    Chemical and biological investigations of Delonix regia (Bojer ex Hook.) Raf.

    U radu je opisana izolacija pet sastojaka petroleterske i diklormetanske frakcije metanolnog ekstrakta kore biljke Delonix regia: lupeol (1), epilupeol (2), β-sitosterol (3), stigmasterol (4) i p-metoksibenzaldehid (5). Nadalje, testirano je antimikrobno djelovanje različitih ekstrakata difuzijskom metodom na disku (15 μg mm2). Zone inhibicije za sastojke topljive u petroleteru, tetraklormetanu i diklormetanu bile su 914 mm, 1113 mm, odnosno 920 mm, dok je zona inhibicije standarda kanamicina bila 2025 mm. U biološkom pokusu smrtnosti morskih kozica najveću toksičnost pokazali su spojevi topljivi u tetraklormetanu (LC50 = 0,83 μg mL1), dok je topljivost sastojaka topljivih u petroleteru i diklormetanu bila LC50 14,94, odnosno 3,29 μg mL1, a standarda vinkristin sulfata 0,812 μg mL1. Ovo je prvo izvješće o izolaciji sastojaka, antimikrobnom djelovanju i citotoksičnosti biljke D. regia.In this study five compounds, lupeol (1), epilupeol (2), β-sitosterol (3), stigmasterol (4) and p-methoxybenzaldehyde (5) were isolated from the petroleum ether and dichloromethane fractions of a methanolic extract of the stem bark of Delonix regia. Antimicrobial screening of the different extracts (15 μg mm2) was conducted by disc diffusion method. The zones of inhibition demonstrated by the petroleum ether, carbon tetrachloride and dichloromethane fractions ranged from 914 mm, 1113 mm and 920 mm, respectively, compared to kanamycin standard with the zone of inhibition of 2025 mm. In brine shrimp lethality bioassay, the carbon tetrachloride soluble materials demonstrated the highest toxicity with LC50 of 0.83 μg mL1, while petroleum ether and dichloromethane soluble partitionates of the methanolic extract revealed LC50 of 14.94 and 3.29 μg mL1, respectively, in comparison with standard vincristine sulphate with LC50 of 0.812 μg mL1. This is the first report on compounds separation from D. regia, their antimicrobial activity and cytotoxicity

    Development and optimisation of spironolactone nanoparticles for enhanced dissolution rates and stability

    Stable solid lipid nanoparticles (SLNs) and nanostructured lipid carriers (NLCs) formulations to enhance the dissolution rates of poorly soluble drug spironolactone (SP) were being developed. Probe ultra-sonication method was used to prepare SLNs and NLCs. All NLCs contained stearic acid (solid lipid carrier) and oleic acid (liquid lipid content), whereas, SLNs were prepared and optimised by using the solid lipid only. The particles were characterised in terms of particle size analysis, thermal behaviour, morphology, stability and in vitro release. The zeta sizer data revealed that the increase in the concentration of oleic acid in the formulations reduced the mean particle size and the zeta potential. The increase in concentration of oleic acid from 0 to 30% (w/w) resulted in a higher entrapment efficiency. All nanoparticles were almost spherically shaped with an average particle size of about ∼170 nm. The DSC traces revealed that the presence of oleic acid in the NLC formulations resulted in a shift in the melting endotherms to a higher temperature. This could be attributed to a good long-term stability of the nanoparticles. The stability results showed that the particle size remained smaller in NLC compared to that of SLN formulations after 6 months at various temperatures. The dissolution study showed about a 5.1- to 7.2-fold increase in the release of the drug in 2 h compared to the raw drug. Comparing all nanoparticle formulations indicated that the NLC composition with a ratio of 70:30 (solid:liquid lipid) is the most suitable formulation with desired drug dissolution rates, entrapment efficiency and physical stability

    Towards analytical typologies of plot systems: quantitative profile of five European cities

    The importance of the plot (also referred to as ‘property’) as one of the fundamental elements of urban form is well recognized within the field of urban morphology. Despite the fact that it is often described as the basic element in the pattern of land divisions, which are essential as organizational frameworks for urban form, studies offering comprehensive descriptions and classifications of plot systems are quite scant. The aim of the paper is to introduce a classification of plot systems into typologies based on five European cities, in order to distinguish particular spatial differences and similarities in terms of their plot structure. The proposed typologies are developed using unsupervised k-means cluster analysis based on numeric attributes derived from central theories in urban morphology. The introduced typologies are essentially configurational, allowing collective systematic properties of plot systems to be captured. Numeric attributes include plot differentiation (or plot size), plot frontage and compactness ratio, corresponding to essential qualities of plot systems such as the capacity to carry differences in space, the ability to operate as interface between street and building and providing a framework for evolution of built form over time. All three attributes are translated into configurational measures in order to capture the context of the plot system, rather than the parameters of individual plots. The combination of these deductively defined variables with algorithmically defined classification methods results in seven plot types that can be used to scale up traditional urban morphological analysis to whole city regions and conduct substantial comparison of patterns within, but also between these regions. Further, it also makes it possible to describe commonly recognized plot patterns and discover new ones
