Isocitrate Lyase from Germinating Soybean Cotyledons: Purification and Characterization

Isocitrate lyase (E.C. 4.1.3.1) was purified from the cotyledons of 7-d-old soybean seedlings. Three molecular forms were detected with pi values of 6·46, 6·25 and 6·0. The main form (pl = 6·46) had an approximate Mr of 130000, a pH optimum of 8·0, a Km (isocitrate) close to 2·0 mol m−3 and a molecular activity of 615 min −1 at 25 °C. Thermodynamic Conditions in Quenching Chamber of Low Voltage Circuit Breaker
Práce se zabývá studiem procesů probíhajících při zhášení silnoproudého oblouku ve zhášecí komoře jističe. Je zaměřena na výpočet dynamiky tekutin a teplotního pole v okolí elektrického oblouku. V práci je dále popsán vliv vzdálenosti plechů v komoře a vliv tvarů plechů z hlediska aerodynamických podmínek uvnitř komory. Dalším cílem dosaženým touto prací je poskytnutí informací o vlivu polohy elektrického oblouku na termodynamické vlastnosti uvnitř komory. Toto je důležité, zejména pokud je oblouk do komory vtahován jinými silami, např. elektromagnetickými a během tohoto vtahovacího procesu mění svůj tvar i polohu. Za účelem co nejjednoduššího, ale zároveň co nejefektivnějšího řešení úkolu, byl vyvinut software určen speciálně pro výpočet dynamiky tekutin numerickou metodou konečných objemů (FVM). Tato metoda je, v porovnání s rozšířenější metodou konečných prvků (FEM), vhodnější pro výpočet dynamiky tekutin (CFD) zejména proto, že režie na výpočet jedné iterace jsou menší v porovnání s ostatními numerickými metodami. Další výhodou tohoto softwarového řešení je jeho modularita a rozšiřitelnost. Cely koncept softwaru je postaven na tzv. zásuvných modulech. Díky tomuto řešení můžeme využít výpočtové jádro pro další numerické analýzy, např. strukturální, elektromagnetickou apod. Jediná potřeba pro úspěšné používání těchto analýz je napsáni solveru pro konečné prvky (FEM). Jelikož je software koncipován jako multi–thread aplikace, využívá výkon současných vícejádrových procesorů naplno. Tato vlastnost se ještě více projeví, pokud se výpočet přesune z CPU na GPU. Jelikož současné grafické karty vyšších tříd mají několik desítek až stovek výpočetních jader a pracují s mnohem rychlejšími pamětmi, než CPU, je výpočetní výkon několikanásobně vyšší.Work deals with the study of processes that attend the electric arc extinction inside the quenching chamber of a circuit breaker. It is focused on several areas. The first one is concerned to fluid dynamics calculations (CFD) and the second one is aimed at thermal field calculations. In this work effects of metal plates distance together with metal plates shapes are described from aerodynamical point of view. Another objective solved by this work is to give information about influence of an electric arc position in a quenching chamber, which changed its shape due to forces acting on it during extinction process. For purpose of this work a new software solution for CFD was developed. Whole software concept is based on plug-ins. Due to this solution, the software§s calculation core can be used for other numerical analyses, like structural, electromagnetic, etc. The only requirement is to write a plug-in for these analyses. Because the software is designed as multi-threaded application, it can use the fully performance of current multi-core processors. Above mentioned property can be especially shown off, when a calculation is moved from CPU to GPU (Graphics Processing Units). Current high-end graphic cards have tens to hundreds cores and work with faster memories than CPU. Due to this fact, the simulation performance can raised manifold.
Sludge Production and Disposal for Small Cold Climate Bio-Treatment Plants
Ultimate disposal of wastewater sludge has long been a problem which
to a large degree has been ignored. Haney (1971) stated that: "Until
process sludge can be handled with minimum environmental impact, we cannot
claim to have a viable wastewater treatment process". The relationship
of sludge disposal to total treatment processes is emphasized by
the fact that sludge handling and disposal represents up to 50 percent
of the total treatment capital and operating costs (Burd, 1968). Processing
of wastewater sludge will, no doubt, receive increased attention
in the future because of environmental concerns for our air, land and
water.
The present technology for processing wastewater treatment plant
sludge is well established and includes conditioning, dewatering, and
disposal. Many of these processes are highly sophisticated and relatively
expensive. Most of the more advanced processes are unsuitable for small
wastewater treatment facilities in Alaska.The work upon which this report is based was supported in part by

Thermomechanical and hygroelastic properties of an epoxy system under humid and cold-warm cycling conditions
In this paper, we study the hygrothermal aging of an anhydride-cured epoxy under temperature and hygrometry conditions simulating those experienced by an aircraft in wet tropical or subtropical regions. Gravimetric and dimensional measurements were performed and they indicate that there are three stages in this aging process: the first one, corresponding to the early cycles can be called the “induction stage”. The second stage of about 1000 cycles duration, could be named the “swelling stage”, during which the volume increase is almost equal to the volume of the (liquid) water absorbed. Both the first and second stages are accompanied by modifications of the mechanical properties and the glass transition temperature. During the third (“equilibrium”) stage, up to 3000 cycles, there is no significant change in the physical properties despite the continuous increase of water uptake. This can be explained by the fact that only physically sorbed water can influence physical properties
Minimum Cut Tree Games
In this paper we introduce a cooperative game based on the minimum cut tree problem which is also known as multi-terminal maximum flow problem. Minimum cut tree games are shown to be totally balanced and a solution in their core can be obtained in polynomial time. This special core allocation is closely related to the solution of the original graph theoretical problem. We give an example showing that the game is not supermodular in general, however, it is for special cases and for some of those we give an explicit formula for the calculation of the Shapley value
Atomic vapor deposition of bismuth titanate thin films
c-axis oriented ferroelectric bismuth titanate (Bi4Ti 3O12) thin films were grown on (001) strontium titanate (SrTiO3) substrates by an atomic vapor deposition technique. The ferroelectric properties of the thin films are greatly affected by the presence of various kinds of defects. Detailed x-ray diffraction data and transmission electron microscopy analysis demonstrated the presence of out-of-phase boundaries (OPBs). It is found that the OPB density changes appreciably with the amount of titanium injected during growth of the thin films. Piezo-responses of the thin films were measured by piezo-force microscopy. It is found that the in-plane piezoresponse is stronger than the out-of-plane response, due to the strong c-axis orientation of the films
Higher plant diversity enhances soil stability in disturbed alpine ecosystems
Plant diversity is hypothesised to increase soil stability by increasing the diversity of root types. To test this hypothesis, we took soil cores from machine-graded ski slopes and from the adjacent undisturbed vegetation as a control. We quantified aggregate stability as an indicator for soil stability in relation to (1) abiotic soil properties, (2) above-ground vegetation characteristics and (3) root parameters. From the three groups of variables, the number of plant species, root density (RD) and sand content showed the highest correlation with soil aggregate stability and explained 54% of its variance. In variance partitioning, the number of plant species was the most relevant factor explaining 19% of the variance in aggregate stability. Further, it explained another 11% through shared effects with RD and sand content. An additional 8% was explained through the shared influence with sand content. Plant species showing the highest correlation with overall diversity were from different functional groups (grasses, forbs and shrubs), meaning that beneficial effects can not only be assigned to one specific functional group, but to the combination of several groups. Our data demonstrate the positive effect of plant diversity on aggregate stability. Megahertz peaked-spectrum sources in the Boötes field I - a route towards finding high-redshift AGN?

Published by Oxford University Press on behalf of the Royal Astronomical Society.We present a 324.5MHz image of the NOAO Bo\"otes field that was made using Very Large Array (VLA) P-band observations. The image has a resolution of 5.6x5.1arcsec, a radius of and a central noise of ~0.2mJy\beam. Both the resolution and noise of the image are an order of magnitude better than what was previously available at this frequency and will serve as a valuable addition to the already extensive multiwavelength data that are available for this field. The final source catalogue contains 1370 sources and has a median 325 to 1400MHz spectral index of -0.72. Using a radio colour-colour diagram of the unresolved sources in our catalogue, we identify 33 megahertz peaked-spectrum (MPS) sources. Based on the turnover frequency linear size relation for the gigahertz peaked-spectrum (GPS) and compact steep-spectrum (CSS) sources, we expect that the MPS sources that are compact on scales of tens of milliarcseconds should be young radio loud active galactic nuclei at high (z>2) redshifts. Of the 33 MPS sources, we were able to determine redshifts for 24, with an average redshift of 1.3. Given that five of the sources are at z>2, that the four faint sources for which we could not find redshifts are likely at even higher redshifts and that we could only select sources that are compact on a scale of ~5arcsec, there is encouraging evidence that the MPS method can be used to search for high-redshift sources.Peer reviewedFinal Published versio
Lability of Pb in soil: effects of soil properties and contaminant source
Environmental Context: There is growing concern that lead (Pb) in the environment may cause adverse health effects in human populations. We investigated the combined use of isotopic abundance and isotopic dilution to show how the origins of soil Pb and soil characteristics affect lability. Soil pH and soil Pb content are the dominant controls on Pb lability; the lability of recent petrol-derived Pb is similar to that of other sources in urban soils but greater than geogenic Pb in rural roadside topsoils.
Lability of lead (Pb) in soils is influenced by both soil properties and source(s) of contamination. We investigated factors controlling Pb lability in soils from (i) land adjacent to a major rural road, (ii) a sewage processing farm and (iii) an archive of the geochemical survey of London. We measured isotopically exchangeable Pb (E-values; PbE), phase fractionation of Pb by a sequential extraction procedure (SEP) and inferred source apportionment from measured Pb isotopic ratios. Isotopic ratios (206Pb/207Pb and 208Pb/207Pb) of total soil Pb fell on a mixing line between those of petrol and UK coal or Pb ore. The main determinant of the isotopically exchangeable Pb fraction (%E-value) was soil pH: %E-values decreased with increasing pH. In rural roadside topsoils and there was also evidence that, petrol-derived Pb remained more labile (35%) than Pb from soil parent material (27%). However, in biosolid-amended and London soils %E-values were low (c. 25%), covered a restricted range, and showed no clear evidence of source-dependent lability
