Detection of Malawi polyomavirus sequences in secondary lymphoid tissues from Italian healthy children: a transient site of infection
BACKGROUND:
The novel Malawi polyomavirus (MWPyV) was initially detected in stool specimens from healthy children and children with gastrointestinal symptoms, mostly diarrhea, indicating that MWPyV might play a role in human gastroenteric diseases. Recently, MWPyV sequences were additionally identified in respiratory secretions from both healthy and acutely ill children suggesting that MWPyV may have a tropism for different human tissues. This study was designed to investigate the possible sites of latency/persistence for MWPyV in a cohort of healthy Italian children.
METHODS:
Specimens (n\ub0 500) of tonsils, adenoids, blood, urines and feces, from 200 healthy and immunocompetent children (age range: 1-15 years) were tested for the amplification of the MWPyV LT antigen sequence by quantitative real-time PCR. Samples (n\ub0 80) of blood and urines from 40 age-matched children with autoimmune diseases, were screened for comparison. Polyomaviruses JC/BK and Epstein-Barr Virus (EBV) were also tested as markers of infection in all samples using the same molecular technique.
RESULTS:
In our series of healthy children, MWPyV was detected only in the lymphoid tissues showing a prevalence of 6 % in tonsils and 1 % in adenoids, although with a low viral load. No JCPyV or BKPyV co-infection was found in MWPyV positive samples, while EBV showed a similar percentage of both in tonsils and adenoids (38 and 37 %). Conversely, no MWPyV DNA was detected in stool from babies with gastroenteric syndrome. With regards to autoimmune children, neither MWPyV nor BKPyV were detected in blood, while JCPyV viremia was observed in 15 % (6/40) of children treated with Infliximab. Urinary BKPyV shedding was observed in 12.5 % (5/40) while JCPyV in 100 % of the samples.
CONCLUSIONS:
The detection of MWPyV sequences in tonsils and adenoids of healthy children suggests that secondary lymphoid tissues can harbour MWPyV probably as transient sites of persistence rather than actual sites of latency
The Impact of IoT Technologies on Our Activities
The evolution of the IoT technologies is spectacular in recent years, from
simple devices able to receive messages and to count the number of steps, to complex
devices which get information about the heartbeat rate monitoring, body position,
calories burned and even warn us about the UV radiation. The large amount of data
from IoT devices is important also for the business environment. We interviewed ten
people from Bucharest, who already used at least one IoT device, to understand the
impact of the use of IoT technologies on our lives. We identified common
characteristics of the people who use IoT applications and devices, and a user profile
was created. This could help to understand the advantages and disadvantages of the
IoT technologies usage and how it influences people in their lives
Social Media Networks as a Business Environment, During COVID-19 Crisis
Companies are now integrating social media marketing into their strategies and policies, for a better business performance. Social media campaigns are cost-effective and flexible in operations, offering many benefits. Between 15 - 30 August 2020 we created and managed a Facebook campaign for a local fashion brand. The online marketing becomes even more valuable in COVID-19 crisis, people shop and spend online more than ever. The marketing campaign generated important data to understand the customers demographic, interests, actions and helps us to make profiles of the potential clients. In this context we analyzed the conversion rate indicator, to understand if the marketing campaign was efficient or not. Conversion rate may not be the best to measure the success, but it is a great tool for tracking performance. We propose also the ROAS indicator, to measure the success of this marketing campaign
Attention to speech and music in young children with bilateral cochlear implants: a pupillometry study
Early bilateral cochlear implants (CIs) may enhance attention to speech, and reduce cognitive load in noisy environments. However, it is sometimes difficult to measure speech perception and listening effort, especially in very young children. Behavioral measures cannot always be obtained in young/uncooperative children, whereas objective measures are either difficult to assess or do not reliably correlate with behavioral measures. Recent studies have thus explored pupillometry as a possible objective measure. Here, pupillometry is introduced to assess attention to speech and music in noise in very young children with bilateral CIs (N = 14, age: 17–47 months), and in the age-matched group of normally-hearing (NH) children (N = 14, age: 22–48 months). The results show that the response to speech was affected by the presence of background noise only in children with CIs, but not NH children. Conversely, the presence of background noise altered pupil response to music only in in NH children. We conclude that whereas speech and music may receive comparable attention in comparable listening conditions, in young children with CIs, controlling for background noise affects attention to speech and speech processing more than in NH children. Potential implementations of the results for rehabilitation procedures are discussed
CaloCube: a novel calorimeter for high-energy cosmic rays in space
In order to extend the direct observation of high-energy cosmic rays up to
the PeV region, highly performing calorimeters with large geometrical
acceptance and high energy resolution are required. Within the constraint of
the total mass of the apparatus, crucial for a space mission, the calorimeters
must be optimized with respect to their geometrical acceptance, granularity and
absorption depth. CaloCube is a homogeneous calorimeter with cubic geometry, to
maximise the acceptance being sensitive to particles from every direction in
space; granularity is obtained by relying on small cubic scintillating crystals
as active elements. Different scintillating materials have been studied. The
crystal sizes and spacing among them have been optimized with respect to the
energy resolution. A prototype, based on CsI(Tl) cubic crystals, has been
constructed and tested with particle beams. Some results of tests with

Implantable hearing devices in clinical practice. Systematic review and consensus statements
Objective. Implantable hearing devices represent a modern and innovative solution for hearing restoration. Over the years, these high-tech devices have increasingly evolved but their use in clinical practice is not universally agreed in the scientific literature. Congresses, meetings, conferences, and consensus statements to achieve international agreement have been made. This work follows this line and aims to answer unsolved questions regarding examinations, selection criteria and surgery for implantable hearing devices. Materials and methods. A Consensus Working Group was established by the Italian Society of Otorhinolaryngology. A method group performed a systematic review for each single question to identify the current best evidence on the topic and to guide a multidisciplinary panel in developing the statements. Results. Twenty-nine consensus statements were approved by the Italian Society of Otorhinolaryngology. These were associated with 4 key area subtopics regarding pre -operative tests, otological, audiological and surgical indications. Conclusions. This consensus can be considered a further step forward to establish realistic guidelines on the debated topic of implantable hearing devices
Raccomandazioni per perfezionare i programmi regionali di screening uditivo neonatale universale in Italia
L’obiettivo dello screening uditivo neonatale universale è di ottenere una diagnosi precoce di ipoacusia congenita. Non appena confermata la perdita uditiva, è necessario intervenire dal punto di vista riabilitativo. Nell’ambito del progetto del Ministero della Salute CCM 2013 “Programma regionale di identificazione, intervento e presa in carico precoci per la prevenzione dei disturbi comunicativi nei bambini con deficit uditivo” un gruppo di professionisti appartenenti a centri di terzo livello con un programma regionale di screening uditivo neonatale, ha analizzato i limiti e i punti di forza dell’attuale impostazione dei programmi regionali di screening uditivo neonatale mediante l’analisi SWOT e la realizzazione di una matrice TOWS. Alcune criticità sono rappresentate dalla copertura dello screening e dal numero di persi allo screening. Sono state sviluppate raccomandazioni volte a migliorare l’efficacia di tali programmi. Sono emerse la necessità di una regolamentazione uniforme dei programmi di screening a livello regionale e nazionale, e il bisogno di ricevere informazioni e formazione aggiornate, di alta qualità , e condivise per familiari e operatori
A Multicenter Clinical Evaluation of Data Logging in Cochlear Implant Recipients Using Automated Scene Classification Technologies
Currently, there are no studies assessing everyday use of cochlear implant (CI) processors by recipients by means of objective tools. The Nucleus 6 sound processor features a data logging system capable of real-time recording of CI use in different acoustic environments and under various categories of loudness levels. In this study, we report data logged for the different scenes and different loudness levels of 1,366 CI patients, as recorded by SCAN. Monitoring device use in cochlear implant recipients of all ages provides important information about the listening conditions encountered in recipients' daily lives that may support counseling and assist in the further management of their device settings. The findings for this large cohort of active CI users confirm differences between age groups concerning device use and exposure to various noise environments, especially between the youngest and oldest age groups, while similar levels of loudness were observed
CaloCube: an innovative homogeneous calorimeter for the next-generation space experiments
The direct measurement of the cosmic-ray spectrum, up to the knee region, is one of the instrumental challenges for next generation space experiments. The main issue for these measurements is a steeply falling spectrum with increasing energy, so the physics performance of the space calorimeters are primarily determined by their geometrical acceptance and energy resolution. CaloCube is a three-year R&D project, approved and financed by INFN in 2014, aiming to optimize the design of a space-born calorimeter. The peculiarity of the design of CaloCube is its capability of detecting particles coming from any direction, and not only those on its upper surface. To ensure that the quality of the measurement does not depend on the arrival direction of the particles, the calorimeter will be designed as homogeneous and isotropic as possible. In addition, to achieve a high discrimination power for hadrons and nuclei with respect to electrons, the sensitive elements of the calorimeter need to have a fine 3-D sampling capability. In order to optimize the detector performances with respect to the total mass of the apparatus, which is the most important constraint for a space launch, a comparative study of different scintillating materials has been performed using detailed Monte Carlo simulation based on the FLUKA package. In parallel to simulation studies, a prototype consisting in 14 layers of 3 x 3 CsI(Tl) crystals per layer has been assembled and tested with particle beams. An overview of the obtained results during the first two years of the project will be presented and the future of the detector will be discussed too
EZH2 is a sensitive marker of malignancy in salivary gland tumors
BACKGROUND: The immunohistochemical detection of Enhancer of zeste homologue 2 (EZH2) proved to be a useful tool to recognize the malignant nature of tumors in a wide variety of neoplasms. The histological diagnostics of salivary gland tumors is a challenging task, and a reliable marker of malignancy would be extremely helpful. METHODS: EZH2 expression was investigated in 54 malignant and 40 benign salivary gland tumors of various histological types by standard immunohistochemistry. RESULTS: The majority (n = 52) of the malignant tumors stained positively, while all the investigated benign tumors were negative for EZH2. CONCLUSIONS: EZH2 expression in salivary gland tumors, similarly to the tumors of other organs is not characteristic for any tumor type, but is a solid marker of the malignant nature of the tumors
