    Qualidade de vida de mulheres com incontinência urinária

    The aim was to measure the quality of life of women with complaints of urinary incontinence. These women received medical care at the Urology Outpatient Care Center of a teaching hospital. Forty-three women who had complaints of UI were interviewed. All ethical principles for research involving human beings were respected. The average age was 50.7 years. 62.5% reported urinary leakage for 5-9 years. There were several allusions to physical and psychological damages caused by urinary incontinence: 33.5% to psychosocial interactions, 23.3% to sex life, 41.9% reported depression and social isolation, 27.9% sleep disorders and 76.7% embarrassment due to urine leakage. The obtained data permit amplified actions for treatment and counseling of women with urinary incontinence.Teve-se, aqui, o objetivo de investigar a qualidade de vida de mulheres com queixa de incontinência urinária que buscaram atendimento médico em ambulatório de urologia, de um hospital de ensino. Foram entrevistadas 43 mulheres com queixa de IU, preservando-se os aspetos éticos de pesquisa em seres humanos. Obteve-se, como resultados, que a idade média foi de 50,7 anos, 62,8% referiram perda de urina entre 5 e 9 anos. Houve muitos relatos de danos físicos e psicológicos decorrentes da IU: 33,5% na interação psicossocial, 23,3% na vida sexual, 41,9% depressão e isolamento social, 27,9% alterações do sono, 76,7% relataram constrangimento por perda de urina. Conclui-se que os dados obtidos permitem o desenvolvimento de ações amplas no atendimento e orientação a mulheres com IU.La finalidad fue investigar la calidad de vida de mujeres con quejas de incontinencia urinaria que buscaron atendimiento médico en ambulatorio de urología de un hospital escuela.     To describe how a community-academic partnership between Taller Salud Inc., a community-based organization, and the Puerto Rico Community Cancer Control Outreach Program of the University of Puerto Rico was crucial in the adaptation and implementation of Cultivando La Salud (CLS), an evidence-based educational outreach program designed to increase breast and cervical cancer screening among Hispanic women living in Puerto Rico. This collaboration facilitated the review and adaptation of the CLS intervention to improve cultural appropriateness, relevance, and acceptability for Puerto Rican women

    BCLXL PROTAC degrader DT2216 targets secondary plasma cell leukemia addicted to BCLXL for survival

    Secondary plasma cell leukemia (sPCL) is a rare form of aggressive plasma cell malignancy arising mostly at end-stage refractory multiple myeloma and consequently presenting limited therapeutic options. We analyzed 13 sPCL for their sensitivity to BH3 mimetics targeting either BCL2 (venetoclax) or BCLXL (A1155463) and showed that 3 sPCL were efficiently killed by venetoclax and 3 sPCL by A1155463. Accordingly, BH3 profiling of 2 sPCL sensitive to BCLXL inhibition confirmed their high BCLXL primed profile. While targeting BCLXL using BH3 mimetics induces platelets on-target drug toxicity, the recent development of DT2216, a clinical-stage BCLXL proteolysis targeting chimera PROTAC compound, provides an alternative strategy to target BCLXL. Indeed, DT2216 specifically degrades BCLXL via VHL E3 ligase, without inducing thrombocytopenia. We demonstrated in human myeloma cell lines and sPCL that sensitivity to DT2216 strongly correlated with the sensitivity to A1155463. Interestingly, we showed that low doses of DT2216 (nM range) were sufficient to specifically degrade BCLXL after 48 hours of treatment, consistent with VHL expression, in all cell lines but irrespectively to DT2216 sensitivity. In myeloma cells, DT2216 induced apoptotic cell death and triggered BAX and BAK activation. In conclusion, our study demonstrated that patients with sPCL addicted to BCLXL, a small but a very challenging group, could potentially receive therapeutic benefit from DT2216.     Assessment of a Novel VEGF Targeted Agent Using Patient-Derived Tumor Tissue Xenograft Models of Colon Carcinoma with Lymphatic and Hepatic Metastases

    The lack of appropriate tumor models of primary tumors and corresponding metastases that can reliably predict for response to anticancer agents remains a major deficiency in the clinical practice of cancer therapy. It was the aim of our study to establish patient-derived tumor tissue (PDTT) xenograft models of colon carcinoma with lymphatic and hepatic metastases useful for testing of novel molecularly targeted agents. PDTT of primary colon carcinoma, lymphatic and hepatic metastases were used to create xenograft models. Hematoxylin and eosin staining, immunohistochemical staining, genome-wide gene expression analysis, pyrosequencing, qRT-PCR, and western blotting were used to determine the biological stability of the xenografts during serial transplantation compared with the original tumor tissues. Early passages of the PDTT xenograft models of primary colon carcinoma, lymphatic and hepatic metastases revealed a high degree of similarity with the original clinical tumor samples with regard to histology, immunohistochemistry, genes expression, and mutation status as well as mRNA expression. After we have ascertained that these xenografts models retained similar histopathological features and molecular signatures as the original tumors, drug sensitivities of the xenografts to a novel VEGF targeted agent, FP3 was evaluated. In this study, PDTT xenograft models of colon carcinoma with lymphatic and hepatic metastasis have been successfully established. They provide appropriate models for testing of novel molecularly targeted agents
