Structural design of concrete filled steel elliptical hollow sections
This paper presents the behaviour and design of axially loaded elliptical steel hollow sections filled with normal and high strength concrete. The experimental investigation was conducted with three nominal wall thickness (4mm, 5mm and 6.3mm) and different infill concrete cube
strengths varied from 30 to 100 MPa. The effect of steel tube thickness, concrete strength, and confinement were discussed together with column strengths and load-axial shortening curves were evaluated. The study is limited to cross-section capacity and has not been validated at
member level. Comparisons of the tests results together with other available results from the literature have been made with current design method used for the design of composite circular steel sections in Eurocode 4 and AISC codes. It was found that existing design guidance for
concrete filled circular hollow sections may generally be safely applied to concrete filled elliptical
steel tubes
Design and evaluation of 16S rRNA sequence based oligonucleotide probes for the detection and quantification of Comamonas testosteroni in mixed microbial communities
Background
The β-proteobacterial species Comamonas testosteroni is capable of biotransformation and also biodegradation of a range of chemical compounds and thus potentially useful in chemical manufacturing and bioremediation. The ability to detect and quantify members of this species in mixed microbial communities thus may be desirable.
Results
We have designed an oligonucleotide probe for use in fluorescent in situ hybridization (FISH) and two pairs of PCR primers targeting a C. testosteroni subgroup. The FISH probe and one of the PCR primer pairs are suitable for quantification of C. testosteroni in mixed microbial communities using FISH followed by quantitative image analysis or real-time quantitative PCR, respectively. This has been shown by analysis of samples from an enrichment of activated sludge on testosterone resulting in an increase in abundance and finally isolation of C. testosteroni. Additionally, we have successfully used quantitative PCR to follow the C. testosteroni abundance during a laboratory scale wastewater bioaugmentation experiment.
Conclusion
The oligonucleotides presented here provide a useful tool to study C. testosteroni population dynamics in mixed microbial communities
Thermal Perception in Mild Climate: Adaptive Thermal Models for Schools
A comprehensive assessment of indoor environmental conditions is performed on a
representative sample of classrooms in schools across southern Spain (Mediterranean climate) to
evaluate the thermal comfort level, thermal perception and preference, and the relationship with
HVAC systems, with a comparison of seasons and personal clothing. Almost fifty classrooms were
studied and around one thousand pool-surveys distributed among their occupants, aged 12 to 17.
These measurements were performed during spring, autumn, and winter, considered the most
representative periods of use for schools. A new proposed protocol has been developed for the
collection and subsequent analysis of data, applying thermal comfort indicators and using the most
frequent predictive models, rational (RTC) and adaptive (ATC), for comparison. Cooling is not
provided in any of the rooms and natural ventilation is found in most of the spaces during midseasons.
Despite the existence of a general heating service in almost all classrooms in the cold period, the use
of mechanical ventilation is limited. Heating did not usually provide standard set-point temperatures.
However, this did not lead to widespread complaints, as occupants perceive the thermal environment
as neutral—varying greatly between users—and show a preference for slightly colder environments.
Comparison of these thermal comfort votes and the thermal comfort indicators used showed a better
fit of thermal preference over thermal sensation and more reliable results when using regional ATC
indicators than the ASHRAE adaptive model. This highlights the significance of inhabitants’ actual
thermal perception. These findings provide useful insight for a more accurate design of this type of
building, as well as a suitable tool for the improvement of existing spaces, improving the conditions
for both comfort and wellbeing in these spaces, as well as providing a better fit of energy use for
actual comfort conditions
