    Autism spectrum disorder (ASD) is a kind of pervasive developmental disorder, and characterized by three main behavioral features including social deficits, impaired communication, and stereotyped and repetitive behaviors. The brain of ASD patients has been characterized with specific features including macrocephaly, imbalanced signal between excitation and inhibition, synaptic abnormalities and so on. Among these features, imbalanced excitatory/inhibitory signal in the autistic brain has been recently magnified as a main cause of ASD symptoms. However, mechanism governing the increased excitatory synaptic activity is unclear yet. Because excitatory/inhibitory imbalance in brain can be induced by aberration in glutamatergic/GABAergic neuronal differentiation, the mechanism of excess excitatory neuronal differentiation was investigated in an animal model of ASD induced by prenatal valproate (VPA) exposure, which may produce symptoms like autistic disorders including skeletal malformation, neural tube defects, and several behavioral abnormalities both in human and rodents. Although exposure to VPA at E9.5 showed strong reproductive toxicity, VPA exposure at E12 showed no major reproductive toxicity but most significant changes in behaviors over control animals with reduced sociability and social preference as well as increased sensitivity to electroshock convulsion. In addition to the altered social behavior, VPA exposure induced increased glutamatergic neuronal differentiation in postnatal brains as exemplified by immunostaining and Western blot using specific marker proteins including PSD-95. Similar finding was observed with primary neural progenitor cell culture treated with VPA in vitro. Expression of transcription factors, which has been implicated in glutamatergic neuronal differentiation, i.e., Pax6, Ngn2, and NeuroD1 was increased in VPA-exposed embryonic brains as well as neural progenitor culture in vitro. When we silenced the expression of Pax6 using siRNA transfection in cultured rat primary neural progenitor cells, VPA induced neither expression of Ngn2 and NeuroD1 nor glutamatergic hyper-differentiation suggesting the essential role of Pax6 induction in this process. The expression of telomerase reverse transcriptase (TERT), that elongates telomere and recently known as a transcription factor interacting with Brg1, was increased by VPA. The binding of TERT protein on Pax6 gene promoter was demonstrated by chromatin immunoprecipitation using anti-TERT antibody, which suggests the possible role of TERT in inducing Pax6 expression. Silence of TERT expression using siRNA transfection in vitro inhibited VPA-induced expression of Pax6, Ngn2 and NeuroD1 as well as excess glutamatergic differentiation. Over-expression of TERT protein using hTERT-GFP plasmid transfection in vitro activated glutamatergic neuronal differentiation by inducing expression of Pax6, Ngn2 and NeuroD1. These results suggest that glutamatergic neuronal differentiation is increased in the VPA-induced autistic animal model and TERT over-expression induces sequential increase of Pax6, Ngn2 and NeuroD1 leading to the increased excitatory neuronal maturation.
