    Interventions at the Supreme Court of Canada: Accuracy, Affiliation, and Acceptance

    Interveners make submissions in about half of the cases heard by the Supreme Court of Canada, but the motivations for and consequences of the practice are not clearly understood. Considered broadly, there are at least three functions that the practice of intervention might perform. The first possibility is that hearing from interveners might provide objectively useful information to the Court (i.e., interveners might promote the accuracy of the Court\u27s decision making). A second possibility is that the practice of intervention allows interveners to provide the best argument for certain partisan interests that judges might want to affiliate with. A third possibility is that interventions are allowed mainly (if not only) so that intervening parties feel they have had their voices heard by the Court and the greater public, including Parliament, regardless of the effect on the outcome of the appeal (i.e., the Court might be promoting the acceptability of its decisions by allowing for an outlet for expression). We examine empirically the role of interveners in all the cases decided by the Supreme Court of Canada from January 2000 to July 2009 and find statistical evidence that interveners matter

    Morphology of the larvae of Rhantaticus congestus (Klug, 1833) and phylogenetic comparison with other known Aciliini (Coleoptera: Dytiscidae: Dytiscinae)

    We describe the second- and third instar larvae of the diving beetle Rhantaticus congestus (Klug, 1833), including detailed morphometric and chaetotaxic analyses of the cephalic capsule, head appendages, legs, terminal abdominal segment and urogomphi in order to discover useful characters for distinguishing Rhantaticus Sharp, 1882 larvae from those of other known Aciliini (Coleoptera: Dytiscidae: Dytiscinae). A parsimony analysis based on 94 larval characteristics of nine Aciliini species in five genera (Acilius Leach, 1817, Graphoderus Dejean, 1833, Rhantaticus, Sandracottus Sharp, 1882, Thermonectus Dejean, 1833) was conducted using the program TNT. Rhantaticus shares with all these genera several larval character states which support its inclusion in the Aciliini. Whereas Rhantaticus larva stands out from other known genera by several unique character states, our parsimony analysis did not recover any clear phylogenetic position of this genus within the Aciliini.Fil: Alarie, Yves.     A microfluidic chip integrating DNA extraction and real-time PCR for the detection of bacteria in saliva

    A microfluidic chip integrating DNA extraction, amplification, and detection for the identification of bacteria in saliva is described. The chip design integrated a monolithic aluminum oxide membrane (AOM) for DNA extraction with seven parallel reaction wells for real-time polymerase chain reaction (rtPCR) amplification of the extracted DNA. Samples were first heated to lyse target organisms and then added to the chip and filtered through the nanoporous AOM to extract the DNA. PCR reagents were added to each of the wells and the chip was thermocycled. Identification of Streptococcus mutans in a saliva sample is demonstrated along with the detection of 300 fg (100–125 copies) of both methicillin-susceptible Staphylococcus aureus (MSSA) and methicillin-resistant S. aureus (MRSA) genomic DNA (gDNA) spiked into a saliva sample. Multiple target species and strains of bacteria can be simultaneously identified in the same sample by varying the primers and probes used in each of the seven reaction wells. In initial tests, as little as 30 fg (8–12 copies) of MSSA gDNA in buffer has been successfully amplified and detected with this device

    Development of a Photothermal Absorbance Detector for Use with Microfluidic Devices

    The development of a photothermal absorbance detector for use with microfluidic devices is described. Unlike thermooptical techniques that rely on measuring refractive index changes, the solution viscosity is probed by continuously monitoring solution conductivity. Platinum electrodes microfabricated on a quartz substrate and bonded to a substrate containing the microchannels enable contact conductivity measurements. The effects of excitation frequency and voltage, electrode spacing, laser power, and laser modulation (chopping) frequency were evaluated experimentally. In the current configuration a limit of detection of 5 nM for DABSYL-tagged glucosamine was obtained using long injections (to give flat-topped peaks). This corresponds to an absorbance of 4.4 × 10−7 AU. Separation and detection of DABSYL-tagged glycine, proline, and tryptophan is also shown to demonstrate the feasibility of the method. In addition, simulations were used to investigate the applicability of the technique to small volume platforms

    A Device for Performing Lateral Conductance Measurements on Individual Double-Stranded DNA Molecules

    A nanofluidic device is described that is capable of electrically monitoring the driven translocation of DNA molecules through a nanochannel. This is achieved by intersecting a long transport channel with a shorter orthogonal nanochannel. The ionic conductance of this transverse nanochannel is monitored while DNA is electrokinetically driven through the transport channel. When DNA passes the intersection, the transverse conductance is altered, resulting in a transient current response. In 1 M KCl solutions, this was found to be a current enhancement of 5–25%, relative to the baseline transverse ionic current. Two different device geometries were investigated. In one device, the DNA was detected after it was fully inserted into and translocating through the transport nanochannel. In the other device, the DNA was detected while it was in the process of entering the nanochannel. It was found that these two conditions are characterized by different transport dynamics. Simultaneous optical and electrical monitoring of DNA translocation confirmed that the transient events originated from DNA transport through the nanochannel intersection

    Risk-stratified approach to breast cancer screening in canada: Women’s knowledge of the legislative context and concerns about discrimination from genetic and other predictive health data

    The success of risk-stratified approaches in improving population-based breast cancer screening programs depends in no small part on women’s buy-in. Fear of genetic discrimination (GD) could be a potential barrier to genetic testing uptake as part of risk assessment. Thus, the objective of this study was twofold. First, to evaluate Canadian women’s knowledge of the legislative context governing GD. Second, to assess their concerns about the possible use of breast cancer risk levels by insurance companies or employers. We use a cross-sectional survey of 4293 (age: 30–69) women, conducted in four Canadian provinces (Alberta, British Colombia, Ontario and QuĂ©bec). Canadian women’s knowledge of the regulatory framework for GD is relatively limited, with some gaps and misconceptions noted. About a third (34.7%) of the participants had a lot of concerns about the use of their health information by employers or insurers; another third had some concerns (31.9%), while 20% had no concerns. There is a need to further educate and inform the Canadian public about GD and the legal protections that exist to prevent it. Enhanced knowledge could facilitate the implementation and uptake of risk prediction informed by genetic factors, such as the risk-stratified approach to breast cancer screening that includes risk levels
