酵素結合免疫吸附法(ELISA) 應用於新城雞瘟血清抗體之測定及單簇群抗體融合瘤之篩選
經16%PEG -6○○○沈澱,及連續蔗糖層高速離心而純化的新城雞瘟石井病毒株
E -3,為間接酵素結合免疫吸附法(ELISA )的抗原,進行測定血清中ELISA 力價
及篩選分泌特異性單簇群抗體的融合瘤。
經血清採用標準系列稀釋法(standard serial dilution)及其吸光度採用扣除法(
subtraction
method)修正後,在單一血清稀釋倍數(16○倍)時,任一血清吸光度修正值與其
ELISA 觀測血清力價(observed serum titers )值,在雙對數座標下,有近乎直線
的關係。經迴歸分析,可得一標準直線及迴歸方程式。藉此,任一稀釋16○倍的血
清,由其吸光度修正值即可預估其ELISA 抗體力價及訂出ELISA 保護力價的標準值。
與血球凝集抑制(Hemagglutination-Inhibition;HI)力價比較,ELISA 的敏感性
(sensitivity )為1○○%(135/135)而HI試驗僅為61.6%(135
/221)。兩者之間有極顯著差異(P <○.○1),其一致性(agreement )為
63.25%(148/234);ELISA 力價至少比HI力價高64倍以上,因此,
ELISA 比HI試驗較易測出低力價的抗體。
同時對融合瘤試驗進行篩選:已有18個多簇群呈ELISA 陽性反應,其中14群亦呈
HI試驗陽性反應。陽性多簇群業經limiting dilution 嘗試單簇群化(monocloniza-
tion)後,已獲9個ELISA 陽性單簇群。進一步的觀察與試驗仍在進行
水禽及火雞類禽流感監控
Sera samples will be collected seasonally from all over the country. Avian influenza antibody of ELISA, agar gel precipitation (AGP), and hemagglutination inhibition (HI) test. Fecal samples will be collected from the antibody positive flocks, and then subjected to RT-PCR for AIV detection and subserotype determination. Domestic turkeys will detects for serological antiboies to avian influenza virus in Taiwan, sampling 20 from each group or farm to testing with enzyme-linked immunosorbent assay (ELISA). The antibody prevalence will be analyzing as a data for prevention of AI.
