The yhiM gene codes for an inner membrane protein involved in GABA export in Escherichia coli
In order to survive the exposure to acid pH, Escherichia coli activates molecular circuits leading from acid tolerance to extreme acid resistance (AR). The activation of the different circuits involves several global and specific regulators affecting the expression of membrane, periplasmic and cytosolic proteins acting at different levels to dampen the harmful consequences of the uncontrolled entry of protons intracellularly. Many genes coding for the structural components of the AR circuits (protecting from pH ≤ 2.5) and their specific transcriptional regulators cluster in a genomic region named AFI (acid fitness island) and respond in the same way to global regulators (such as RpoS and H-NS) as well as to anaerobiosis, alkaline, cold and respiratory stresses, in addition to the acid stress. Notably some genes coding for structural components of AR, though similarly regulated, are non-AFI localised. Amongst these the gadBC operon, coding for the major structural components of the glutamate-based AR system, and the ybaS gene, coding for a glutaminase required for the glutamine-based AR system. The yhiM gene, a non-AFI gene, appears to belong to this group. We mapped the transcription start of the 1.1 kb monocistronic yhiM transcript: it is an adenine residue located 22 nt upstream a GTG start codon. By real-time PCR we show that GadE and GadX equally affect the expression of yhiM under oxidative growth conditions. While YhiM is partially involved in the RpoS-dependent AR, we failed to detect a significant involvement in the glutamate- or glutamine-dependent AR at pH ≤ 2.5. However, when grown in EG at pH 5.0, the yhiM mutant displays impaired GABA export, whereas when YhiM is overexpressed, an increases of GABA export in EG medium in the pH range 2.5–5.5 is observed. Our data suggest that YhiM is a GABA transporter with a physiological role more relevant at mildly acidic pH, but not a key component of AR at pH < 2.5
Work-related teaching and learning methods to foster generic skills in Higher Education. An Italian experience
open5siWithin the framework of modernisation of higher education systems in Europe, universities are invited to go beyond a knowledge-based perspective focused on disciplinary approaches and to be more concentrated on encouraging generic skills to deal with today’s complex and unpredictable career paths. The literature about Work-Related Learning and Work-Integrated Learning offers evidence to research regarding contributions of work-related experiences to the development of generic skills. The first part of the article presents a literature review carried out following the matching among three main keywords: work-related learning, generic skills, and higher education. Resources focused on the integration/teaching of generic skills in formal curriculum or in co-curriculum work-related activities and they were collected in order to explore the link between work-related learning in higher education and the development of generic skills. The focus is to identify valuable considerations to improve teaching strategies and methods. The second part presents an Italian work-related experience developed within the course of “Organizational Intervention Research Methods,” which involved 22 master’s degree students. Ultrasonic and Densitometric Characterization of Hyaluronan and its Interaction with Surfactant
This disertation thesis is focused on the study of physico-chemical interactions of hyaluronan (with molecular weights from 10 to 1750 kDa) with cationic surfactants measured using uncommon technique named high resolution ultrasonic spectroscopy. Densitometer was also used for the study of these interactions, in measuring of density and ultrasonic velocity of hyaluronan with different molecular weight in dependence on elevated temperature (25 50 °C). The aim is the determination of critical micelle concentration (CMC) and critical aggregation concentration (CAC) of the suractants in the absence and in the presence of hyaluronan with various molecular weights. Interactions in this system are important for the design of the systems for the targeted delivery, especially for the drugs. The experiments were made in water and sodium chloride solution. The significant breakpoint in the ultrasonic velocity showed changes in the system hyaluronan-surfactant.Tato disertační práce se zaměřuje na fyzikálně-chemické interakce hyaluronanu (molekulové hmotností od 10 do 1750 kDa) s kationickými tenzidy. Pro zkoumání a měření vzájemného působení byla použita technika s názvem ultrazvuková spektroskopie s vysokým rozlišením (HR-US). Při zkoumání interakcí byl též použit denzitometr, a to při měření hustoty a uktrazvukové rychlosti hyaluronanu o různé molekulové hmotnosti v závislosti na vybrané teplotě (25-50 °C). Práce se zaměřuje se na studium kritické micelární (CMC) a agregační (CAC) koncentrace tenzidů v přítomnosti a nepřítomnosti hyaluronanu o různé molekulové hmotnosti. Interakce hyaluronanu s kationickými tenzidy jsou důležité pro systémy s cíleným transportem, zejména léčiv. Měření interakcí bylo prováděno ve vodě a v roztoku chloridu sodného. V získaných datech lze pozorovat významné zlomy v ultrazvukové rychlosti, které nám ukazují změny v systému hyaluronan-tenzid.
Framing audience prefigurations of The Hobbit: An Unexpected Journey: The roles of fandom, politics and idealised intertexts
Audiences for blockbuster event-film sequels and adaptations often formulate highly developed expectations, motivations, understandings and opinions well before the films are released. A range of intertextual and paratextual influences inform these audience prefigurations, and are believed to frame subsequent audience engagement and response. In our study of prefigurative engagements with Peter Jackson’s 2012 film, The Hobbit: An Unexpected Journey, we used Q methodology to identify five distinct subjective orientations within the film’s global audience. As this paper illustrates, each group privileges a different set of extratextual referents – notably J.R.R. Tolkien’s original novels, Peter Jackson’s The Lord of The Rings film trilogy, highly localised political debates relating to the film’s production, and the previous associations of the film’s various stars. These interpretive frames, we suggest, competed for ascendancy within public and private discourse in the lead up to The Hobbit’s international debut, effectively fragmenting and indeed polarising the film’s prospective global audience
Control, Process Facilitation, and Requirements Change in Offshore Requirements Analysis: The Provider Perspective
Process, technology, and project factors have been increasingly driving organizations to offshore early software development phases, such as requirements analysis. This emerging trend necessitates greater control and process facilitation between client and vendor sites. The effectiveness of control and facilitation has, however, not been examined within the context of requirements analysis and change. In this study, we examine the role of control and facilitation in managing changing requirements and on success of requirements gathering in the Indian offshore software development environment. Firms found that control by client-site coordinators had a positive impact on requirements analysis success while vender site-coordinators did not have similar influence. Process facilitation by client site-coordinators affected requirements phase success indirectly through control. The study concludes with recommendations for research and practice
Method for finding metabolic properties based on the general growth law. Liver examples. A General framework for biological modeling
We propose a method for finding metabolic parameters of cells, organs and
whole organisms, which is based on the earlier discovered general growth law.
Based on the obtained results and analysis of available biological models, we
propose a general framework for modeling biological phenomena and discuss how
it can be used in Virtual Liver Network project. The foundational idea of the
study is that growth of cells, organs, systems and whole organisms, besides
biomolecular machinery, is influenced by biophysical mechanisms acting at
different scale levels. In particular, the general growth law uniquely defines
distribution of nutritional resources between maintenance needs and biomass
synthesis at each phase of growth and at each scale level. We exemplify the
approach considering metabolic properties of growing human and dog livers and
liver transplants. A procedure for verification of obtained results has been
introduced too. We found that two examined dogs have high metabolic rates
consuming about 0.62 and 1 gram of nutrients per cubic centimeter of liver per
day, and verified this using the proposed verification procedure. We also
evaluated consumption rate of nutrients in human livers, determining it to be
about 0.088 gram of nutrients per cubic centimeter of liver per day for males,
and about 0.098 for females. This noticeable difference can be explained by
evolutionary development, which required females to have greater liver
processing capacity to support pregnancy. We also found how much nutrients go
to biomass synthesis and maintenance at each phase of liver and liver
transplant growth. Obtained results demonstrate that the proposed approach can
be used for finding metabolic characteristics of cells, organs, and whole
organisms, which can further serve as important inputs for many applications in
biology (protein expression), biotechnology (synthesis of substances), and

The Prevalence of Workaholism: A Survey Study in a Nationally Representative Sample of Norwegian Employees
Workaholism has become an increasingly popular area for empirical study. However, most studies examining the prevalence of workaholism have used non-representative samples and measures with poorly defined cut-off scores. To overcome these methodological limitations, a nationally representative survey among employees in Norway (N = 1,124) was conducted. Questions relating to gender, age, marital status, caretaker responsibility for children, percentage of full-time equivalent, and educational level were asked. Workaholism was assessed by the use of a psychometrically validated instrument (i.e., Bergen Work Addiction Scale). Personality was assessed using the Mini-International Personality Item Pool. Results showed that the prevalence of workaholism was 8.3% (95% CI = 6.7–9.9%). An adjusted logistic regression analysis showed that workaholism was negatively related to age and positively related to the personality dimensions agreeableness, neuroticism, and intellect/imagination.
