Effect of Cyperus Rotundus Rhizomes on Blood Glucose, Lipid, Insulin and Hepatic Enzymes in Insulin Resistance Model of Male Rats
<p><strong><em>Background and Objectives:</em></strong><strong> </strong>Nowadays herbal medicine is excessively used in traditional medicine for diabetes. The Cyperus rotundus rhizomes in traditional medicine are used for decreasing blood glucose and lipid. Therefore, the aim of this study was to examine the effect of rhizomes extract of this plant in insulin resistance model of male rats.<strong></strong></p><p> </p><p><strong><em>Methods:</em></strong><strong> </strong>This experimental study was carried out on 36 adult male wistar rats in the weight range of 150-200(g). Diabetes was induced by fructose 10% in drinking water for 8 weeks, then the animals were divided randomly into 4 groups: control, Fructose induce, aqueous extracts of Cyperus rotundus rhizomes (500mg/kg), MeOH extracts of Cyperus rotundus rhizomes (500mg/kg). 48 hours after the last gavage with extracts, the rats were fasted for 12h, then blood samples were collected from heart, serum was dissented and blood glucose, insulin, fasting insulin resistance index (FIRI), lipid profile and hepatic enzymes were determined.</p><p> </p><p><strong><em>Results:</em></strong><strong> </strong>Glucose in fructose group in comparison with control group showed a significant increase. Insulin in aqueous and MeOH extracts groups in comparison with control group significantly increased. FIRI in aqueous extract group in comparison with control group was significantly higher (p&lt;0.05). Lipid profile was not changed in any of them. MeOH extract showed a decrease in all of the hepatic enzymes (Alkaline Phosphatase, Aspartate, and AST, Alanine Transferase).<strong></strong></p><p> </p><p><strong><em>Conclusion:</em></strong><strong> </strong>Aqueous has could not probably be effective in decreasing diabetic complications. But it is believed that with extensive study on MeOH extracts, we can reach a more efficient decrease in diabetes and antihepatotxicity.</p><p><em> </em></p>

Effect Of Aqueous And Hydroalcoholic Extract Of Beberis Vulgaris On Insulin Secretion From Islets Of Langerhans Isolated From Male Mice
Background & Aim: considering the use of Beberis vulgaris in traditional medicine as a blood sugar depressant, in this study, the effect of Beberis vulgaris extracts were investigated on the level of insulin secretion from islets isolated of langerhans in male mice.
Methods: This experimental study was carried out on 90 adult male mice, NMARI strains weighing 20-25 g. Pancreatic islets from normal mice were isolated by collagenase digestion method. Then the aqueous and hydro-alcoholic extract of Beberis vulgaris at 0.05, 0.1, and 1 mg/ml concentrations and glyburide at 1 and 10 μM concentrations were applied on islets isolated in three different concentration of glucose solution (2.8, 5.6 and 16.7 mM). Insulin secretion from hand-picked islets were evaluated in the static incubation system. The level of Insulin secretion was measured by the ELISA insulin kit. Data were analyzed with variance analysis.
Results: Insulin secretion was significantly increased at 16.7 mM glucose concentration in comparison with 2.8 and 5.6 mM glucose concentration (p<0.05). Incubation of pancreatic islets isolated at 2.8 and 5.6 mM glucose concentration and low concentrations of extract (0.05 and 0.1mg/ml) significantly increased the insulin secretion (p<0.05). Glyburide at 10 μM concentration was more effective than aqueous and hydro alcoholic extract of Beberis vulgaris at 16.7 mM glucose.
Conclusion: The present study supported the anti-diabetic effect of Beberis vulgaris extracts in vitro with low glucose concentration and it suggests that one of the anti diabetic mechanisms of this plant is via pancreatic islets
Effect of Arctium Lappa Root Extract on Glucose Levels and Insulin Resistance in Rats with High Sucrose Diet
Introduction: Diabetes Mellitus is a growing health problem in all over the world. Arctium Lappa has been used therapeutically in Europe, North America and Asia. Antioxidants and antidiabetic compounds have been found in the root of Arctium Lappa. This study intends to investigate the effects of Arctium Lappa root aqueous extract on glucose, insulin levels and Fasting Insulin Resistance Index in female rats with high sucrose diet. Methods: 40 female Wistar rats weighting 150-250(g) were applied. After having a diet induced by sucrose 50% in drinking water for 5 weeks, the animals were randomly divided into two groups of control, sucrose induced, and three groups of sucrose induced along with Arctium Lappa root aqueous extract (50,100,200 mg/Kg) (8 rats in each group). Treatment by extracts was used during 2 weeks (i.p.) and 24 hours after the last treatment, heart blood samples were gathered. After Blood samples were centrifuged, fasting plasma glucose (12 h) was determined by kit and fasting insulin concentration was assayed by Enzyme-linked immunosorbent assay (Elisa) methods. Result: Glucose levels, insulin and FIRI in sucrose group significantly increased in comparison with control group. Glucose levels in aqueous extract groups; 50 mg/kg (116.14±16.64mg/dl) and 200 mg/kg (90.66±22.58 mg/dl) in comparison with sucrose group (140.5±18.73 mg/dl) significantly decreased. Insulin level and FIRI in all of aqueous extract groups were significantly decreased (P<0.001) in comparison with sucrose group. Conclusions: Arctium Lappa root aqueous extracts in animal model has revealed significant decrease in blood glucose and insulin levels
