Lucas Galon’s Concerto for Violin, Percussion and Strings (2018): a composer in search of his voice
In this paper, I investigate how the composer searches for his own voice in his violin concerto while using a blend of influences such as Bartok, twelve-tone and Brazilian popular music. Galon argues that composers such as Bartok, Stravinsky and Villa-Lobos followed an independent, more varied compositional style without subscribing to any specific method.[1]On the other hand, the self-proclaimed mainstream of the Second Viennese School established a very structured, particular way of writing music. The composer seems to put into question the mechanization of composition of the dodecaphonic method, but validates its use as a way of refraining his creative impulse.[2]While Bartok’s Violin Concerto No. 2provides a framework for his piece; the tools he uses to manipulate the musical material are drawn from a free use of serialism and Brazilian contemporary music philosophy and aesthetic.

The presence of the viola caipira in Kilza Setti’s Missa Caiçara: objective and subjective aspects
This article analyzes the Brazilian composer Kilza Setti’s Missa Caiçara (Caiçara Mass), focusing on the fact that the viola caipira (peasant viola) was widely used in this piece, along with other instruments that are frequently employed in this musical genre. Some objective and subjective aspects of the compositional process of this piece are discussed here, as an attempt to clarify the technical and stylistic procedures used by the musician in her oeuvre. Among the objective aspects is the employment of specific musical elements typically found in the fishing communities’ music. Two of them are strumming chords and rhythmic motives that derive from the Portuguese fandango. On a more subjective tone Kilza pays tribute to São Gonçalo by quoting tunes that were traditionally sang in folk festivities. São Gonçalo is a saint who is worshipped by the fishing communities for protecting infertile married women and prostitutes. Due to the limited amount of publications on Kilza Setti’s Missa Caiçara, the main sources used were: an interview with the composer, which occurred in 2016; Kilza’s Doctoral research about the coastline communities’ music; José Luiz Chamorro Ribalta’s Masters Dissertation about Missa caiçara, and the score and recording of the piece itself.
This paper presents the genuine initiative of pianist Beatriz Balzi (1936-2001) to improve the quality of the 20th century piano repertoire through alternative choices. From 1984 to 2000 the Argentinian-Brazilian interpreter produced and performed a CD collection of seven volumes, enclosing 54 contemporary piano pieces to exemplify 13 countries of Latin America. Aware of the public’s inexperience about this music, hardly divulged by the ordinary Western media at that time, Balzi adopted a pedagogical posture while organizing the compositions on her CDs, as we show in this article
Music is powerful as a social mode, and a music performance offers a compelling means of communication which reveals aspects of the performer’s personality and musicianship. A pianist’s creative imagination is the key driver giving music its emotional appeal, with the performance built on the score interpretation, which involves a fluid concept, so that each performer can offer a distinctive rendition. The soundtrack involves continuous interaction with the music, with musicians evaluating the quality and fit of the sounds with the dramatic concept of the score. The playing actions generating the sounds communicates the music’s structural and emotional features, with these actions increasing in amplitude with the pianist’s increasing skill and connection with the music. Although the aural aspect was previously considered core to the domain of music, the visual information also influences the observers’ perception of the music’s expressive intent, and this is important particularly for music-naïve audience members. The presence of observers during a performance provides a motivating factor for musicians, affecting the amplitude and intensity of movements which, in turn, increase the audience engagement with the playing. At this level of performance, skilled musicians sense the playing movements as intrinsically rewarding, and undergo a state of effortless attention and high arousal which generates in them a subjective state of optimal experience. Sensing the musician’s connection with the music engages the attention of audience members, forging a bond between them which results in neural activity in the same brain regions of both, enabling a sharing of the affective experience. Successful performance communication involves the performer capturing the imagination of an audience, and this review explores the different performance elements shaping the communication of music and conveying the music’s expressivity, and the features that affect observers and enhance their enjoyment of the music. The review concludes with an examination of the performance appraisal process.
Pianism: performance communication and the playing technique
A pianist’s movements are fundamental to music-making by producing the musical sounds and the expressive movements of the trunk and arms which communicate the music’s structural and emotional information making it valuable for this review to examine upper-body movement in the performance process in combination with the factors important in skill acquisition. The underpinning playing technique must be efficient with economic muscle use by using body segments according to their design and movement potential with the arm segments mechanically linked to produce coordinated and fluent movement. Two physiologically and pianistically important actions proposed by early music scientists to deliver the keystroke involve dropping the hand from the shoulders towards the keys via a wave action with the joints activated sequentially, and forearm rotation to position the fingers for the keystroke, an action followed by the elbow/upper-arm rotating in the opposite direction. Both actions spare the forearm muscles by generating the energy needed in the larger shoulder muscles. The hand in the playing position has a curved palm through action of the metacarpal (knuckle) joints and curved fingers. Palm/finger posture controls sound quality from loud, high tempo sounds to a more mellow legato articulation, and to perform effectively the forearms should slope down towards the keyboard. The technique must be automatic through systematic practice which develops the motor skills for proficient playing, with practice duration tempered to reduce the risk of causing injury through overuse of the forearm muscles. Efficient movement patterns and strategic muscle relaxation which results in faster movement are realized only through extensive training. The constant movements of the head and trunk, and flowing arm movement with frequent hand lifts and rotational elbow movements, although generated in producing the playing technique, resonate with audience members who perceive them as expressive and thereby creating in them an empathic engagement with the music. It was proposed that music students be trained in the mechanical aspects of upper-body use in the playing technique, and practice strategies, with specialist pedagogy for children to develop motor skills for efficient playing, and training methods fostering an appreciation of the communicative aspects of music performance
Physical, genetic and functional interactions between the eisosome protein Pil1 and the MBOAT O-acyltransferase Gup1
The Saccharomyces cerevisiae MBOAT O-acyltransferase Gup1 is involved in many processes, including cell wall and membrane composition and integrity, and acetic acid-induced cell death. Gup1 was previously shown to interact physically with the mitochondrial membrane VDAC (Voltage-Dependent Anion Channel) protein Por1 and the ammonium transceptor Mep2. By co-immunoprecipitation, the eisosome core component Pil1 was identified as a novel physical interaction partner of Gup1. The expression of PIL1 and Pil1 protein levels were found to be unaffected by GUP1 deletion. In ∆gup1 cells, Pil1 was distributed in dots (likely representing eisosomes) in the membrane, identically to wt cells. However, ∆gup1 cells presented 50% less Pil1-GFP dots/eisosomes, suggesting that Gup1 is important for eisosome formation. The two proteins also interact genetically in the maintenance of cell wall integrity, and during arsenite and acetic acid exposure. We show that Δgup1 Δpil1 cells take up more arsenite than wt and are extremely sensitive to arsenite and to acetic acid treatments. The latter causes a severe apoptotic wt-like cell death phenotype, epistatically reverting the ∆gup1 necrotic type of death. Gup1 and Pil1 are thus physically, genetically and functionally connected.JT and MA-R are Ph.D. students from FCT (Fundação para a Ciência e Tecnologia), Portugal (grant numbers SFRH/BD/76025/2011 and SFRH/BD/145354/2019). Saccharomyces cerevisiae glycerol/H+ symporter Stl1p is essential for cold/near-freeze and freeze stress adaptation. A simple recipe with high biotechnological potential is given
<p>Abstract</p> <p>Background</p> <p>Freezing is an increasingly important means of preservation and storage of microbial strains used for many types of industrial applications including food processing. However, the yeast mechanisms of tolerance and sensitivity to freeze or near-freeze stress are still poorly understood. More knowledge on this regard would improve their biotechnological potential. Glycerol, in particular intracellular glycerol, has been assigned as a cryoprotectant, also important for cold/near-freeze stress adaptation. The <it>S. cerevisiae </it>glycerol active transporter Stl1p plays an important role on the fast accumulation of glycerol. This gene is expressed under gluconeogenic conditions, under osmotic shock and stress, as well as under high temperatures.</p> <p>Results</p> <p>We found that cells grown on <it>STL1 </it>induction medium (YPGE) and subjected to cold/near-freeze stress, displayed an extremely high expression of this gene, also visible at glycerol/H+ symporter activity level. Under the same conditions, the strains harbouring this transporter accumulated more than 400 mM glycerol, whereas the glycerol/H+ symporter mutant presented less than 1 mM. Consistently, the strains able to accumulate glycerol survive 25-50% more than the <it>stl1Δ </it>mutant.</p> <p>Conclusions</p> <p>In this work, we report the contribution of the glycerol/H+ symporter Stl1p for the accumulation and maintenance of glycerol intracellular levels, and consequently cell survival at cold/near-freeze and freeze temperatures. These findings have a high biotechnological impact, as they show that any <it>S. cerevisiae </it>strain already in use can become more resistant to cold/freeze-thaw stress just by simply adding glycerol to the broth. The combination of low temperatures with extracellular glycerol will induce the transporter Stl1p. This solution avoids the use of transgenic strains, in particular in food industry.</p
Characterization of ScGup1 partners in the yeast putative Hedgehog-like morphogenic pathway
associated with glycerol uptake, and then involved in a wide range of cellular processes,
including: (i) plasma membrane and cell wall composition, (ii) rafts assembly and
integrity, (iii) lipid metabolism and GPI anchor remodeling, (iv) trafficking, (v)
cytoskeleton polarization and budding pattern, (vi) telomere length, (vii) cell death, and
(viii) ECM composition among others. Candida albicans Gup1 was also associated with
morphogenesis and differentiation. The disruption of GUP1 in this pathogenic yeast
reduces virulence, affecting its capacity to adhere/invade, to differentiate into hyphae
and to form biofilms. Yeast Gup1 and Gup2 proteins in higher Eukaryotes, respectively
HHATL and HHAT, are regulators of the morphogenic cell-cell signalling Hedgehog
pathway. HHAT is responsible for the palmitoylation of the Hedgehog secreted
morphogen, and HHATL for its negative regulation. The existence of a paracrine
signaling pathway similar to Hedgehog was never described in microbial cells.
However, unicellular organism can form large aggregates of cells like colonies or
biofilms that have a tissue-like behavior, where cells differentiate, specialize, and
spatially organize, supported by a complex saccharide and proteinatious ECM.
Therefore, cell-cell communication must underlie these numerous communities. It
remains unclear, however, whether this occurs through a diffusible chemical, like
ammonia or quorum-sensing chemicals, or through a peptide signal like the Hh
morphogen from higher Eukaryotes. The presence of a Gup/HHAT(L) protein in all
Eukaryotes suggests a conserved mechanism in which these proteins might be involved.
The main goal of this work was to identify and characterize the proteins interacting
physically with Gup1 in S. cerevisiae, as a first step to disclose the function(s) of Gup
proteins in yeast. Several proteins were previously suggested to putatively interact with
Gup1, though only one did not arise from HTP surveys, the ammonium transceptor
Mep2. In this work, two novel Gup1 physical interactions were found: the yeast outer
mitochondrial membrane VDAC (Por1), and the eisosome core component Pil1. The
interaction between Gup1 and the newly identified Por1 and Pil1 partners, as well as the
previously identified Mep2, was studied: (i) the expression and localization of these
partners was assessed by RT-PCR and GFP fluorescence respectively, and (ii) several
processes commonly associated to Gup1 were evaluated phenotypically, for which
purpose new single and double deleted strains were built. This work was supported by the strategic programme UID/BIA/04050/2013 (POCI-01-0145-FEDER-
007569) funded by national funds through the FCT I.P. and by the ERDF through the COMPETE2020 -
Programa Operacional Competitividade e Internacionalização (POCI). Joana Tulha is a PhD student
SFRH/BD/76025/2011 from FCT (Fundação para a Ciência e Tecnologia). Although the expression of neither Gup1 partner seems to be significantly altered by
the deletion of GUP1, its absence affects the distribution of Por1, and Mep2.
Importantly, the interaction between Gup1 and Por1, proved to be determinant for the
nature of acetic acid-induced cell death, which changes from a necrosis-like program in
Δgup1 cells, to what seems to be an apoptotic-like cell death in the absence of both
proteins. In spite of the mitochondrial localization of Por1, its interaction with Gup1 is
also important for the control of cell wall integrity, possible through the regulation CWI
signaling, and for the differentiation of structured colonies and development of
multicellular aggregates/mats. On the other hand, the interaction between Gup1 and the
physical partner Pil1 seems to be important for the organization and/or stability of the
plasma membrane. In the absence of Gup1, the number of eisosomes was reduced,
suggesting an inefficient Pil1 assembly at the membrane, possibly related to the altered
levels of phosphoinositide. Moreover, the absence of Pil1 increases the susceptibility of
yeast cells to SDS, a phenotype that is exacerbated in Δgup1Δpil1 mutants. Finally,
Gup1 and Mep2 seem to collaborate in the definition of cell wall composition/structure.
Deletion of Mep2 in Δgup1 cells increases their sensitivity to some cell wall related
stresses, suggesting that Mep2-associated transported and/or signaling could be
important for cell survival when the cell wall is affected. Accordingly, both proteins
appear to be essential to adherence/invasive growth of yeast cells.
The work developed in this thesis represents the first systematic effort to identify
Gup1 physical interactors, and a first step to understand the biological relevance and the
niche of these interactions. Previous data suggest that yeast Gup1 is, or locates at, a hub
between CWI, TORC1, TORC2/YPK, and HOG pathways. The present work results are
compatible with this possibility, and highlight the intricate and complex role of Gup
proteins in yeast cells, by showing that Gup1 interacts with mitochondrial, membrane
and eisosomal proteins in the regulation of processes as different as cell death or plasma
membrane and cell wall composition/organization.
A deleção do GUP1 reduz a
A comunicação intercelular deverá
Finalmente, as proteínas Gup1 e Mep2
CWI, TORC1, TORC2/YPK e HOG.
