New insights into the tonoplast architecture of plant vacuoles and vacuolar dynamics during osmotic stress
BACKGROUND: The vegetative plant vacuole occupies >90% of the volume in mature plant cells. Vacuoles play fundamental roles in adjusting cellular homeostasis and allowing cell growth. The composition of the vacuole and the regulation of its volume depend on the coordinated activities of the transporters and channels localized in the membrane (named tonoplast) surrounding the vacuole. While the tonoplast protein complexes are well studied, the tonoplast itself is less well described. To extend our knowledge of how the vacuole folds inside the plant cell, we present three-dimensional reconstructions of vacuoles from tobacco suspension cells expressing the tonoplast aquaporin fusion gene BobTIP26-1::gfp. RESULTS: 3-D reconstruction of the cell vacuole made possible an accurate analysis of large spanning folds of the vacuolar membrane under both normal and stressed conditions, and suggested interactions between surrounding plastids. Dynamic, high resolution 3-D pictures of the vacuole in tobacco suspension cells monitored under different growth conditions provide additional details about vacuolar architecture. The GFP-decorated vacuole is a single continuous compartment transected by tubular-like transvacuolar strands and large membrane surfaces. Cell culture under osmotic stress led to a complex vacuolar network with an increased tonoplast surface area. In-depth 3-D realistic inspections showed that the unity of the vacuole is maintained during acclimation to osmotic stress. Vacuolar unity exhibited during stress adaptation, coupled with the intimate associations of vacuoles with other organelles, suggests a physiological role for the vacuole in metabolism, and communication between the vacuole and organelles, respectively, in plant cells. Desiccation stress ensuing from PEG treatment generates "double" membrane structures closely linked to the tonoplast within the vacuole. These membrane structures may serve as membrane reservoirs for membrane reversion when cells are reintroduced to normal growth conditions. CONCLUSION: 3-D processing of a GFP-labeled tonoplast provides compelling visual constructions of the plant cell vacuole and elaborates on the nature of tonoplast folding and architecture. Computergestützte Visualisierung eines human-embryonalen Gehirns
In der vorliegenden Arbeit wurde das 3-D-Modell des Gehirns eines frühen humanen Embryos angefertigt,
des Weiteren eine 3-D-Software entwickelt, die es erlaubt, das Modell in Echtzeit
manipulierbar darzustellen und es schließlich vollwertig stereoskopisch betrachten zu können. Diese
Software wird Studierenden auf dem Server des Leibniz-Rechenzentrums zur Verfügung gestellt.
Damit können sie am eigenen Rechner virtuelle 3-D-Modelle, die am Lehrstuhl III der
Anatomischen Anstalt erarbeitet und bereit gestellt werden, plastisch (auch stereoskopisch)
studieren. So besteht in Zukunft die Möglichkeit, die embryonale Entwicklung mit zeitgemäßen
Methoden leicht verständlich zu veranschaulichen.
Dem 3-D-Modell diente als Quellmaterial eine Schnittserie aus 574 Schichten eines
menschlichen Embryos im Carnegie-Stadium 18. Die Schichten wurden über ein Mikroskop
digitalisiert und am Computer wieder räumlich zueinander ausgerichtet. Um die ursprünglichen
anatomischen Verhältnisse trotz der verzerrten Schnitte mit dem kommerziell verfügbaren
Programm AmiraDev 3.0 möglichst korrekt herzustellen, wurde dieser elementare aber komplizierte
Schritt durch selbst entwickelte Techniken unterstützt und sichtbar verbessert. Im so entstandenen
Bilderstapel wurde das Gehirn markiert und dann zum virtuellen Modell trianguliert.
Die hier entwickelte 3-D-Software erlaubt es, das willkürlich drehbare 3-D-Modell sowie andere
Rekonstruktionen am Rechner anzuzeigen. Eine frei wählbare Schnittebene und die Transparenzfunktion
geben Aufschluss über den inneren Aufbau des 3-D-Modells, z. B. über das Ventrikelsystem.
In der Programmiersprache C++ wurden hocheffiziente, handoptimierte Bibliotheken für
lineare Algebra und Computergrafik entwickelt, die eine ruckfreie Betrachtung ermöglichen. Im
Hinblick auf Effizienz, Erweiterbarkeit und Fehlervermeidung wurde auf ein wohl überlegtes
Software-Design mit sicherer Semantik Wert gelegt.
Auch wenn ein virtuelles 3-D-Modell bereits einen besseren räumlichen Eindruck als eine plane
Abbildung verschafft, kommt eine echte Tiefenwirkung erst durch stereoskopische Darstellung
zustande. Diese wurde lege artis als asymmetrische perspektivische Projektion so implementiert,
dass sie unkompliziert auf Tastendruck genutzt werden kann. Die ausgereifte Software beherrscht
das Anaglyphenverfahren (Rot-Grün-Brille) genauso wie auch aufwendigere Projektionsverfahren.
Die Arbeit stellt darüber hinaus in kurzer Form die für die Programmentwicklung relevanten
mathematischen Grundlagen dar. Ferner wird ein Überblick über die im Internet verfügbaren, teils
kommerziell vertriebenen Datensätze – speziell zur Embryologie – gegeben und das selbst
entwickelte Darstellungsmodell mit seinen Vorteilen und den (selbst auferlegten) Beschränkungen
in dieses Bezugssystem eingeordnet
Volume visualization of multicolor laser confocal microscope data. Comput Graph 25:371
The objective of this work is to give biologists the ability to visualize Multicolor Laser Confocal Microscope (MLCM) data sets. We present the application of volume ray casting techniques to create fast and accurate visualization of the spatial structures in MLCM data sets. We provide various volume pre-processing routines for noise removal, smoothing and edge enhancement. The tool has an interactive interface to assist the operator in classifying the volume using various transparency mappings. We have developed volume rendering using volumetric ray casting based upon a perspective camera model. We illustrate an innovative way to simultaneously visualize LCM data set from three lasers using voxel level merging. A method to generate key frame animation sequence of an arbitrary fly through path is also included in the visualization tool
