects on the histological and biochemical factors of testis and sperm parameters of male newborns,  which can be mitigated by taking melatonin after the end of the breastfeeding period

    Ameliorative effects of crocin against electromagnetic field-induced oxidative stress and liver and kidney injuries in mice

    Objective: The current study's goal was to examine how crocin affects organ damage such as damage to the kidney and liver in mice treated by 2100 MHz Electro Magnetic Field.Materials and Methods: The liver and kidneys of mice exposed to EMFs were used in this study to examine how crocin affected them. 24 male NMARI mice were randomly divided into 4 groups: EMF group (2100 MHZ); Crocin (Cr) group (50 mg/kg); EMF+Crocin group (2100 MHZ+50 mg/kg), and control group. The antioxidant enzymes and some serum biochemical parameters were assessed in blood samples collected after the experiment. After the animals were put to sleep, liver and kidney samples were taken for histopathological and liver samples were taken for ultrastructural analysis.Results: The serum levels of urea and creatinine, and serum activities of alanine transaminase, aspartate transaminase, and alkaline phosphatase were higher in the EMF group than the control group, and this difference was significant. When compared to the control group, the EMF group's antioxidants, (catalase and superoxide dismutase) activity were decreased. These metrics significantly improved in the EMF + Cr group when compared to the EMF group. Different pathological damages were present in the liver and kidney of the EMF group, and the liver's ultrastructure had changed. Crocin administration decrease these changes.Conclusion: Crocin, an antioxidant agent, may provide defense against tissue damage brought on by EMF by reducing oxidative stress

    In vivo effects of allogeneic mesenchymal stem cells in a rat model of acute ischemic kidney injury

    Objective(s): Renal ischemia-reperfusion injury (IRI) as a severe condition of acute kidney injury (AKI) is the most common clinical problem with high mortality rates of 35-60% deaths in hospital. Mesenchymal stem cells (MSC) due to unique regenerative characteristics are ideal candidates for the treatment of the ischemic injuries. This work is focused on the administration of MSC to IRI-induced AKI Wistar rats and evaluating their significance in AKI treatment. Material and Methods: Animals underwent surgical procedure and AKI was induced by 40 min bilateral renal pedicle clamping. Immediately after reperfusion, 2×106 rat bone marrow derived MSCs were injected via intra-parenchymal or intra-aortic route. Results: Animals subjected to AKI after days 1 and 3 showed significant increase in the serum creatinine and blood urea nitrogen (BUN) concentration along with a declined glomerular filtration rate (GFR) when compared with non-ischemic animals. On the other hand, treated animals showed a significant enhanced regeneration as compared to ischemic animals in both administration route groups. Conclusion: According to the results concluded from the renoprotective effects of MSC in IRI/AKI, MSCs could be considered as promising therapeutic approach for AKI in clinical applications

    Objective(s):Silver nanoparticles (Ag-NPs) are used widely in bedding, water purification, tooth paste and toys. These nanoparticles can enter into the body and move into the hippocampus. The aim of this study was to investigate the neurotoxicity of silver nanoparticles in the adult rat hippocampus. Materials and Methods:12 male Wistar rats were randomly divided into two experimental and control groups (6 rats in each group). Animals in the experimental group received Ag-NPs (30 mg/kg) orally (gavage) for 28 consecutive days. Control group in the same period was treated with distilled water via gavage. At the end of experiment, animals were deeply anesthetized, sacrificed, and their brains were collected from each group. Finally the brain sections were stained using toluidine blue and TUNEL. Then to compare the groups, dark neurons (DNs) and apoptotic neurons were counted by morphometric method. Results: Results showed that the num­bers of DNs and apoptotic cells in the CA1, CA2, CA3, and dentate gyrus (DG) of hippocampus significantly increased in the Ag-NPs group in comparison to the control group (

    Stanozolol is an anabolic-androgenic steroid which is commonly abused by athletes for improved energy, appearance, and physical size. It has been previously shown to cause changes in behaviour and has various physical effects. Studies have previously been conducted on its neurotoxic effect on the central nervous system (CNS), which are typically psychological in nature. This study was performed to investigate the apoptotic effect of stanozolol on different parts of the rat hippocampus. Sixteen male Wistar rats were divided randomly into two groups (experimental and control). The experimental group received subcutaneous injections of stanozolol (5mg/kg/day) for consecutive 28 days, whereas the control group received saline using the same dosing schedule and administration route. After routine procedures, coronal sections of rat brain were stained with Toluidine blue and TUNEL for pre-apoptotic and apoptotic cell detection, respectively. In order to compare groups, the mean number of TUNEL-positive and pre-apoptotic neurons per unit area were calculated and analysed. Histopathological examination revealed that the mean number of pre-apoptotic and apoptotic neurons in the CA1, CA2, CA3 and DG areas of the hippocampus were significantly increased in the stanozolol treated group. In conclusion, stanozolol abuse may induce pre-apoptotic and apoptotic cell formation in different regions of the hippocampus

    Anticonvulsant and neuroprotective effects of Rosa damascena hydro-alcoholic extract on rat hippocampus

    Objective: Previously, analgesic, hypnotic, and anticonvulsant effects have been suggested for Rosa damascena (R. damascena). In the present study, possible anti-seizure and neuro-protective effects of hydro-alcoholic extract of R. damascena has been investigated after inducing seizures in rats by pentylenetetrazole (PTZ). Materials and Methods: The rats were divided to five groups: (1) Control: received saline, (2) PTZ: 100 mg/kg, i.p., (3) PTZ-Extract 50 mg/kg(PTZ-Ext 50), (4) PTZ- Extract 100 mg/kg(PTZ-Ext 100), and (5) PTZ- Extract 200 mg/kg(PTZ-Ext 200) groups which were treated with 50, 100, and 200 mg/kg respectively of hydro-alcoholic extract of R. damascena for one week before PTZ injection. The animals were examined for electrocorticography (ECoG) recording and finally, the brains were removed for histological study. Results: The hydro-alcoholic extract of R. damascena significantly prolonged the latency of seizure attacks and reduced the frequency and amplitude of epileptiform burst discharges induced by PTZ injection. Moreover, all three doses of the extract significantly inhibited production of dark neurons in different regions of the hippocampus in the mentioned animal model. Conclusion: The present study showed that the hydro-alcoholic extract of R. damascena has anticonvulsant and neuroprotective effects. More investigations are needed to be done in order to better understand the responsible compound(s) as well as the possible mechanism(s)

    Objective:Coriandrum sativum (C. sativum) as a medicinal plant has been pointed to have analgesic, hypnotic and anti-oxidant effects. In the current study, a possible preventive effect of the hydro-alcoholic extract of the plant on neuronal damages was examined in pentylenetetrazole (PTZ) rat model of seizure.Materials and Methods: Forty male rats were divided into five main groups and treated by (1) saline, (2) PTZ: 100 mg/kg PTZ (i.p) and (3-5) 50, 100 and 200 mg/kg of hydro-alcoholic extract of C. sativum during seven consecutive days before PTZ injection.     Objective(s):Skin extracellular matrix, which contains type I and type III collagens, is involved in skin development. The aim of this study was to investigate type III collagen distribution pattern as well as its changes during pre and post-natal skin morphogenesis in rats. Materials and Methods: Ventral skins of Wistar rat embryos at different stages from 10 to 20 gestational day (E10-E20) and also one month and one year post natal rat pups were fixed in normalin, embedded in paraffin and 5 µm thick sections were incubated with Anti type III collagen antibody. In order to detect staining intensity, the reactions were observed and graded by three examiners separately. Kruskal-Wallis non-parametric statistical test and SPSS software version 11.5 were used to compare differences between samples. Results: Immunoreactivity of type III collagen was distributed weakly in the mesenchymal connective tissue on day 10 (E10). The observed reaction was increased onE12 and E14. This reaction was clear in basement membrane, relatively intensive in dermal papillae and moderate in dermal reticularis on E14. This immunoreactivity pattern was increased afterward on E16, not changed on E18 and decreased in dermal reticularis on E20. The density of collagen type III in dermal papillae and dermal reticularis in skin of one year old rats were decreased comparing to one month old rats. Conclusion: Our results showed that type III collagen is expressed and timely regulated during pre and post natal rat skin morphogenesis
