    Guz Krukenberga – problem interdyscyplinarny – opis przypadku

    Metastatic lesions within the ovary constitute a serious diagnostic problem in daily practice. We present an interesting case of Krukenberg tumor in a woman 13 years after partial gastrectomy due to stomach cancer. Our case confirms that every woman with history of cancer should remain under gynecological control. It is important due to a high risk of metastatic changes localized in the ovaries, regardless of the time elapsed since the diagnosis of the primary tumor.Ogniska przerzutowe w obrębie jajnika stanowią trudny interdyscyplinarny problem diagnostyczny. Część zmian wykrywana jest przypadkowo, a brak pełnych danych klinicznych może utrudnić rozpoznanie zarówno radiologowi analizującemu badania obrazowe jak również patomorfologowi oceniającemu materiał histopatologiczny. W pracy przedstawiono kazuistyczny przypadek pacjentki z guzem Krukenberga, rozpoznanym trzynaście lat po częściowej gastrektomii z powodu raka gruczołowego żołądka. Potwierdza on konieczność okresowej kontroli ginekologicznej pacjentek z przeszłością nowotworową ze względu na wysokie ryzyko wystąpienia zmian przerzutowych w obrębie jajników, niezależnie od okresu jaki upłynął od rozpoznania nowotworu pierwotnego

    Ultrasensitive electrochemical genosensor for direct detection of specific RNA sequences derived from avian influenza viruses present in biological samples

    An electrochemical genosensor based on an epoxyphenanthroline–Fe(III)–NH2-ssDNA layer for the detection of RNA derived from Avian Influenza is presented. The biosensor preparation consists of: (I) modification of gold electrodes with aminoethanethiol, (II) modification of the self-assembled monolayer of aminoethanethiol with 5,6-epoxy-5,6-dihydro-[1,10]-phenanthroline using “click” chemistry, (III) a first step of complexation of Fe(III) by 5,6-epoxy-5,6-dihydro-[1,10]-phenanthroline, (IV) a second step of complexation of Fe(III) by 5,6-epoxy-5,6-dihydro-[1,10]-phenanthroline, (V) immobilization of the single stranded amino-DNA probe via “click” chemistry between epoxy and amino groups. The interactions between the ssDNA probe and RNA targets were explored with Osteryoung Square Wave Voltammetry. The genosensor showed a remarkable detection limit of 3 copies/μL (5 aM) for RNA extracted from A/swan/Poland/305/06 (H5N1) containing a fully complementary sequence. A linear dynamic range for this sequence was observed from 3.0×103 to 3.0×105 [copies/μl]. RNA extracted from A/mallard/Poland/446/09 (H7N7), containing a non-complementary sequence, generated a much weaker response. Moreover, the developed genosensor allows to distinguish RNA present in biological samples having 2, 3 and 4 mismatches. This biosensing approach can become a potential alternative tool for detecting RNA samples in biomedical research and early clinical diagnosis of avian influenza viruses

    Bornaviruses are a diverse family of viruses infecting various hosts, including birds. Aquatic bird bornavirus 1 (ABBV-1) and aquatic bird bornavirus 2 (ABBV-2) have been found in wild waterfowl but data on their prevalence are scarce. To gain knowledge on the occurrence of ABBVs in Poland, samples originating from dead birds of the Anseriformes order collected in 2016–2021 were tested with a real time RT-PCR method targeting the ABBVs genome. A total of 514 birds were examined, including 401 swans, 96 ducks and 17 geese. The presence of ABBV-1 RNA was detected in 52 swans (10.1% of all tested birds) from 40 different locations. No positive results were obtained for ducks and geese. Sequences of about 2300 bases were generated for 18 viruses and phylogenetic analysis was performed. A relatively low genetic diversity of the examined ABBV-1 strains was observed as all were gathered in a single cluster in the phylogenetic tree and the minimum nucleotide identity was 99.14%. The Polish strains were closely related to ABBV-1 identified previously in Denmark and Germany, but a limited number of sequences from Europe hinders the drawing of conclusions about interconnections between Polish and other European ABBVs. The results of the present study provide new insights into the distribution and genetic characteristics of ABBVs in wild birds in Europe

    Avian influenza virus (AIV) is a highly diverse and widespread poultry pathogen. Its evolution and adaptation may be affected by multiple host and ecological factors, which are still poorly understood. In the present study, a turkey-origin H9N2 AIV was used as a model to investigate the within-host diversity of the virus in turkeys, quail and ducks in conjunction with the clinical course, shedding and seroconversion. Ten birds were inoculated oculonasally with a dose of 106 EID50 of the virus and monitored for 14 days. Virus shedding, transmission and seroconversion were evaluated, and swabs collected at selected time-points were characterized in deep sequencing to assess virus diversity. In general, the virus showed low pathogenicity for the examined bird species, but differences in shedding patterns, seroconversion and clinical outcome were noted. The highest heterogeneity of the virus population as measured by the number of single nucleotide polymorphisms and Shannon entropy was found in oropharyngeal swabs from quail, followed by turkeys and ducks. This suggests a strong bottleneck was imposed on the virus during replication in ducks, which can be explained by its poor adaptation and stronger selection pressure in waterfowl. The high within-host virus diversity in quail with high level of respiratory shedding and asymptomatic course of infection may contribute to our understanding of the role of quail as an intermediate host for adaptation of AIV to other species of poultry. In contrast, low virus complexity was observed in cloacal swabs, mainly from turkeys, showing that the within-host diversity may vary between different replication sites. Consequences of these observations on the virus evolution and adaptation require further investigation

    Porównanie cystouretrografii mikcyjnej i sonocystografii mikcyjnej z użyciem ultrasonograficznego środka kontrastującego drugiej generacji w badaniu prospektywnym

    Background: The invasiveness and exposure to radiation in voiding cystourethrography led to the introduction of alternative methods of diagnosis of vesicoureteral reflux, including contrast enhanced voiding urosonography. While there is a limited number of studies comparing these methods using new generation ultrasound contrast agents, none of them compared both methods simultaneously. This study is aimed at assessing agreement between contrast enhanced voiding urosonography with second-generation ultrasound contrast agents and voiding cystourethrography. Methods: From April 2013 to May 2014, 83 children (37 female and 46 male), mean age 3.5 years, age range from 1 month to 17.5 years, underwent prospective simultaneous assessment by contrast enhanced voiding urosonography and voiding cystourethrography, with a total of 166 uretero-renal units evaluated. Results: The sensitivity of voiding cystourethrography and contrast enhanced voiding urosonography were comparable, amounting to 88%, however, neither reached 100% for the entire studied population. The negative predictive value of voiding urosonography and voiding cystourethrography was 97%, and there was no difference between both methods. Conclusion: Voiding cystourethrography and contrast enhanced voiding urosonography are comparable methods in diagnosis of vesicoureteral reflux, and can be performed alternatively. However, some limitations of contrast enhanced voiding urosonography must be remembered.Wprowadzenie: Negatywne skutki promieniowania jonizującego wykorzystywanego do badań cystouretrografii mikcyjnej wymuszają poszukiwanie alternatywnych metod diagnostycznych. Takim badaniem może być sonocystografia mikcyjna z użyciem ultrasonograficznych środków kontrastujących. Liczba prac porównujących klasyczne badanie cystouretrografii mikcyjnej i badanie ultrasonograficzne z użyciem środków kontrastujących drugiej generacji jest jednak niewielka. Dotąd nie opublikowano pracy, która porównałaby jednoczasowo obie metody w diagnostyce odpływów pęcherzowo-moczowodowych. Prezentowana praca ma na celu ocenę zgodności między badaniem ultrasonograficznym z użyciem środków kontrastujących drugiej generacji i badaniem cystouretrografii mikcyjnej. Materiał i metody: W okresie od kwietnia 2013 do maja 2014 roku jednoczasowe badanie ultrasonograficzne z użyciem środków kontrastujących drugiej generacji i cystouretrografii mikcyjnej wykonano u 83 dzieci (37 dziewczynek i 46 chłopców, średnia wieku 3,5 roku, zakres wieku od 1 miesiąca do 17,5 roku). Prospektywnej ocenie zostało poddanych 166 jednostek moczowodowo-nerkowych. Wyniki: W badaniu wykazano podobną czułość obu metod diagnostycznych w diagnostyce odpływów pęcherzowo- moczowodowych, na poziomie 88%, żadna z metod nie osiągnęła wyniku 100%. Ujemna wartość predykcyjna dla obu metod wyniosła 97% i nie wykazano żadnej istotnej różnicy pomiędzy obiema metodami w zakresie diagnostyki odpływów pęcherzowo-moczowodowych. Wniosek: Obie metody wykazały podobną czułość w ocenie odpływów pęcherzowo-moczowodowych. Należy jednak pamiętać o możliwych ograniczeniach badania ultrasonograficznego z użyciem środków kontrastujących drugiej generacji w diagnostyce fazy mikcyjnej u niespokojnych, płaczących w trakcie badania dzieci
