    Feeding, tentacle and gut morphology in five species of southern African intertidal holothuroids (Echinodermata)

    Light, scanning and transmission electron microscopy were used to compare the structure of the tentacles and digestive tracts of four species of intertidal dendrochlrote (Roweia stephensoni, Pseudocneila sykion, Aslia spyridophora, R. frauenfeldi frauenfeldi),and one species of aspidochirote holothuroid (Neostichopus grammatus). In addition, gut lengths and contents of the five species were compared. Gut contents were sieved to determine the size of the particulate matter ingested. Roweia stephensoni, P. sykion and A. spyridophora were found to be suspension feeders using dendritic tentacles to capture and ingest food particles mostly <53 μm in size. Roweia f. frauenfeldi was also a suspension feeder but, had atypical (reduced) dendritic tentacles which captured food particles between 250 μm-1.18 mm in size. Neostichopus grammatus was a deposit feeder, ingesting sediments mostly between 106-500 μm using tentacles which are peltate with ramified processes. The gut lengths of the four suspension-feeding species were found to be significantly (p <0.001) longer than that of the deposit feeder. The digestive tract of all species was composed of four tissue layers, with the digestive epithelial layer of the anterior and posterior ends of the intestine of suspension feeders being significantly thicker (52 to 57 μm) than that of the deposit feeder (about 19 to 29 μm). In addition, the epithelial layer of the intestine of suspension feeders contained more highly vesicular enterocytes than that of the deposit feeder

    Morphological and genetic differentiation of Patella granularis (Gastropoda: Patellidae): recognition of two sibling species along the coast of southern Africa

    Morphological and isozyme variations between 13 populations of the species hitherto named Patella granularis were investigated to see whether differences in shell structure between the west coast versus the south and east coasts of southern Africa are supported by other morphological features or by genetic differences. The shells showed a definite decrease in size from west to east, but this is correlated with productivity and is of no diagnostic use in distinguishing between populations. Discriminant functions analysis based on shell morphometrics failed to separate populations from the three coastal regions. Shells from the northern east coast do, however, have shell nodules with a dark pigmentation, distinctly separating them from those further south and west. No differences in radular or soft part morphology were detected between the populations, but the four northernmost populations on the east coast have a significantly shorter Z looping of the gut than the other populations along the coast. Significant microstructural differences in the sperm were also detected between these two groups of populations. Electrophoretic analysis of 16 enzyme loci failed to detect any significant differences between the west and south coast populations, but revealed a genetic identity (Nei) of 0.528 as well as four diagnostic alleles between the four northernmost populations from the east coast compared with those to the south and west. The two genetically distinct forms occurred sympatrically at one of the study sites on the east coast (Coffee Bay). It was concluded the two groupings were sufficiently different to warrant the recognition of a separate species, which is centred in KwaZulu-Natal on the east coast and extends south to Coffee Bay, from where it is replaced by P. granularis. There is, however, no evidence at all that the west coast populations are in any way separable from the remaining populations of P. granularis on the south coast

    The magnetic field structure in CTA 102 from high-resolution mm-VLBI observations during the flaring state in 2016-2017

    CONTEXT: Investigating the magnetic field structure in the innermost regions of relativistic jets is fundamental to understanding the crucial physical processes giving rise to jet formation, as well as to their extraordinary radiation output up to γ-ray energies. AIMS: We study the magnetic field structure of the quasar CTA 102 with 3 and 7 mm VLBI polarimetric observations, reaching an unprecedented resolution (∼50 μas). We also investigate the variability and physical processes occurring in the source during the observing period, which coincides with a very active state of the source over the entire electromagnetic spectrum. METHODS: We perform the Faraday rotation analysis using 3 and 7 mm data and we compare the obtained rotation measure (RM) map with the polarization evolution in 7 mm VLBA images. We study the kinematics and variability at 7 mm and infer the physical parameters associated with variability. From the analysis of γ-ray and X-ray data, we compute a minimum Doppler factor value required to explain the observed high-energy emission. RESULTS: Faraday rotation analysis shows a gradient in RM with a maximum value of ∼6 × 104⁴ rad m⁻² and intrinsic electric vector position angles (EVPAs) oriented around the centroid of the core, suggesting the presence of large-scale helical magnetic fields. Such a magnetic field structure is also visible in 7 mm images when a new superluminal component is crossing the core region. The 7 mm EVPA orientation is different when the component is exiting the core or crossing a stationary feature at ∼0.1 mas. The interaction between the superluminal component and a recollimation shock at ∼0.1 mas could have triggered the multi-wavelength flares. The variability Doppler factor associated with such an interaction is large enough to explain the high-energy emission and the remarkable optical flare occurred very close in time.Accepted manuscrip

    Spermiogenesis in three species of cicadas (Hemiptera: Cicadidae)

    Spermiogenesis in three species of cicadas representing one cicadettine (Monomatapa matoposa Boulard) and two cicadines (Diceroprocta biconica [Walker] and Kongota punctigera [Walker]) was investigated by light and electron microscopy. Although spermiogenesis was occurring in the testis of adult males of all species, earlier spermiogenic stages were observed in D. biconica only. While spermiogenesis was similar to that described for other insects, some differences were noted. For example granular material did not assemble around the centriole to form a centriolar adjunct but did accumulate in the cytoplasm of early spermatids adjacent to a region of the nuclear membrane where nuclear pores were aggregated. In late spermatids this material accumulated anterior to the mitochondrial derivatives in a developing postero-lateral nuclear groove. While this material has been named the ‘centriolar adjunct’ by previous authors, its formation away from the centriole raises questions about its true identity. Second, during acrosome maturation an ante-acrosomal region of cytoplasm develops. Although present in later spermatids, this region is lost in spermatozoa.     Reefs at Risk: A Map-Based Indicator of Threats to the Worlds Coral Reefs

    This report presents the first-ever detailed, map-based assessment of potential threats to coral reef ecosystems around the world. "Reefs at Risk" draws on 14 data sets (including maps of land cover, ports, settle-ments, and shipping lanes), information on 800 sites known to be degraded by people, and scientific expertise to model areas where reef degradation is predicted to occur, given existing human pressures on these areas. Results are an indicator of potential threat (risk), not a measure of actual condition. In some places, particularly where good management is practiced, reefs may be at risk but remain relatively healthy. In others, this indicator underestimates the degree to which reefs are threatened and degraded.Our results indicate that:Fifty-eight percent of the world's reefs are poten-tially threatened by human activity -- ranging from coastal development and destructive fishing practices to overexploitation of resources, marine pollution, and runoff from inland deforestation and farming.Coral reefs of Asia (Southeastern); the most species-rich on earth, are the most threatened of any region. More than 80 percent are at risk (undermedium and high potential threat), and over half are at high risk, primarily from coastal development and fishing-related pressures.Overexploitation and coastal development pose the greatest potential threat of the four risk categories considered in this study. Each, individually, affects a third of all reefs.The Pacific, which houses more reef area than any other region, is also the least threatened. About 60 percent of reefs here are at low risk.Outside of the Pacific, 70 percent of all reefs are at risk.At least 11 percent of the world's coral reefs contain high levels of reef fish biodiversity and are under high threat from human activities. These "hot spot" areas include almost all Philippine reefs, and coral communities off the coasts of Asia, the Comoros, and the Lesser Antilles in the Caribbean.Almost half a billion people -- 8 percent of the total global population -- live within 100 kilometers of a coral reef.Globally, more than 400 marine parks, sanctuaries, and reserves (marine protected areas) contain coral reefs. Most of these sites are very small -- more than 150 are under one square kilometer in size. At least 40 countries lack any marine protected areas for conserving their coral reef systems

    Increasing follow-up questionnaire response rates in a randomized controlled trial of telehealth for depression: three embedded controlled studies

    Background: Attrition is problematic in trials, and may be exacerbated in longer studies, telehealth trials and participants with depression – three features of The Healthlines Study. Advance notification, including a photograph and using action-oriented email subject lines might increase response rates, but require further investigation. We examined the effectiveness of these interventions in three embedded Healthlines studies. Methods: Based in different trial sites, participants with depression were alternately allocated to be pre-called or not ahead of the 8-month follow-up questionnaire (Study 1), randomized to receive a research team photograph or not with their 12-month questionnaire (Study 2), and randomized to receive an action-oriented (‘ACTION REQUIRED’) or standard (‘Questionnaire reminder’) 12-month email reminder (Study 3). Participants could complete online or postal questionnaires, and received up to five questionnaire reminders. The primary outcome was completion of the Patient Health Questionnaire (PHQ-9). Secondary outcome measures were the number of reminders and time to questionnaire completion. Results: Of a total of 609 Healthlines depression participants, 190, 251 and 231 participants were included in Studies 1–3 (intervention: 95, 126 and 115), respectively. Outcome completion was ≥90 % across studies, with no differences between trial arms (Study 1: OR 0.38, 95 % CI 0.07–2.10; Study 2: OR 0.84, 95 % CI 0.26–2.66; Study 3: OR 0.53 95 % CI 0.19–1.49). Pre-called participants were less likely to require a reminder (48.4 % vs 62.1 %, OR 0.41, 95 % CI 0.21–0.78), required fewer reminders (adjusted difference in means −0.67, 95 % CI −1.13 to −0.20), and completed follow-up quicker (median 8 vs 15 days, HR 1.35, 95 % CI 1.00–1.82) than control subjects. There were no significant between-group differences in Studies 2 or 3. Conclusions: Eventual response rates in this trial were high, with no further improvement from these interventions. While the photograph and email interventions were ineffective, pre-calling participants reduced time to completion. This strategy might be helpful when the timing of study completion is important. Researchers perceived a substantial benefit from the reduction in reminders with pre-calling, despite no overall decrease in net effort after accounting for pre-notification

    Multiple novel prostate cancer susceptibility signals identified by fine-mapping of known risk loci among Europeans

    Genome-wide association studies (GWAS) have identified numerous common prostate cancer (PrCa) susceptibility loci. We have fine-mapped 64 GWAS regions known at the conclusion of the iCOGS study using large-scale genotyping and imputation in 25 723 PrCa cases and 26 274 controls of European ancestry. We detected evidence for multiple independent signals at 16 regions, 12 of which contained additional newly identified significant associations. A single signal comprising a spectrum of correlated variation was observed at 39 regions; 35 of which are now described by a novel more significantly associated lead SNP, while the originally reported variant remained as the lead SNP only in 4 regions. We also confirmed two association signals in Europeans that had been previously reported only in East-Asian GWAS. Based on statistical evidence and linkage disequilibrium (LD) structure, we have curated and narrowed down the list of the most likely candidate causal variants for each region. Functional annotation using data from ENCODE filtered for PrCa cell lines and eQTL analysis demonstrated significant enrichment for overlap with bio-features within this set. By incorporating the novel risk variants identified here alongside the refined data for existing association signals, we estimate that these loci now explain ∼38.9% of the familial relative risk of PrCa, an 8.9% improvement over the previously reported GWAS tag SNPs. This suggests that a significant fraction of the heritability of PrCa may have been hidden during the discovery phase of GWAS, in particular due to the presence of multiple independent signals within the same regio

    Associations of common breast cancer susceptibility alleles with risk of breast cancer subtypes in BRCA1 and BRCA2 mutation carriers

    Introduction: More than 70 common alleles are known to be involved in breast cancer (BC) susceptibility, and several exhibit significant heterogeneity in their associations with different BC subtypes. Although there are differences in the association patterns between BRCA1 and BRCA2 mutation carriers and the general population for several loci, no study has comprehensively evaluated the associations of all known BC susceptibility alleles with risk of BC subtypes in BRCA1 and BRCA2 carriers. Methods: We used data from 15,252 BRCA1 and 8,211 BRCA2 carriers to analyze the associations between approximately 200,000 genetic variants on the iCOGS array and risk of BC subtypes defined by estrogen receptor (ER), progesterone receptor (PR), human epidermal growth factor receptor 2 (HER2) and triple-negative- (TN) status; morphologic subtypes; histological grade; and nodal involvement. Results: The estimated BC hazard ratios (HRs) for the 74 known BC alleles in BRCA1 carriers exhibited moderate correlations with the corresponding odds ratios from the general population.     Genetic mechanisms of critical illness in COVID-19.

    Host-mediated lung inflammation is present1, and drives mortality2, in the critical illness caused by coronavirus disease 2019 (COVID-19). Host genetic variants associated with critical illness may identify mechanistic targets for therapeutic development3. Here we report the results of the GenOMICC (Genetics Of Mortality In Critical Care) genome-wide association study in 2,244 critically ill patients with COVID-19 from 208 UK intensive care units. We have identified and replicated the following new genome-wide significant associations: on chromosome 12q24.13 (rs10735079, P = 1.65 × 10-8) in a gene cluster that encodes antiviral restriction enzyme activators (OAS1, OAS2 and OAS3); on chromosome 19p13.2 (rs74956615, P = 2.3 × 10-8) near the gene that encodes tyrosine kinase 2 (TYK2); on chromosome 19p13.3 (rs2109069, P = 3.98 ×  10-12) within the gene that encodes dipeptidyl peptidase 9 (DPP9); and on chromosome 21q22.1 (rs2236757, P = 4.99 × 10-8) in the interferon receptor gene IFNAR2. We identified potential targets for repurposing of licensed medications: using Mendelian randomization, we found evidence that low expression of IFNAR2, or high expression of TYK2, are associated with life-threatening disease; and transcriptome-wide association in lung tissue revealed that high expression of the monocyte-macrophage chemotactic receptor CCR2 is associated with severe COVID-19. Our results identify robust genetic signals relating to key host antiviral defence mechanisms and mediators of inflammatory organ damage in COVID-19. Both mechanisms may be amenable to targeted treatment with existing drugs. However, large-scale randomized clinical trials will be essential before any change to clinical practice
