A preliminary transcriptomic approach to elucidate post harvest ripening of plum fruit
The aim of the present study was to dissect common and/or diverse mechanisms regulating plum (Prunus salicina) fruit ripening in genotypes characterized by different patterns of ethylene production. Fruit of an ethylenesuppressed cultivar ('Shiro') and a cultivar characterized by the typical increase of ethylene production during fruit ripening ('Santa Rosa') were harvested at commercial maturity stage and allowed to further ripen at room temperature (23°C) up to 4 days. While non-detectable amounts of ethylene were recorded in 'Shiro' fruit, a typical climacteric behavior was observed in 'Santa Rosa' plums. For comparative purposes, the peach microarray μPEACH 1.0 containing 4,806 oligonucleotides corresponding to an equal number of genes expressed in peach fruit was employed for transcript profiling during postharvest ripening of both cultivars. Intriguingly, transcript levels of genes involved in ethylene biosynthesis, primarily 1-aminocyclopropane-1-carboxylate synthase, appeared to increase during the progress of 'Shiro' fruit ripening, following the same pattern as in 'Santa Rosa' plums. These data suggest that an induction of the ethylene biosynthetic pathway is present also in plum cultivars in which the burst of ethylene is not detectable. Expression levels of other genes implicated in auxin metabolism, antioxidant system and stress response followed the same pattern in both cultivars. Overall, this preliminary transcriptomic approach tried to elucidate the flow of events that accompany postharvest ripening of plum cultivars with diverse properties in relation to ethylene evolutio
Discovery of non-climacteric and suppressed climacteric bud sport mutations originating from a climacteric Japanese plum cultivar (Prunus salicina Lindl.).
Japanese plums are classified as climacteric; however, some economically important cultivars selected in California produce very little ethylene and require long ripening both "on" and "off" the tree to reach eating-ripe firmness. To unravel the ripening behavior of different Japanese plum cultivars, ripening was examined in the absence (air) or in the presence of ethylene or propylene (an ethylene analog) following a treatment or not with 1-methylcyclopropene (1-MCP, an ethylene action inhibitor). Detailed physiological studies revealed for the first time three distinct ripening types in plum fruit: climacteric, suppressed-climacteric, and non-climacteric. Responding to exogenous ethylene or propylene, the slow-softening supressed-climacteric cultivars produced detectable amounts of ethylene, in contrast to the novel non-climacteric cultivar that produced no ethylene and softened extremely slowly. Genetic analysis using microsatellite markers produced identical DNA profiles for the climacteric cultivars "Santa Rosa" and "July Santa Rosa," the suppressed-climacteric cultivars "Late Santa Rosa," "Casselman," and "Roysum" and the novel non-climacteric "Sweet Miriam," as expected since historic records present most of these cultivars as bud-sport mutations derived initially from "Santa Rosa." This present study provides a novel fruit system to address the molecular basis of ripening and to develop markers that assist breeders in providing high-quality stone fruit cultivars that can remain "on-tree," increasing fruit flavor, saving harvesting costs, and potentially reducing the need for low-temperature storage during postharvest handling

Sea turtle nesting activity along Eglin Air Force Base on Cape San Blas and Santa Rosa Island, Florida from 1994 to 1997.
Along EAFB on Cape San BIas, the only sea turtle species nest observed has been
the loggerhead turtle. The first green turtle nest documented along the Florida panhandle
coast was observed on EAFB property, however (D. Atencio, EAFB, pers. comm). Santa
Rosa Island, located approximately 150 miles west of Cape San BIas supports a small but
consistent, group of nesting green turtles (Fig. 2). Although erosion is not as severe along
Santa Rosa Island as it is on Cape San BIas, and vehicular traffic is not permitted, sea
turtles nesting on this barrier island must survive severe tropical storms, predation, and
artificial lighting to be successful. Because this area supports a rare group of nesting green
turtles and is disturbed by intense artificial lighting from Air Force missions and adjacent
resort towns, continued monitoring is necessary. The sea turtle species that nest along this
barrier island, and the human activities that disturb those sea turtles present unique
circumstances for management ofthis area. Protection ofthe significant nesting
populations of sea turtles on EAFB properties on Cape San BIas and Santa Rosa Island
requires yearly monitoring of the nesting activity and the natural and human disturbances
influencing the nesting females.
The objectives ofthis study were to monitor sea turtle nesting along EAFB on
Cape San BIas to determine number of nests and hatching success, assess disturbances,
and determine proper management to ensure successful nesting and hatching.(56 page document

Newspapers Turning Ideas into Dollars: Four Revenue Success Stories
The report follows a year-long effort to identify newspaper successes in the search for new business models. This report analyzes four such dailies -- the Naples (Fla.) Daily News, the Santa Rosa (Calif.) Press Democrat, the (Salt Lake City) Deseret News, the Columbia (Tenn.) Daily Herald -- whose executives explained, in detail, the motivation and strategy behind their experiments and shared internal data about the results. Their innovations-ranging from sales force restructuring to rebranding the print product to web consulting for local merchants-are generating significant new income
