Ancient feeding ecology inferred from stable isotopic evidence from fossil horses in South America over the past 3 Ma
[Background]: Stable isotope ratios (13C/12C and 18O/16O) in fossil teeth and bone provide key archives for understanding the ecology of extinct horses during the Plio-Pleistocene in South America; however, what happened in areas of sympatry between Equus (Amerhippus) and Hippidion is less understood.[Results]: Here, we use stable carbon and oxygen isotopes preserved in 67 fossil tooth and bone samples for seven species of horses from 25 different localities to document the magnitude of the dietary shifts of horses and ancient floral change during the Plio-Pleistocene. Dietary reconstructions inferred from stable isotopes of both genera of horses present in South America document dietary separation and environmental changes in ancient ecosystems, including C3/C4 transitions. Stable isotope data demonstrate changes in C4 grass consumption, inter-species dietary partitioning and variation in isotopic niche breadth of mixed feeders with latitudinal gradient.[Conclusions]: The data for Hippidion indicate a preference varying from C3 plants to mixed C3-C4 plants in their diet. Equus (Amerhippus) shows three different patterns of dietary partitioning Equus (A.) neogeus from the province of Buenos Aires indicate a preference for C3 plants in the diet. Equus (A.) andium from Ecuador and Equus (A.) insulatus from Bolivia show a preference for to a diet of mixed C3-C4 plants, while Equus (A.) santaeelenae from La Carolina (sea level of Ecuador) and Brazil are mostly C4 feeders. These results confirm that ancient feeding ecology cannot always be inferred from dental morphology. While the carbon isotope composition of horses skeletal material decreased as latitude increased, we found evidence of boundary between a mixed C3/C4 diet signal and a pure C4 signal around 32° S and a change from a mixed diet signal to an exclusively C3 signal around 35°S. We found that the horses living at high altitudes and at low to middle latitude still have a C4 component in their diet, except the specimens from 4000 m, which have a pure C3 diet. The change in altitudinal vegetation gradients during the Pleistocene is one of several possibilities to explain the C4 dietary component in horses living at high altitudes. Changes in white adipose tissue metabolism induced by resveratrol in rats.
BACKGROUND: A remarkable range of biological functions have been ascribed to resveratrol. Recently, this polyphenol has been shown to have body fat lowering effects. The aim of the present study was to assess some of the potential underlying mechanisms of action which take place in adipose tissue. METHODS: Sixteen male Sprague-Dawley rats were randomly divided into two groups: control and treated with 30 mg resveratrol/kg body weight/d. All rats were fed an obesogenic diet and after six weeks of treatment white adipose tissues were dissected. Lipoprotein lipase activity was assessed by fluorimetry, acetyl-CoA carboxylase by radiometry, and malic enzyme, glucose-6P-dehydrogenase and fatty acid synthase by spectrophotometry. Gene expression levels of acetyl-CoA carboxylase, fatty acid synthase, lipoprotein lipase, hormone-sensitive lipase, adipose triglyceride lipase, PPAR-gamma, SREBP-1c and perilipin were assessed by Real time RT-PCR. The amount of resveratrol metabolites in adipose tissue was measured by chromatography. RESULTS: There was no difference in the final body weight of the rats; however, adipose tissues were significantly decreased in the resveratrol-treated group. Resveratrol reduced the activity of lipogenic enzymes, as well as that of heparin-releasable lipoprotein lipase. Moreover, a significant reduction was induced by this polyphenol in hormone-sensitive lipase mRNA levels. No significant changes were observed in other genes. Total amount of resveratrol metabolites in adipose tissue was 2.66 ± 0.55 nmol/g tissue. CONCLUSIONS: It can be proposed that the body fat-lowering effect of resveratrol is mediated, at least in part, by a reduction in fatty acid uptake from circulating triacylglycerols and also in de novo lipogenesis
La fauna del pleistoceno inferior de la sierra de Quibas (Abanilla, Murcia)
El yacimiento kárstico cuaternario de la Sierra de Quibas (Abanilla, Murcia) ha proporcionado
una amplia lista faunística constituida por 53 especies repartidas entre gasterópodos,
miriápodos, anfibios, reptiles, aves y mamíferos. En el conjunto de los gasterópodos
destaca Palaeoglandina, un género que se extingue en el resto de Europa durante el Plioceno
y que se mantiene como relicto en el Pleistoceno de la Península Ibérica. Dos de las
especies de aves determinadas, Gypaetus barbatus y Gerontieus eremita, y una de las serpientes,
Elaphe cf. E. sealaris, apenas se hallan representadas en el registro fósil. Hay además
varias especies de aves cuya aparición en Quibas supone la primera cita en el Pleistoceno
inferior de la Península Ibérica. Entre los mamíferos posee una especial relevancia la
presencia del cercopitécido Macaca sylvanus. La asociación de los taxones Arvieola deuealíon,
Castillomys rivas rivas, Elíomys intermedius, Equus altidens y Capra sp. aff. C.
alba permite la correlación con Plines 1, Orce 3 y Venta Micena, entre otros. El yacimiento
de Quibas puede situarse, por tanto, antes del final del Pleistoceno inferior, con una antigüedad
entre 1.3 y 1 Ma. Por lo que se refiere a las condiciones paleoclimáticas, podemos
inferir a partir de la asociación faunística un régimen xerófilo, muy semejante al actual en
el área geográfica, aunque quizás con valores de humedad y temperatura algo superiores.
El entorno de la cavidad kárstica estaba formado por roquedo calcáreo con áreas abiertas
de matorral, pero en las proximidades se desarrollaron humedales y zonas arboladas, como
así lo atestigua la presencia de aves y micromamíferos típicos de estos hábitats.The Quaternary karstic site of Sierra de Quibas (Abanilla, Murcia, Spain) has provided
a wide faunallist with 53 species distributed among gastropods, myriapods, amphibians,
reptiles, birds and mammals. Especially interesting among the gastropods is
Palaeoglandina that became extinct in Europe during the Pliocene. It remains as a relict
genus in the Pleistocene of the Iberian Peninsula. Two of the birds, Gypaetus barbatus
and Geronticus eremita, and a snake, Elaphe cf. E. sealaris, are scarcely represented in the fossil record. For several birds, it is the first record in the Lower Pleistocene of the
Iberian Peninsula. Among the mammals, the presence of the Cercopithecine Macaca sylvanus
is especially relevant. The assemblage of the taxa Arvicola deucalion, Castillomys
rivas rivas, Eliomys intermedius, Equus altidens and Capra sp. aff. C. alba allows the
correlation with Plines 1, Orce 3 and Venta Micena, among other sites. Therefore Quibas
can be clated before the end of Lower Pleistocene, between 1.3 and 1 Ma. A dry paleoclimatic
regime, very similar to the current climate in the geographical area, though perhaps
slightly wetter and warmer, can be inferred from the faunal assemblage. The environment
of the karstic cavity was a rocky place with open brushwood areas, but in the
proximities there were wetlands and woodlands, as can be inferred from the presence of
birds and micromammals characteristic of these [email protected] [email protected]
Future mmVLBI Research with ALMA: a European vision
Very long baseline interferometry at millimetre/submillimetre wavelengths (mmVLBI) offers the highest achievable spatial resolution at any wavelength in astronomy. The anticipated inclusion of ALMA as a phased array into a global VLBI network will bring unprecedented sensitivity and a transformational leap in capabilities for mmVLBI. Building on years of pioneering efforts in the US and Europe the ongoing ALMA Phasing Project (APP), a US-led international collaboration with MPIfR-led European contributions, is expected to deliver a beamformer and VLBI capability to ALMA by the end of 2014 (APP: Fish et al. 2013, arXiv:1309.3519). This report focuses on the future use of mmVLBI by the international users community from a European viewpoint. Firstly, it highlights the intense science interest in Europe in future mmVLBI observations as compiled from the responses to a general call to the European community for future research projects. A wide range of research is presented that includes, amongst others: - Imaging the event horizon of the black hole at the centre of the Galaxy - Testing the theory of General Relativity an/or searching for alternative theories - Studying the origin of AGN jets and jet formation - Cosmological evolution of galaxies and BHs, AGN feedback - Masers in the Milky Way (in stars and star-forming regions) - Extragalactic emission lines and astro-chemistry - Redshifted absorption lines in distant galaxies and study of the ISM and circumnuclear gas - Pulsars, neutron stars, X-ray binaries - Testing cosmology - Testing fundamental physical constant
DNA methylation epigenotypes in breast cancer molecular subtypes
12 páginas, 3 figuras, 3 tablas.-- et al.[Introduction]: Identification of gene expression-based breast cancer subtypes is considered a critical means of prognostication. Genetic mutations along with epigenetic alterations contribute to gene-expression changes occurring in breast cancer. So far, these epigenetic contributions to sporadic breast cancer subtypes have not been well characterized, and only a limited understanding exists of the epigenetic mechanisms affected in those particular breast cancer subtypes. The present study was undertaken to dissect the breast cancer methylome and to deliver specific epigenotypes associated with particular breast cancer subtypes.
[Methods]: By using a microarray approach, we analyzed DNA methylation in regulatory regions of 806 cancer-related genes in 28 breast cancer paired samples. We subsequently performed substantial technical and biologic validation by pyrosequencing, investigating the top qualifying 19 CpG regions in independent cohorts encompassing 47 basal-like, 44 ERBB2+ overexpressing, 48 luminal A, and 48 luminal B paired breast cancer/adjacent tissues. With the all-subset selection method, we identified the most subtype-predictive methylation profiles in multivariable logistic regression analysis.
[Results]: The approach efficiently recognized 15 individual CpG loci differentially methylated in breast cancer tumor subtypes. We further identified novel subtype-specific epigenotypes that clearly demonstrate the differences in the methylation profiles of basal-like and human epidermal growth factor 2 (HER2)-overexpressing tumors.
[Conclusions]: Our results provide evidence that well-defined DNA methylation profiles enable breast cancer subtype prediction and support the utilization of this biomarker for prognostication and therapeutic stratification of patients with breast cancer.This work was supported by grants from project CGL2008-01131

Effect of remote ischaemic conditioning on clinical outcomes in patients with acute myocardial infarction (CONDI-2/ERIC-PPCI): a single-blind randomised controlled trial.
BACKGROUND: Remote ischaemic conditioning with transient ischaemia and reperfusion applied to the arm has been shown to reduce myocardial infarct size in patients with ST-elevation myocardial infarction (STEMI) undergoing primary percutaneous coronary intervention (PPCI). We investigated whether remote ischaemic conditioning could reduce the incidence of cardiac death and hospitalisation for heart failure at 12 months. METHODS: We did an international investigator-initiated, prospective, single-blind, randomised controlled trial (CONDI-2/ERIC-PPCI) at 33 centres across the UK, Denmark, Spain, and Serbia. Patients (age >18 years) with suspected STEMI and who were eligible for PPCI were randomly allocated (1:1, stratified by centre with a permuted block method) to receive standard treatment (including a sham simulated remote ischaemic conditioning intervention at UK sites only) or remote ischaemic conditioning treatment (intermittent ischaemia and reperfusion applied to the arm through four cycles of 5-min inflation and 5-min deflation of an automated cuff device) before PPCI. Investigators responsible for data collection and outcome assessment were masked to treatment allocation. The primary combined endpoint was cardiac death or hospitalisation for heart failure at 12 months in the intention-to-treat population. This trial is registered with ClinicalTrials.gov (NCT02342522) and is completed. FINDINGS: Between Nov 6, 2013, and March 31, 2018, 5401 patients were randomly allocated to either the control group (n=2701) or the remote ischaemic conditioning group (n=2700). After exclusion of patients upon hospital arrival or loss to follow-up, 2569 patients in the control group and 2546 in the intervention group were included in the intention-to-treat analysis. At 12 months post-PPCI, the Kaplan-Meier-estimated frequencies of cardiac death or hospitalisation for heart failure (the primary endpoint) were 220 (8·6%) patients in the control group and 239 (9·4%) in the remote ischaemic conditioning group (hazard ratio 1·10 [95% CI 0·91-1·32], p=0·32 for intervention versus control). No important unexpected adverse events or side effects of remote ischaemic conditioning were observed. INTERPRETATION: Remote ischaemic conditioning does not improve clinical outcomes (cardiac death or hospitalisation for heart failure) at 12 months in patients with STEMI undergoing PPCI. Etude et comparaison des restes d' <i>Hipparion crassum</i> gervais (Perissodactyla, Mammalia) proyenant de la nouvelle localite «Le Soler (Lit de la Tet)», Pyrenees Orientales, France
The objective of the present work is the study of fossil remains of <i>Hipparion crassum</i> ssp. collected in Lit de la Tet, district of «Le Soler», by J. Aymar, in November 1974.<br>A comparison is carried out between the other forms of Upper Miocene and Pliocene <i>Hipparion</i> (Turolian and Lower Ruscinian) from the Mediterranean basin and other sites where <i>H. crassum</i> is known to be found.<br><br>El objetivo de este trabajo es el estudio de los restos fósiles de <i>Hipparion crassum</i> ssp. encontrados por J.
