    A computer-generated supercoiled model of the pUC19 plasmid

    DNA models have become a powerful tool in the simulation of radiation-induced molecular damage. Here, a computer code was developed which calculates the coordinates of individual atoms in supercoiled plasmid DNA. In this prototype study, the known base-pair sequence of the pUC19 plasmid has been utilized. The model was built in a three-step process. Firstly, a Monte Carlo simulation was performed to shape a segment chain skeleton. Checks on elastic energy, distance and unknotting were applied. The temperature was considered in two different ways: (1) it was kept constant at 293 K and (2) it was gradually reduced from 350 K to less than 10 K. Secondly, a special smoothing procedure was introduced here to remove the edges from the segment chain without changing the total curve length while avoiding the production of overshooting arcs. Finally, the base pair sequence was placed along the smoothed segment chain and the positions of all the atoms were calculated. As a first result, a few examples of the supercoiled plasmid models will be presented, demonstrating the strong influence of appropriate control of the system temperature

    Automated sleep stage classification in sleep apnoea using convolutional neural networks

    A sleep disorder is a condition that adversely impacts one\u27s ability to sleep well on a regular schedule. It also occurs as a consequence of numerous neurological sicknesses. These types of disorders can be investigated using laboratory-based polysomnography (PSG) signals. The detection of neurological disorders is exact and efficient thanks to the automated monitoring of sleep relegation stages. This automation method publicly presents a flexible deep learning model and machine learning approach utilizing raw electroencephalogram (EEG) signals. The deep learning model is a Deep Convolutional Neural Network (CNN) that analyses invariant time capacities and frequency actualities and collects assessment adaptations. It also captures the inviolate and long brief length setting conditions between the epochs and the degree of sleep stage relegation. This method uses an innovative function to calculate data loss and misclassified errors found while training the network for the sleep stage, considering the restrictions found in the publicly available sleep datasets. It is used in conjunction with machine learning techniques to forecast the best approach for the process. Its effectiveness is determined by using two open-source, public databases available from PhysioNet: two recordings with 5402 epoch counts. The technique used in this approach achieves an accuracy of 90.70%, precision of 90.50%, recall of 92.70%, and F-measure of 90.60%. The proposed method is more significant than existing models like AlexNet, ResNet, VGGNet, and LeNet. The comparative study of the models could be adopted for clinical use and modified based on the requirements

    Uncertainty Principle for Control of Ensembles of Oscillators Driven by Common Noise

    We discuss control techniques for noisy self-sustained oscillators with a focus on reliability, stability of the response to noisy driving, and oscillation coherence understood in the sense of constancy of oscillation frequency. For any kind of linear feedback control--single and multiple delay feedback, linear frequency filter, etc.--the phase diffusion constant, quantifying coherence, and the Lyapunov exponent, quantifying reliability, can be efficiently controlled but their ratio remains constant. Thus, an "uncertainty principle" can be formulated: the loss of reliability occurs when coherence is enhanced and, vice versa, coherence is weakened when reliability is enhanced. Treatment of this principle for ensembles of oscillators synchronized by common noise or global coupling reveals a substantial difference between the cases of slightly non-identical oscillators and identical ones with intrinsic noise.Comment: 10 pages, 5 figure

    Lymph node core biopsies reliably permit diagnosis of lymphoproliferative diseases. Real‐World Experience from 554 sequential core biopsies from a single centre

    INTRODUCTION: Whilst excision biopsy is traditionally preferred, advances in radiological and histological techniques warrant a re-look at core biopsy as a viable primary diagnostic method. METHOD: Over a 3-year period, all patients who underwent core biopsy to investigate lymphoma at our centre were included. RESULTS: 554 consecutive patients were included (40.1% prior lymphoma and 59.4% new presentations). Three or more cores were taken in 420 (75.8%) cases. Median time from request to biopsy and biopsy to histology report was 2 (0-40) days and 7 (1-24) days respectively. 510/544 (93.8%) biopsies were diagnostic. There was no difference in whether the biopsy was diagnostic based on indication (new vs. relapsed lymphoma) (p=0.445), whether biopsy was PET-directed (p=0.507), for T-cell lymphoma (p=0.468) or nodal vs. extra-nodal (p=0.693). Thirty-eight patients (6.9%) required a second biopsy due to inadequate tissue. In a patient experience survey, only 13.9% reported any complications (1 self-limiting minor bleeding, 4 bruising) whilst 16.7% reported any discomfort beyond 12 hours. CONCLUSION: Core biopsy performed by experienced radiologists and analysed by expert haemato-pathologists is a reliable, well-tolerated method for diagnosing lymphoma and confirming relapse. Multiple cores can be obtained under local anaesthetic yielding sufficient material in the majority of cases

    Shorter Lines Facilitate Reading in Those Who Struggle

    People with dyslexia, who ordinarily struggle to read, sometimes remark that reading is easier when e-readers are used. Here, we used eye tracking to observe high school students with dyslexia as they read using these devices. Among the factors investigated, we found that reading using a small device resulted in substantial benefits, improving reading speeds by 27%, reducing the number of fixations by 11%, and importantly, reducing the number of regressive saccades by more than a factor of 2, with no cost to comprehension. Given that an expected trade-off between horizontal and vertical regression was not observed when line lengths were altered, we speculate that these effects occur because sluggish attention spreads perception to the left as the gaze shifts during reading. Short lines eliminate crowded text to the left, reducing regression. The effects of attention modulation by the hand, and of increased letter spacing to reduce crowding, were also found to modulate the oculomotor dynamics in reading, but whether these factors resulted in benefits or costs depended on characteristics, such as visual attention span, that varied within our sample

    Graphene-based modulation-doped superlattice structures

    The electronic transport properties of graphene-based superlattice structures are investigated. A graphene-based modulation-doped superlattice structure geometry is proposed and consist of periodically arranged alternate layers: InAs/graphene/GaAs/graphene/GaSb. Undoped graphene/GaAs/graphene structure displays relatively high conductance and enhanced mobilities at elevated temperatures unlike modulation-doped superlattice structure more steady and less sensitive to temperature and robust electrical tunable control on the screening length scale. Thermionic current density exhibits enhanced behaviour due to presence of metallic (graphene) mono-layers in superlattice structure. The proposed superlattice structure might become of great use for new types of wide-band energy gap quantum devices.Comment: 5 figure

    Novel markers in pediatric-type follicular lymphoma

    The aim of this study was to review the histopathological, phenotypic, and molecular characteristics of pediatric-type follicular lymphoma (PTFL) and to assess the diagnostic value of novel immunohistochemical markers in distinguishing PTFL from follicular hyperplasia (FH). A total of 13 nodal PTFLs were investigated using immunohistochemistry, fluorescence in situ hybridization (FISH), and PCR and were compared with a further 20 reactive lymph nodes showing FH. Morphologically, PTFL cases exhibited a follicular growth pattern with irregular lymphoid follicles in which the germinal centers were composed of numerous blastoid cells showing a starry-sky appearance. Immunohistochemistry highlighted preserved CD10 (13/13) and BCL6 (13/13) staining, CD20 (13/13) positivity, a K light chain predominance (7/13), and partial BCL2 expression in 6/13 cases (using antibodies 124, E17, and SP66). The germinal center (GC)–associated markers stathmin and LLT-1 were positive in most of the cases (12/13 and 12/13, respectively). Interestingly, FOXP-1 was uniformly positive in PTFL (12/13 cases) in contrast to reactive GCs in FH, where only a few isolated positive cells were observed. FISH revealed no evidence of BCL2, BCL6, or MYC rearrangements in the examined cases. By PCR, clonal immunoglobulin gene rearrangements were detected in 100% of the tested PTFL cases. Our study confirmed the unique morphological and immunophenotypic features of PTFL and suggests that FOXP-1 can represent a novel useful diagnostic marker in the differential diagnosis between PTFL and FH

    Gender plays no role in student ability to perform on computer-based examinations

    BACKGROUND: To see if there is a difference in performance when students switch from traditional paper-and-pencil examinations to computer-based examinations, and to determine whether there are gender differences in student performance in these two examination formats. METHODS: This study involved first year medical students at the University of Illinois at Urbana-Champaign over three Academic Years 2002–03/2003–04 and 2003–05. Comparisons of student performance by overall class and gender were made. Specific comparisons within courses that utilized both the paper-and-pencil and computer formats were analyzed. RESULTS: Overall performance scores for students among the various Academic Years revealed no differences between exams given in the traditional pen-and-paper and computer formats. Further, when we looked specifically for gender differences in performance between these two testing formats, we found none. CONCLUSION: The format for examinations in the courses analyzed does not affect student performance. We find no evidence for gender differences in performance on exams on pen-and-paper or computer-based exams
