Circulating leukocyte telomere length is highly heritable among families of Arab descent
Background
Telomere length, an indicator of ageing and longevity, has been correlated with several biomarkers of cardiometabolic disease in both Arab children and adults. It is not known, however, whether or not telomere length is a highly conserved inheritable trait in this homogeneous cohort, where age-related diseases are highly prevalent. As such, the aim of this study was to address the inheritability of telomere length in Saudi families and the impact of cardiometabolic disease biomarkers on telomere length.
Methods
A total of 119 randomly selected Saudi families (123 adults and 131 children) were included in this cross-sectional study. Anthropometrics were obtained and fasting blood samples were taken for routine analyses of fasting glucose and lipid profile. Leukocyte telomere length was determined using quantitative real time PCR.
Results
Telomere length was highly heritable as assessed by a parent-offspring regression [h2 = 0.64 (p = 0.0006)]. Telomere length was modestly associated with BMI (R2 0.07; p-value 0.0087), total cholesterol (R2 0.08; p-value 0.0033), and LDL-cholesterol (R2 0.15; p-value 3 x 10-5) after adjustments for gender, age and age within generation.
Conclusion
The high heritability of telomere length in Arab families, and the associations of telomere length with various cardiometabolic parameters suggest heritable genetic fetal and/or epigenetic influences on the early predisposition of Arab children to age-related diseases and accelerated ageing
Diabetes mellitus type 2 and other chronic non-communicable diseases in the central region, Saudi Arabia (riyadh cohort 2): a decade of an epidemic
<p>Abstract</p> <p>Background</p> <p>Follow-up epidemiologic studies are needed to assess trends and patterns of disease spread. No follow-up epidemiologic study has been done in the Kingdom of Saudi Arabia to assess the current prevalence of major chronic, noncommunicable diseases, specifically in the urban region, where modifiable risk factors remain rampant. This study aims to fill this gap.</p> <p>Methods</p> <p>A total of 9,149 adult Saudis ages seven to eighty years (5,357 males (58.6%) and 3,792 females (41.4%)) were randomly selected from the Riyadh Cohort Study for inclusion. Diagnosis of type 2 diabetes mellitus (DMT2) and obesity were based on the World Health Organization definitions. Diagnoses of hypertension and coronary artery disease (CAD) were based on the Seventh Joint National Committee on Prevention, Detection, Evaluation, and Treatment of High Blood Pressure and American Heart Association criteria, respectively.</p> <p>Results</p> <p>The overall crude prevalence of DMT2 was 23.1% (95% confidence interval (95% CI) 20.47 to 22.15). The age-adjusted prevalence of DMT2 was 31.6%. DMT2 prevalence was significantly higher in males, with an overall age-adjusted prevalence of 34.7% (95% CI 32.6 to 35.4), than in females, who had an overall age-adjusted prevalence of 28.6% (95% CI 26.7 to 29.3) (<it>P </it>< 0.001). The overall crude prevalence of obesity was 31.1% (95% CI 30.1 to 32.0). The age-adjusted prevalence of obesity was 40.0%. The prevalence of obesity was higher in females, with an overall prevalence of 36.5% (95% CI 35.1 to 37.83), than in males (25.1% (95% CI 23.7 to 26.3)) (<it>P </it>< 0.001). The age-adjusted prevalence of hypertension and CAD were 32.6% (95% CI 31.7 to 33.6) and 6.9% (95% CI 6.4 to 7.4), respectively.</p> <p>Conclusion</p> <p>Comparisons of our findings with earlier data show that the prevalence of DMT2, hypertension and CAD in Riyadh, Saudi Arabia, has alarmingly worsened. Aggressive promotion of public awareness, continued screening and early intervention are pivotal to boosting a positive response.</p
Gut Microbiota, Probiotics and Diabetes
Diabetes is a condition of multifactorial origin, involving several molecular mechanisms related to the intestinal
microbiota for its development. In type 2 diabetes, receptor activation and recognition by microorganisms from
the intestinal lumen may trigger inflammatory responses, inducing the phosphorylation of serine residues in insulin
receptor substrate-1, reducing insulin sensitivity. In type 1 diabetes, the lowered expression of adhesion proteins
within the intestinal epithelium favours a greater immune response that may result in destruction of pancreatic
β cells by CD8+ T-lymphocytes, and increased expression of interleukin-17, related to autoimmunity. Research in
animal models and humans has hypothesized whether the administration of probiotics may improve the prognosis
of diabetes through modulation of gut microbiota. We have shown in this review that a large body of evidence
suggests probiotics reduce the inflammatory response and oxidative stress, as well as increase the expression of
adhesion proteins within the intestinal epithelium, reducing intestinal permeability. Such effects increase insulin sensitivity and reduce autoimmune response. However, further investigations are required to clarify whether the administration of probiotics can be efficiently used for the prevention and management of diabetes
Quantitative Spectrophotometric Determination of Propranolol Hydrochloride in Pharmaceuticals Using Cerium(IV) Sulphate as Oxidimetric Reagent
Two simple, selective and economic spectrophotometric methods have been developed and validated for the analysis of the propranolol hydrochloride (PPH) in bulk drug and pharmaceuticals. The methods are based on the oxidation of PPH with cerium(IV) in acid medium and subsequent measurement of the excess cerium(IV) either by its reaction with p-dimethylamino benzaldehyde (pDMAB) and measuring the resulting color at 460 nm (method A) or by its reaction with sulphanilic acid (SAA) to give a pink colored product peaking at 540 nm (method B). The decrease in the absorption intensity of the colored products, due to the presence of the drug has been correlated with its concentration in the sample solution. Different experimental variables affecting the reaction are carefully studied and optimized. Under the optimum conditions, linear relationships with good correlation coefficients are found in the concentration ranges of 0.4-7.0 μg ml -1 for method A and 10-200 μg ml -1 for method B, respectively. The calculated molar absorptivity values are 3.72 � 10 4 for method A and 1.43 � 10 3 for method B. The methods have been validated for accuracy, precision, ruggedness and robustness and the results are found to be satisfactory. The proposed methods are successfully applied to the determination of PPH in pure and pharmaceutical dosage forms without any interference from the common excipients. The results obtained by the proposed methods are comparable with those obtained by the official methods
