Isolation of recombinant antibodies directed against surface proteins of Clostridium difficile
Clostridium difficile has emerged as an increasingly important nosocomial pathogen and the prime causative agent of antibiotic-associated diarrhoea and pseudomembranous colitis in humans. In addition to toxins A and B, immunological studies using antisera from patients infected with C. difficile have shown that a number of other bacterial factors contribute to the pathogenesis, including surface proteins, which are responsible for adhesion, motility and other interactions with the human host. In this study, various clostridial targets, including FliC, FliD and cell wall protein 66, were expressed and purified. Phage antibody display yielded a large panel of specific recombinant antibodies, which were expressed, purified and characterised. Reactions of the recombinant antibodies with their targets were detected by enzyme-linked immunosorbent assay; and Western blotting suggested that linear rather than conformational epitopes were recognised. Binding of the recombinant antibodies to surface-layer proteins and their components showed strain specificity, with good recognition of proteins from C. difficile 630. However, no reaction was observed for strain R20291—a representative of the 027 ribotype. Binding of the recombinant antibodies to C. difficile M120 extracts indicated that a component of a surface-layer protein of this strain might possess immunoglobulin-binding activities. Second-order weak lensing from modified gravity
We explore the sensitivity of weak gravitational lensing to second-order
corrections to the spacetime metric within a cosmological adaptation of the
parameterized post-Newtonian framework. Whereas one might expect nonlinearities
of the gravitational field to introduce non-Gaussianity into the statistics of
the lensing convergence field, we show that such corrections are actually
always small within a broad class of scalar-tensor theories of gravity. We show
this by first computing the weak lensing convergence within our parameterized
framework to second order in the gravitational potential, and then computing
the relevant post-Newtonian parameters for scalar-tensor gravity theories. In
doing so we show that this potential systematic factor is generically
negligible, thus clearing the way for weak lensing to provide a direct tracer
of mass on cosmological scales for a wide class of gravity theories despite

ELASTICITY: Topological Characterization of Robustness in Complex Networks
Just as a herd of animals relies on its robust social structure to survive in
the wild, similarly robustness is a crucial characteristic for the survival of
a complex network under attack. The capacity to measure robustness in complex
networks defines the resolve of a network to maintain functionality in the
advent of classical component failures and at the onset of cryptic malicious
attacks. To date, robustness metrics are deficient and unfortunately the
following dilemmas exist: accurate models necessitate complex analysis while
conversely, simple models lack applicability to our definition of robustness.
In this paper, we define robustness and present a novel metric, elasticity- a
bridge between accuracy and complexity-a link in the chain of network
robustness. Additionally, we explore the performance of elasticity on Internet
topologies and online social networks, and articulate results
Thermal-structural analyses of Space Shuttle Main Engine (SSME) hot section components
Three dimensional nonlinear finite element heat transfer and structural analyses were performed for the first stage high pressure fuel turbopump (HPFTP) blade of the space shuttle main engine (SSME). Directionally solidified (DS) MAR-M 246 and single crystal (SC) PWA-1480 material properties were used for the analyses. Analytical conditions were based on a typical test stand engine cycle. Blade temperature and stress strain histories were calculated by using the MARC finite element computer code. The structural response of an SSME turbine blade was assessed and a greater understanding of blade damage mechanisms, convective cooling effects, and thermal mechanical effects was gained
Price and tax policy for semi-subsistence agriculture in Ethiopia
In the case of semi-subsistence agriculture where wage employment is not available, the role played by prices and taxes in determining production and consumption decisions is not clearly established by economic theories of household choice. This study demonstrates that where choices in production, consumption, and leisure can be made independently, farmers will decide what to grow on the basis of their preferences for marketed goods. The paper also points out that the choice will be affected by the level and type of taxation imposed. The paper shows the impact of four taxes -- agricultural revenue, land, production and marketed goods consumption -- on crop production and tax revenues. This paper also reports on a model of production in Ethiopia. The results of this study give strong evidence of the role of producer and consumer prices in semi-subsistence agriculture. 3D angle-of-arrival positioning using von Mises-Fisher distribution
We propose modeling an angle-of-arrival (AOA) positioning measurement as a
von Mises-Fisher (VMF) distributed unit vector instead of the conventional
normally distributed azimuth and elevation measurements. Describing the
2-dimensional AOA measurement with three numbers removes discontinuities and
reduces nonlinearity at the poles of the azimuth-elevation coordinate system.
Our computer simulations show that the proposed VMF measurement noise model
based filters outperform the normal distribution based algorithms in accuracy
in a scenario where close-to-pole measurements occur frequently.Comment: 5 page
Coordinating Complementary Waveforms for Sidelobe Suppression
We present a general method for constructing radar transmit pulse trains and
receive filters for which the radar point-spread function in delay and Doppler,
given by the cross-ambiguity function of the transmit pulse train and the pulse
train used in the receive filter, is essentially free of range sidelobes inside
a Doppler interval around the zero-Doppler axis. The transmit pulse train is
constructed by coordinating the transmission of a pair of Golay complementary
waveforms across time according to zeros and ones in a binary sequence P. The
pulse train used to filter the received signal is constructed in a similar way,
in terms of sequencing the Golay waveforms, but each waveform in the pulse
train is weighted by an element from another sequence Q. We show that a
spectrum jointly determined by P and Q sequences controls the size of the range
sidelobes of the cross-ambiguity function and by properly choosing P and Q we
can clear out the range sidelobes inside a Doppler interval around the zero-
Doppler axis. The joint design of P and Q enables a tradeoff between the order
of the spectral null for range sidelobe suppression and the signal-to-noise
ratio at the receiver output. We establish this trade-off and derive a
necessary and sufficient condition for the construction of P and Q sequences
that produce a null of a desired order
