    Exposure to aluminium during gestation causes changes in mammalian brain development and behavior. Our aim was to perform spectral analysis on electrocortical activity of Sprague Dawley male pups (30Ā±3 days of age) whose mothers were treated with aluminium during gestation and lactation. There was a higher presence of power spectra in the delta range of parietal electrocortical activity, a lower presence in the theta range and increased values of the parameter DT as the ratio of delta to theta range in pups indirectly exposed to aluminium (whose mothers were drinking a 0.5% water solution of aluminium chloride during the gestation and lactation periods), compared to controls.Izlaganje dejstvu aluminijuma tokom gestacije i laktacije može dovesti do promena u razviću mozga i ponaÅ”anja kod sisara. NaÅ” cilj je bio da se uradi spektralna analiza elektrokortikalne aktivnosti mladih Sprague Dawley pacova muÅ”kog pola (starosti 30Ā±3 dana) čije su majke bile tretirane aluminijumom tokom gestacije i laktacije. U poređenju sa kontrolom, kod mladih pacova koji su bili indirektno trovani aluminijumom (čije su majke pile 0,5 % vodeni rastvor AlCl3 tokom gestacionog i laktacionog perioda) postoji veća zastupljenost delta ritma aktivnosti kore velikog mozga, manje prisustvo teta ritma, kao i porast parametra DT kao odnos između delta i teta opsega.Projekat ministarstva br. 14302

    Dynamic and Assembly Characteristics of Deep-Cavity Basket Acting as a Host for Inclusion Complexation of Mitoxantrone in Biotic and Abiotic Systems

    We describe the preparation, dynamic, assembly characteristics of vase-shaped basket 13āˆ’ along with its ability to form an inclusion complex with anticancer drug mitoxantrone in abiotic and biotic systems. This novel cavitand has a deep nonpolar pocket consisting of three naphthalimide sides fused to a bicyclic platform at the bottom while carrying polar glycines at the top. The results of 1H Nuclear Magnetic Resonance (NMR), 1H NMR Chemical Exchange Saturation Transfer (CEST), Calorimetry, Hybrid Replica Exchange Molecular Dynamics (REMD), and Microcrystal Electron Diffraction (MicroED) measurements are in line with 1 forming dimer [12]6āˆ’, to be in equilibrium with monomers 1(R)3āˆ’ (relaxed) and 1(S)3āˆ’ (squeezed). Through simultaneous line-shape analysis of 1H NMR data, kinetic and thermodynamic parameters characterizing these equilibria were quantified. Basket 1(R)3āˆ’ includes anticancer drug mitoxantrone (MTO2+) in its pocket to give stable binary complex [MTOāŠ‚1]āˆ’ (Kd=2.1 Ī¼M) that can be precipitated in vitro with UV light or pH as stimuli. Both in vitro and in vivo studies showed that the basket is nontoxic, while at a higher proportion with respect to MTO it reduced its cytotoxicity in vitro. With well-characterized internal dynamics and dimerization, the ability to include mitoxantrone, and biocompatibility, the stage is set to develop sequestering agents from deep-cavity baskets

    Antihyperlipidemic potential of dietary supplementation with carnosine in high-fat diet-fed rats

    OBJECTIVE: The aim of this study was to investigate the hypolipidemic effects of carnosine and a commercial carnosine supplement on lipid status, liver and kidney function, and inflammation associated with dyslipidemia in rats with high-fat diet-induced hyperlipidemia. MATERIALS AND METHODS: The study was conducted on adult male Wistar rats, divided into control and experimental groups. Animals were kept in standard laboratory conditions and according to groups were treated with saline, carnosine, carnosine dietary supplement, simvastatin, and their combinations. All substances were prepared fresh every day and used by oral gavage. RESULTS: Treatment with a carnosine-based supplement significantly improved total and LDL cholesterol levels in serum, especially in the combination with simvastatin as a conventional drug in dyslipidemia treatment. The effect of carnosine on the metabolism of triglycerides was not as evident as in the case of cholesterol. Nevertheless, the values of the atherogenic index showed that the combinations of carnosine and carnosine supplement with simvastatin were the most effective in lowering this comprehensive lipid index. Dietary carnosine supplementation resulted also in anti-inflammatory effects, as demonstrated by immunohistochemical analyses. Besides, the good safety profile of carnosine in terms of its effect on liver and kidney functions was also confirmed. CONCLUSIONS: The use of carnosine supplements in preventing and/or treatment of metabolic disorders requires further investigations into the mechanisms of action and potential interactions with conventional therapy
