Recovery and Detection of Artifitial DNA (plasmid) originated from LMO (Living Modified Organisms) from Sea water and Marine Sediment.
To investigate the long term effects of artifitial DNA (plasmid) originated from LMO (living Modified Organisms) on marine environment, whole DNA, dissolved as well as particulated, was isolated from large scale sample (20 liter sea water and 10 gram of marine sediment). Using tangential filtration, ultrafiltration and resin adsorption, a proper protocol for enough DNA sample for PCR and sequencing for LMO marker was constructed. In field works as well as microcosm analysis using a bacterial plasmids (e.g. pTP1) with luminescent lux gene as a marker, vectors routinely used in many molecular biology studies were found to remain in natural marine ecosystem longer than 6 months.2
Comparison Of Expressed Protein Pattern in Muscle and Liver of Flat Fish, Paralichthys olivaceus during Induced Hibernation by Starvation at Different Temperatures.
To investigate the key proteins roled in the hibernation of flat fish and rockfish, this study evaluated the effect of induced hybernation caused by starvation at low temperature on the expressed protein pattern in muscle and liver in Paralichthys olivaceus and Sebastes pachycephalus. A series of 2-D electrophoresis works as well as HPLC analysis were performed with protein samples isolated from the muscle and liver of specimens maintained at 5, 15, and 25 oC sea water bath, with and without food supply. For 20 days of hybernation induction periods, a series of proteins of marked difference in expression level were recognized.2

Purification and Characterization of Extracellular Protease from Psychrotrophic Antarctic Bacteria
저온에서 최적 생육을 보이는 저온성 균주를 남극해양에서 분리하여 생화학적 특성 및 165 rRNA 염기서열로부터 Shewanella sp.에 속하는 균주로 동정하고 Shewanella sp. L93으로명명하였다. 본 균주에서 생산되는 저온성 세포외 단백질 분해 효소(extracellular protease)를 ammonium sulfate precipitation, High-Q column chromatography, 일차 gel permeation chromatography, BioScale Q2 ion exchange chromatography 및 gel permeation chromatography를 통하여 purification fold 19.3, yield 0.7 %로 정제하였고 그 특성을 조사하였다.
A psychrotrophic bacterium was isolated from Antarctic marine sediment and identified as Shewanella sp. species based on the biochemical properties and 16S rRNA sequence, and designated as Shewanella sp. L93. Extracellular protease produced by this strain was purified through ammonium sulfate precipitation, High-Q column chromatography, first gel permeation chromatography, BioScale Q2 ion exchange chromatography and second gel permeation chromatography, and basic properties of this enzyme were investigated.22Nkc
