    Rapidly changing logistics environments such as development of port and hinterland, advent of ULCVs and containerization are causing the life cycle of ports and its facilities to shorten, which gives rise to a need of proper conversion of some port function. Some advanced countries such as Australia, UK and Japan redeveloped their old port areas and are now utilizing them as citizen-friendly waterfront and valuable tourism resources. After the 1997 Asian financial crisis, Korea successfully restarted its drive for economic growth and achieved a USD 20,000 per capita income level. Furthermore, as a five-workday week system sets in, there is an increasingly rising demand of port redevelopment for citizen-friendly recreational waterfront. Conventional piers in old port area of North Port in Busan Port are getting dilapidated and New Port is taking up some cargo handling function that was partly done by the piers in the past. Moreover, people are now demanding another aspect of port such as maritime leisure, tourism and passenger transportation center. All of the facts mentioned above spurred Busan Port Authority to undertake Busan North Port Redevelopment Project. The purpose of this study is to derive the best type of investment method in North Port Redevelopment Project and prioritize evaluation item in evaluation detail list.     DoctorSynaptic vesicle fusion is a key process for the communication between neuron cells. For fast and precise communication at synapses, the synaptic vesicles fusion should be highly regulated. To do this, SNARE proteins and their regulatory proteins interact actively and cooperatively during fusion processes. Controllable vesicle system and measurements are required to understand this complex fusion processes. An emerging technique, so called single-molecule fluorescence, in biophysics facilitates measurement of the dynamic and static heterogeneity of biomolecules.In this thesis, single-vesicle measurement in solution was achieved for studying SNARE-mediated vesicle fusion, neurotoxicity of ฮฑ-synculein oligomers and fast millisecond dynamics of vesicle tethered single-molecule. Firstly, a single-molecule technique, so called Alternating-laser excitation (ALEX), was applied for studying synaptic vesicle docking and fusion. We demonstrated that ALEX technique can simultaneously measure the docking and fusion of individual vesicles in a solution. Using ALEX, we found that synaptoatagmin-1(Syt1) that exists in the synaptic vesicle can act as a docking factor. This Syt1-mediated vesicle docking necessarily requires PIP2 and t-SNARE interactions, which results in tremendous increment in the rate of vesicle docking compared with the SNARE-mediated docking. This enhancement in docking rate by Syt1 also increases the overall fusion rate. This find that Syt1 helps vesicle docking but does not inhibit SNARE assembly for fusion. Following the Syt1-mediated vesicle docking, the replacement of Syt1 by v-SNARE, synaptobrevin/VAMP-2, and rebinding of Syt1 by the addition of Ca2+ were sequentially observed. On the basis of these sequential actions of Syt1 on SNAREs, we suggested a novel model for synaptic vesicle fusion.Secondly, the neurotoxicity of ฮฑ-synculein oligomers on synaptic vesicle fusion has been investigated by single-vesicle assay. ฮฑ-synculein is known as the pathogenic molecule for Parkinsonโ€™s disease. For the first time, the inhibition effect of ฮฑ-synculein oligomers on the vesicle fusion has been reported in this work. Specifically, ฮฑ-synculein oligomer inhibits SNARE-mediated vesicle docking, which is caused by its ability to interact with v-SNARE.Lastly, a single-vesicle technique for measuring fast single-molecule dynamics on a millisecond time scale has been investigated. Diffusion-based single-molecule FRET has been limited by the short duration of observation time. By tethering, single-molecule of interest to vesicles, a huge increase in observation time was achieved.
