    Optimization of Microwave-Assisted Extraction Technology for Pingtang Capsules B by Orthogonal Test

    目的:优选平糖胶囊b方的微波辅助提取工艺。方法:采用微波辅助提取平糖胶囊b方,以栀子苷提取量为检测指标,选择微波功率、料液比、提取时间、提取次数为考察指标,通过正交试验优选微波辅助提取工艺。采用HPlC测定栀子苷含量,色谱条件为dIAMOnSIlTMrP C18色谱柱(4.6 MMx250 MM,5μM),流动相乙腈-水(11∶89),流速1.0 Ml·MIn-1,检测波长238 nM,柱温室温,进样量20μl。结果:最佳工艺条件为微波功率15 kW,料液比1∶5,提取时间20 MIn,提取数2次,栀子苷平均提取量9.08 Mg·g-1。结论:优选的微波辅助提取工艺具有高效、节能、省时等优点,具有极大的工业化推广价值。Objective: To optimize microwave-assisted extraction technology of Pingtang capsules B.Method: Pingtang capsules B was extracted by microwave assisted technology,with yield of geniposide as index,choosing microwave power, material-liquid ratio, extraction time, extraction times as investigating factors,orthogonal test was adopted to optimize microwave assisted extraction technology.The content of geniposide was determined by HPLC,chromatographic conditions were as follows: DiamonsilTMRP C18column(4.6 mm × 250mm,5 μm),mobile phase of acetonitrile-water(11∶ 89),flow rate 1.0 mL·min- 1,detection wavelength 238nm,column temperature at room temperature,injection volume 20 μL.Result: Optimum extraction technology conditions were as follows: microwave power 15 kW,solid-liquid rate 1∶ 5,extraction time 20 min,extraction 2times; Under these conditions,average yield of geniposide was 9.08 mg·g- 1.Conclusion: Optimized microwave assisted extraction technology had advantages of high efficiency, energy saving and time saving with great application value for industrial production of Pingtang capsules B.厦门市科技局科研项目(3502Z20044013

    临床药师及用药软件对门诊处方审核结果分析

    目的了解处方合理用药软件及临床药师对门诊处方审核情况,为优化处方合理用药软件提供参考。方法收集我院2017年7—12月门诊处方合理用药软件及临床药师对门诊处方人工审核的监测数据,并对有关数据进行统计分析。结果我院2017年7—12月处方总量为1 127 455份,其中用药提示474 618份,占42.1%。其中,由临床药师人工审核通过共30 133份,返回修改的1 072份;系统自动拦截共1 734份。结论处方合理用药软件同临床药师人工审方相结合能大幅提高调剂药师工作效率,进一步提升为患者服务的质量

    肚液散GC指纹图谱建立及60Co-γ辐照灭菌对4种成分的影响

    目的建立肚液散(甘草、麦芽、鸡内金等)GC指纹图谱,并考察60Co-γ辐照灭菌对乙酸龙脑酯、桂皮醛、丁香酚、肉豆蔻醚含有量的影响。方法该药物乙酸乙酯提取液的分析采用HP-5毛细管柱(0.32 mm×30 m, 0.25μm);进样口温度240℃;FID检测器温度260℃。结果 13批样品指纹图谱中有13个共有峰,相似度大于0.930。4种成分在各自范围内线性关系良好(r≥0.999 7),平均加样回收率93.62%~107.1%,RSD 2.3%~3.2%。辐照灭菌后,桂皮醛、丁香酚、肉豆蔻醚含有量无明显变化(P>0.05);乙酸龙脑酯含有量在4.0 kGy下较稳定,在8.0 kGy下显著降低(P<0.05)。结论 60Co-γ辐照灭菌对肚液散挥发油成分稳定性有一定影响,故在保证灭菌效果前提下应选择低辐照剂量

    重组戊型肝炎病毒衣壳蛋白工程菌的高密度培养

    在 10L发酵罐中对戊型肝炎病毒衣壳蛋白在重组大肠杆菌中表达发酵工艺进行了研究 ,用分批培养方法探讨了不同培养基、培养基中磷酸盐浓度和Mg2 + 浓度等因素对菌体生长与重组蛋白表达的影响 ;用分批补料培养研究了不同的补料工艺对菌体生长与重组蛋白表达的影响 ,同时对重组菌诱导时期、诱导持续时间以及不同诱导温度表达包含体在尿素溶液中的溶解性进行了研究。结果表明 ,在优化后的培养基中 ,磷酸盐浓度、Mg2 + 浓度分别为 80mmol L与 2 0mmol L时菌体生长与表达效果较好 ;分批补料培养中 ,37℃培养 9h菌体达到对数期中期 (约 4 5OD6 0 0 )为适宜诱导时期 ,加入终浓度为 1 0mmol LIPTG后诱导 5h ,OD6 0 0 达到 80以上 ,重组蛋白表达量达到2 9 74 % ,为最适收获菌体时间 ;37℃表达的包含体 80 %以上溶解在 4mol L的尿素溶液中 ,最终浓度达到 14mg mL ;10L发酵罐中确定的发酵工艺参数在 30L发酵罐中进行了放大培养 ,10L发酵罐中确定的发酵工艺参数在 30L发酵罐上具有可放大性与重复性 ,可以应用于工业生产

    表面改性金属材料双极板前处理的研究

    在不锈钢表面制备一层导电性好、耐腐蚀的薄膜。是获得高性能质子交换膜燃料电池双极板材料的有效方法。实验分别采用抛光、喷砂和拉砂三种方法对不锈钢基体进行了前处理,然后利用物理气相沉积技术在不锈钢表面制备了TiN膜。观察了双极板材料的微观结构。并对其导电性和耐蚀性进行评价,以研究基体前处理对性能的影响。经过不同的前处理,双极板材料表面呈现出完全不同的微观结构。双极板材料的界面导电性与有效接触面积密切相关。当与碳纸接触时,抛光前处理的双极板材料导电性最好;当与膨胀石墨接触时,喷砂前处理的双极板材料电阻最低。在模拟电池腐蚀环境下0.5mol/L H2SO4+2×10^-6F^-的电化学腐蚀结果显示。喷砂前处理的双极板材料具有最好的耐腐蚀能力

    海洋微生物是当前抗肿瘤药物开发的重要新资源.应用硅胶柱层析结合活性跟踪分离的方法,从海洋放线菌菌株N350的脂溶性发酵产物中纯化并鉴定了4个化合物,分别为反式-13-二十二碳单烯酰胺,7,4'-二羟基异黄酮、6-甲氧基-7,4'-二羟基异黄酮及β-胡萝卜甙.其中,反式-13-二十二碳单烯酰胺、7,4'-二羟基异黄酮及6-甲氧基-7,4'-二羟基异黄酮具有抗肿瘤活性,抑制人口腔上皮癌KB细胞生长的ID50分别为15,8 1,5 2μg/cm3,抑制人白血病细胞HL-60生长的ID50分别为7,2 8,1 9μg/cm3.这些化合物均首次在海洋放线菌中得到分离.Marine microorganism, which can be easily reproduced in large scale in industry, is being exploited for new antitumor drugs with unique structure and excellent bioactivity. Four lipophilic cytotoxic compounds produced by a marine actinomycetes, strain N350, were purified by bioassay-guided isolation, and identified as cis-13-docosenmamide, 7, 4′-dihydroxyisoflavone, 6-methoxy-7, 4′-dihydroxyisoflavone, and β-sitosterol-β-D-glucopyranoside, respectively. Cis-13-docosenamide, 7, 4′-dihydroxyisoflavone and 6-methoxy-7, 4′-dihydroxyisoflavone demonstrated significant cytotoxic activity in vitro. Against human leukemia cell line, HL-60, their IC_(50) was 2.8, 1.9, 7 μg/cm~3, respectively, and against human epidermal carcinoma of oral cavity, KB cell line, IC_(50) was 8.1, 5.2, 15 μg/cm~3, respectively.     A new lactone (1), named musellactone, was isolated from the aerial parts of Musella lasiocarpa, along with eight known compounds. The structures were chemically characterized by spectroscopic methods. Musellactone (1), and 2-methyl-9-(4'-hydroxyphenyl)-phenalen-1-one (2), showed different effects against five bacteria strains, and 1,2,3,4-tetrahydro-1,2-dimethyl-6,7-isoquinolinediol (3), showed significant cytotoxic activities in vitro against KB, HL-60 and BEL-7404 human carcinoma cell lines
