The cytoplasmic A and B domains of mannitol transporter enzymes are well-conserved across bacterial strains. Domain A and B are covalently connected by a flexible linker in Escherichia coli, whereas they are separated in the thermophilic bacterium Thermoanaerobacter Tengcongensis (Tt). In this study, I investigated changes in domain association changes when the domains have evolved with or without their connecting linkers by characterizing the structure and interaction of the Tt mannitol transporter A (TtIIAmtl) and phosphorylated B (phospho-TtIIBmtl) domains. The backbone folds and interface of individual domains were well conserved between the two bacterial strains. However, separated domains showed 28 times higher affinity than the linked ones. The increase in binding affinity mainly resulted from different compositions of interfacial residues, with the general backbone fold of the complex remaining unperturbed. Phosphorylation of the active site loop significantly contributed to the binding affinity by reducing conformational dynamics at the binding interface, and a few key mutations at the interface were sufficient to account for the enhanced binding affinity. Specifically, the complex was stabilized by extended hydrophobic interactions through T542I and P543T and from hydrogen bonds between the hydroxyl group of V557S and the carboxyl backbone group of A386, which enhances the affinity thereby enabling a proper domain association regardless of the presence of the linker. Interestingly, the population of the association state in vivo was estimated to be nearly identical between the non-linked and linked domains. These results suggest that the binding affinity between domains is, modulated in response to changes in the connecting linkers, which would help to maintain the domain association at a functional level regardless of the presence of the linker.1.     The purpose of this study was to analyze the dental consultation of pediatric inpatients to the department of pediatric dentistry in Yonsei University Severance Hospital, and to investigate the change in patterns. In 2017, 268 pediatric patients (384 cases) admitted to the Severance Hospital were referred to the department of pediatric dentistry. The mean age was 6.6 years, and most of the patients were referred from the Department of Pediatric Hematology Oncology and Rehabilitation Medicine. The chief complaints were as follows: oral examination (31%), dental caries (20%), oral pain (10%), tooth mobility (10%), pre-operative evaluation (9%) and others (20%). 41% of the patients received only oral examination without treatment. Dental caries were the most frequent dental diagnosis of the patients. 28% (111 cases) of patients received operative treatments, 22 cases were treated under general anesthesia. Oral health is closely related to systemic diseases, especially for hospitalized patients. Dental consultations should be encouraged for prevention and early appropriate treatments. For this purpose, it is necessary to establish a referral system and perform dental treatment under general anesthesia.ope

    Replacement and inflammatory resorption are serious complications associated with the delayed replantation of avulsed teeth. In this study, we aimed to assess whether deferoxamine (DFO) can suppress inflammation and osteoclastogenesis in vitro and attenuate inflammation and bone resorption in a replanted rat tooth model. Cell viability and inflammation were evaluated in RAW264.7 cells. Osteoclastogenesis was confirmed by tartrate-resistant acid phosphatase staining, reactive oxygen species (ROS) measurement, and quantitative reverse transcriptase-polymerase chain reaction in teeth exposed to different concentrations of DFO. In vivo, molars of 31 six-week-old male Sprague-Dawley rats were extracted and stored in saline (n = 10) or DFO solution (n = 21) before replantation. Micro-computed tomography (micro-CT) imaging and histological analysis were performed to evaluate inflammation and root and alveolar bone resorption. DFO downregulated the genes related to inflammation and osteoclastogenesis. DFO also reduced ROS production and regulated specific pathways. Furthermore, the results of the micro-CT and histological analyses provided evidence of the decrease in inflammation and hard tissue resorption in the DFO group. Overall, these results suggest that DFO reduces inflammation and osteoclastogenesis in a tooth replantation model, and thus, it has to be further investigated as a root surface treatment option for an avulsed tooth.ope

In the past, most of the steel products produced in Korea were consumed by domestic demand. However, the portion of steel export in Korean total steel production has been increased because of the fall in domestic demand and steel supply increase by the Korean steel company. At same time, countries use of trade remedy actions in the steel trade have proliferated globally, and Korea became the most frequently subjected country to AD/CVD investigations since 1990. The proliferated AD initiations toward Korean steel products became one of the main obstacles to Korean steel export. Many literatures analyzed whether the initiation of AD is motivated by other factors such as countries macroeconomic conditions or retaliatory purposes. However, theres limited studies about Korea(Korean steel industry). Therefore, it is important to investigate the determinants of trade remedy actions(especially AD cases) toward Korean steel products. This study examines the determinants of AD initiation to Korean Steel Products using the panel Probit analysis. AD initiation database includes 14 initiation countries against Korea during the year 1995~2011, and target products are limited in steel products(HS 7201~ 7326). For the analysis of countrys income-level effect, high income, upper-middle-income and lower-middle-income dummy variables are used for the analysis. The panel data analysis suggests that although steel price index and AD initiation shows negative relationship, price factor is not a significant determinant in AD initiation. Instead, fall in GDP growth and deterioration in trade specialization index(TSI) in the world steel trade leads to significant increase in AD initiation. Additionally, countries initiate AD investigation more when their steel over-supply increases. In terms of countries income-level analysis, upper-middle-income country group shows strong negative relations with AD initiation whereas hi-income country group has positive relations.
