    Modulating Anti-MicroRNA-21 Activity and Specificity Using Oligonucleotide Derivatives and Length Optimization

    MicroRNAs are short, endogenous RNAs that direct posttranscriptional regulation of gene expression vital for many developmental and cellular functions. Implicated in the pathogenesis of several human diseases, this group of RNAs provides interesting targets for therapeutic intervention. Anti-microRNA oligonucleotides constitute a class of synthetic antisense oligonucleotides used to interfere with microRNAs. In this study, we investigate the effects of chemical modifications and truncations on activity and specificity of anti-microRNA oligonucleotides targeting microRNA-21. We observed an increased activity but reduced specificity when incorporating locked nucleic acid monomers, whereas the opposite was observed when introducing unlocked nucleic acid monomers. Our data suggest that phosphorothioate anti-microRNA oligonucleotides yield a greater activity than their phosphodiester counterparts and that a moderate truncation of the anti-microRNA oligonucleotide improves specificity without significantly losing activity. These results provide useful insights for design of anti-microRNA oligonucleotides to achieve both high activity as well as efficient mismatch discrimination

    Blood gases, biochemistry and haematology of Galápagos hawksbill turtles (Eretmochelys imbricata)

    The hawksbill turtle, Eretmochelys imbricata, is a marine chelonian with a circum-global distribution, but the species is critically endangered and has nearly vanished from the eastern Pacific. Although reference blood parameter intervals have been published for many chelonian species and populations, including nesting Atlantic hawksbills, no such baseline biochemical and blood gas values have been reported for wild Pacific hawksbill turtles. Blood samples were drawn from eight hawksbill turtles captured in near shore foraging locations within the Galápagos archipelago over a period of four sequential years; three of these turtles were recaptured and sampled on multiple occasions. Of the eight sea turtles sampled, five were immature and of unknown sex, and the other three were females. A portable blood analyzer was used to obtain near immediate field results for a suite of blood gas and chemistry parameters. Values affected by temperature were corrected in two ways: (i) with standard formulas and (ii) with auto-corrections made by the portable analyzer. A bench top blood chemistry analyzer was used to measure a series of biochemistry parameters from plasma. Standard laboratory haematology techniques were employed for red and white blood cell counts and to determine haematocrit manually, which was compared to the haematocrit values generated by the portable analyzer. The values reported in this study provide reference data that may be useful in comparisons among populations and in detecting changes in health status among Galápagos sea turtles. The findings might also be helpful in future efforts to demonstrate associations between specific biochemical parameters and disease or environmental disasters

    Recent trends on glacier area retreat over the group of Nevados Caullaraju - Pastoruri (Cordillera Blanca, Peru) using Landsat imagery

    The Cordillera Blanca, located in the central zone of the Andes Mountains in Peru, has shown a retreat in its glaciers. This paper presents a trend analysis of the glacier area over the groups of Nevados Caullaraju-Pastoruri from 1975 to 2010 using Landsat-5 Thematic Mapper (TM) imagery. In the case of the Nevados Pastoruri/Tuco, the study period was extended back to 1957 by using an aerial photograph taken that year. The extent of clean glacier ice was estimated using Normalized Difference Snow Index (NDSI) thresholds. Moreover, the estimation of debris-covered glacier ice was retrieved by means of a decision tree classification method using NDSI, Normalized Difference Vegetation Index (NDVI) and Land Surface Temperature (LST). Area estimations derived from Landsat imagery were compared to the glacier ground-truth data in 1975 and 2010. Results show a statistically significant (p < 0.05) decreasing trend over the whole study area. Total glacier area decreased at a rate of 4.5 km2 per decade from 1975 to 2010, with a total loss of 22.5 km2 (58%). Lower decreasing rates were found for the period 1987–2010: 3.5 km2 per decade with a total loss of 7.7 km2 (32.5%). In the case of the Nevados Pastoruri/Tuco, decreasing rates of clean ice extent were constant for the periods 1957–2010, 1975–2010 and 1987–2010, with values close to 1.4 km2 per decade and a total loss between 1957 and 2010 estimated at about 5 km2 (54%). This work shows an evident area decrease in the Caullaraju-Pastoruri tropical glaciers, which needs to be included in a future hydrological scenario of local adaptability and water management

    First use of a compound-specific stable isotope (CSSI) technique to trace sediment transport in upland forest catchments of Chile.

    Land degradation is a problem affecting the sustainability of commercial forest plantations. The identification of critical areas prone to erosion can assist this activity to better target soil conservation efforts. Here we present the first use of the carbon-13 signatures of fatty acids (C14 to C24) in soil samples for spatial and temporal tracing of sediment transport in river bodies of upland commercial forest catchments in Chile. This compound-specific stable isotope (CSSI) technique was tested as a fingerprinting approach to determine the degree of soil erosion in pre-harvested forest catchments with surface areas ranging from 12 to 40ha. For soil apportionment a mixing model based on a Bayesian inference framework was used (CSSIAR v.2.0). Approximately four potential sediment sources were used for the calculations of all of the selected catchments. Unpaved forestry roads were shown to be the main source of sediment deposited at the outlet of the catchments (30-75%). Furthermore, sampling along the stream channel demonstrated that sediments were mainly comprised of sediment coming from the unpaved roads in the upper part of the catchments (74-98%). From this it was possible to identify the location and type of primary land use contributing to the sediment delivered at the outlet of the catchments. The derived information will allow management to focus efforts to control or mitigate soil erosion by improving the runoff features of the forest roads. The use of this CSSI technique has a high potential to help forestry managers and decision makers to evaluate and mitigate sources of soil erosion in upland forest catchments. It is important to highlight that this technique can also be a good complement to other soil erosion assessment and geological fingerprinting techniques, especially when attempting to quantify (sediment loads) and differentiate which type of land use most contributes to sediment accumulation

    Plastic contamination of a Galapagos Island (Ecuador) and the relative risks to native marine species

We explored the presence, composition and environmental drivers of plastic contamination across the marine ecosystem at an island scale, investigated uptake in marine invertebrates and designed a systematic priority scoring analysis to identify the most vulnerable vertebrate species. Beach contamination varied by site (macroplastic 0-0.66 items·m-2, microplastics 0-448.8 particles·m-2 or 0-74.6 particles·kg-1), with high plastic accumulation on east-facing beaches that are influenced by the Humboldt Current. Local littering and waste management leakages accounted for just 2% of macroplastic. Microplastics (including anthropogenic cellulosics) were ubiquitous but in low concentrations in benthic sediments (6.7-86.7 particles·kg-1) and surface seawater (0.04-0.89 particles·m-3), with elevated concentrations in the harbour suggesting some local input. Microplastics were present in all seven marine invertebrate species examined, found in 52% of individuals (n = 123) confirming uptake of microplastics in the Galapagos marine food web.     AbstractThe use of endovascular catheters is a routine practice in secondary and tertiary care level hospitals. The short-term use of peripheral catheters has been found to be associated with the risk of nosocomial bacteraemia, resulting in morbidity and mortality. Staphylococcus aureus is mostly associated with peripheral catheter insertion. This Consensus Document has been prepared by a panel of experts of the Spanish Society of Cardiovascular Infections, in cooperation with experts from the Spanish Society of Internal Medicine, Spanish Society of Chemotherapy, and the Spanish Society of Thoracic-Cardiovascular Surgery, and aims to define and establish guidelines for the management of short duration peripheral vascular catheters. The document addresses the indications for insertion, catheter maintenance, registering, diagnosis and treatment of infection, indications for removal, as well as placing an emphasis on continuous education as a drive toward quality. Implementation of these guidelines will allow uniformity in use, thus minimizing the risk of infections and their complications

Se ha evolucionado desde el concepto de remoción completa de caries hacia tratamientos más conservadores como lo es la eliminación parcial de caries, actualmente conocida como remoción selective de tejido cariado, propiciando un escenario favorable para la mantención de la vitalidad pulpar y la permanencia del diente en boca. El objetivo de esta revisión es realizar una actualización de tratamientos para el manejo de caries profunda de dientes permanente de niños y adolescentes apoyados en la evidencia científica que ha sido reportada en la literatura. Se realizó una búsqueda de la literatura desde el año 2010 al 2020 en base de datos electrónicos como: Pubmed, Scielo, Scopus, Web of Science, Cochrane. Dejando en evidencia que las técnicas que remueven parcialmente el tejido infectado reducen significativamente el riesgo exposición pulpar y sintomatología postoperatoria comparados con tratamientos más invasivos, brindando una mayor comodidad al paciente y reduciendo los costos del tratamiento. Cabe destacar que el éxito clínico dependerá del correcto diagnóstico del paciente considerando aspectos importantes como la valoración de riesgo cariogénico y el pronóstico del diente.     A contemporary picture of enterococcal endocarditis

    BACKGROUND: Enterococcal endocarditis (EE) is a growing entity in Western countries. However, quality data from large studies is lacking. OBJECTIVES: The purpose of this study was to describe the characteristics and analyze the prognostic factors of EE in the GAMES cohort. METHODS: This was a post hoc analysis of a prospectively collected cohort of patients from 35 Spanish centers from 2008 to 2016. Characteristics and outcomes of 516 cases of EE were compared with those of 3,308 cases of nonenterococcal endocarditis (NEE). Logistic regression and Cox proportional hazards regression analysis were performed to investigate risk factors for in-hospital and 1-year mortality, as well as relapses. RESULTS: Patients with EE were significantly older; more frequently presented chronic lung disease, chronic heart failure, prior endocarditis, and degenerative valve disease; and had higher median age-adjusted Charlson score. EE more frequently involved the aortic valve and prosthesis (64.3% vs. 46.7%; p < 0.001; and 35.9% vs. 28.9%; p = 0.002, respectively) but less frequently pacemakers/defibrillators (1.5% vs. 10.5%; p < 0.001), and showed higher rates of acute heart failure (45% vs. 38.3%; p = 0.005). Cardiac surgery was less frequently performed in EE (40.7% vs. 45.9%; p = 0.024). No differences in in-hospital and 1-year mortality were found, whereas relapses were significantly higher in EE (3.5% vs. 1.7%; p = 0.035). Increasing Charlson score, LogEuroSCORE, acute heart failure, septic shock, and paravalvular complications were risk factors for mortality, whereas prior endocarditis was protective and persistent bacteremia constituted the sole risk factor for relapse. CONCLUSIONS: Besides other baseline and clinical differences, EE more frequently affects prosthetic valves and less frequently pacemakers/defibrillators. EE presents higher rates of relapse than NEE.     Endodontic regenerative treatment for internal radicular resorption using bio-ceramic material, case report

    The internal resorption of the internal radicular conduct is a process than can be both physiological or pathological, being the osteoclasts, odontoclasts and dentinoclast responsible for said process. 49-year-old female patient, refers orthodontic treatment at age 20, attends a dental check-up due to pain when chewing. Dental organ (DO) #11 was diagnosed with internal root resorption and symptomatic, suppurative apical periodontitis. Treatment started performing an endodontic access and taking a conductometry reading with an apical foramen locator, using a precision hybrid instrumentation technique and applying hypochlorite irrigation, the intra-canal was medicated with chemically pure calcium hydroxide for 7 days. The canal obturation was repaired infiltrating a bio-ceramic material (BIO-C Sealer) followed by the placement of the single cone using a vertical condensation technique
