Perceived thickness and creaminess modulates the short-term satiating effects of high protein drinks
Previous research suggests that increasing beverage protein content enhances subsequent satiety, but whether this effect is entirely attributable to post-ingestive effects of protein or is partly caused by the distinct sensory characteristics imparted by the presence of protein remains unclear. To try and discriminate nutritive from sensory effects of added protein, we contrasted effects of three higher energy (c. 1.2MJ) and one lower energy (LE: 0.35MJ) drink preloads on subsequent appetite and lunch intake. Two higher energy drinks had 44% of energy from protein, one with the sensory characteristics of a juice drink (HP-) and the second thicker and more creamy (HP+). The high-carbohydrate preload (HC+) was matched for thickness and creaminess to the HP+ drink. Participants (healthy male volunteers, n=26) consumed significantly less at lunch after the HP+ (566g) and HC+ (572g) than after HP- (623g) and LE (668g) drinks, although the compensation for drink energy accounted for only 50% of extra energy at best. Appetite ratings indicated that participants felt significantly less hungry and more full immediately before lunch in HP+ and HC+ compared to LE, with HP- intermediate. The finding that protein generated stronger satiety in the context of a thicker creamier drink (HP+ but not HP-), and that an isoenergetic carbohydrate drink (HC+) matched in thickness and creaminess to the HP+ drink generated the same pattern of satiety as HP+ both suggest an important role for these sensory cues in the development of protein-based satiety
The effect of fishmeal or peameal on milk fatty acid composition in organic farming
Feed fatty acid (FA) composition influences the FA composition of cow milk. In a continuous production experiment with 32 Norwegian red dairy cows fishmeal (FM) was compared to peameal (PM) as protein supplement to home-grown cereals and grass silage in organic farming. The protein supplements were together with cereals formulated to be isonitrogeneous and isoenergetic (NEL) and were compared at high (HC) and low concentrate (LC) level. The concentrate rations did not affect the intake of silage. Fishmeal resulted in significantly higher milk yield (kg) with a lower fat concentration (HC) compared to PM. Lower concentrations of urea and FFA were found in milk produced with FM compared to PM. Milk flavour and odour was equal or better when FM rather than PM was fed. Fishmeal diets increased significantly the proportions of several long-chain FAs: oleic acid (C18:1c9), vaccenic acid (C18:1c11), CLA (C18:2c9,t11, not significant at HC), C20:0, C18:1t10, and DHA (C22:6 n-3) in milk fat compared to PM. DHA, which is found in high concentrations in FM (14 g/100g FAME), had the most significant increase. The proportion of C18:3 n-3 (ALA) was significantly lower when FM was fed compared to PM. The percentage of saturated FA was significantly lower and the percentage of monounsaturated FA was higher when FM rather than PM was fed. For cows on HC the n-6/n-3 ratio was lower in the FM group than in the PM group, and the ratio was lower at LC than at HC (p = 0.006, interaction p = 0.02). Fishmeal diets included higher proportions of oats than PM diets. Oats have high content of oleic acid and may therefore have influenced the composition of FAs in milk fat as well as the protein supplements. Fishmeal increased the proportion of beneficial FAs without reducing the sensoric quality of milk. It remains unclear whether this is an effect of protein source or an effect of the higher oat proportions in FM diets
Human collagen Krox up-regulates type I collagen expression in normal and scleroderma fibroblasts through interaction with Sp1 and Sp3 transcription factors.
Despite several investigations, the transcriptional mechanisms that regulate the expression of both type I collagen genes (COL1A1 and COL1A2) in either physiological or pathological situations, such as scleroderma, are not completely known. We have investigated the role of hc-Krox transcription factor on type I collagen expression by human dermal fibroblasts. hc-Krox exerted a stimulating effect on type I collagen protein synthesis and enhanced the corresponding mRNA steady-state levels of COL1A1 and COL1A2 in foreskin fibroblasts (FF), adult normal fibroblasts (ANF), and scleroderma fibroblasts (SF). Forced hc-Krox expression was found to up-regulate COL1A1 transcription through a -112/-61-bp sequence in FF, ANF, and SF. Knockdown of hc-Krox by short interfering RNA and decoy strategies confirmed the transactivating effect of hc-Krox and decreased substantially COL1A1 transcription levels in all fibro-blast types. The -112/-61-bp sequence bound specifically hc-Krox but also Sp1 and CBF. Attempts to elucidate the potential interactions between hc-Krox, Sp1, and Sp3 revealed that all of them co-immunoprecipitate from FF cellular extracts when a c-Krox antibody was used and bind to the COL1A1 promoter in chromatin immunoprecipitation assays. Moreover, hc-Krox DNA binding activity to its COL1A1-responsive element is increased in SF, cells producing higher amounts of type I collagen compared with ANF and FF. These data suggest that the regulation of COL1A1 gene transcription in human dermal fibroblasts involves a complex machinery that implicates at least three transcription proteins, hc-Krox, Sp1, and Sp3, which could act in concert to up-regulate COL1A1 transcriptional activity and provide evidence for a pro-fibrotic role of hc-Krox
Low-carbohydrate diets affect energy balance and fuel homeostasis differentially in lean and obese rats
In parallel with increased prevalence of overweight people in affluent societies are individuals trying to lose weight, often using low-carbohydrate diets. Nevertheless, long-term metabolic consequences of those diets, usually high in (saturated) fat, remain unclear. Therefore, we investigated long-term effects of high-fat diets with different carbohydrate/protein ratios on energy balance and fuel homeostasis in obese (fa/fa) Zucker and lean Wistar rats. Animals were fed high-carbohydrate (HC), high-fat (HsF), or low-carbohydrate, high-fat, high-protein (LC-HsF-HP) diets for 60 days. Both lines fed the LC-HsF-HP diet displayed reduced energy intake compared with those fed the HsF diet (Zucker, -3.7%) or the HC diet (Wistar rats, -12.4%). This was not associated with lower weight gain relative to HC fed rats, because of increased food efficiencies in each line fed HsF and particularly LC-HsF-HP food. Zucker rats were less glucose tolerant than Wistar rats. Lowest glucose tolerances were found in HsF and particularly in LC-HsF-HP-fed animals irrespective of line, but this paralleled reduced plasma adiponectin levels, elevated plasma resistin levels, higher retroperitoneal fat masses, and reduced insulin sensitivity (indexed by insulin-induced hypoglycemia) only in Wistar rats. In Zucker rats, however, improved insulin responses during glucose tolerance testing and tendency toward increased insulin sensitivities were observed with HsF or LC-HsF-HP feeding relative to HC feeding. Thus, despite adverse consequences of LC-HsF diets on blood glucose homeostasis, principal differences exist in the underlying hormonal regulatory mechanisms, which could have benefits for B-cell functioning and insulin action in the obese state but not in the lean state.
The effect of preservation method of barley, maturity of grass silage, and type of protein supplementation on sensoric milk quality in organic farming
Three continuous production experiments and three short term cross over experiments were carried out in Northern Norway to investigate different feeding strategies for dairy cows in organic farming. This paper focuses on the effects on sensoric milk quality. Half of the 32 Norwegian red dairy cows in the production experiments were fed 40% (HC) concentrates (on energy basis per year) and the other half 10% (LC). Twelve cows (HC) participated in the short term cross over experiments. The experimental factors in the continuous production experiments were barley preservation method (P1), grass silage maturity (P2) and type of protein supplement (P3), and in the short term cross over experiments barley preservation method (C1), type of protein supplement (C2) and time for fishmeal feeding (C3). In all experiments the cows were offered grass silage ad libitum, restricted amounts of cereals and protein feeds, and mineral and vitamin supplements. The sensoric quality of milk was in general high. In the production experiments, milk from cows in HC had slightly higher quality than from cows in LC (significant in P2, p = 0.04). Neither the preservation method of barley (dried or ensiled with molasses), maturity of grass silage or type of protein supplement (fishmeal or peameal) influenced the sensoric milk quality significantly. Also in the cross over experiments no effect of the studied factors was found in milk flavour and odour. Early harvested grass silage gave significantly lower FFA contents than grass silage cut at normal time (P2) and FM gave significantly lower FFA concentrations than peameal (P3). These results indicate that organic farmers with different feeding regimes can produce milk of first class sensoric quality. Also the content of FFA has been low in all experiments (except C1). However, feeding regimes containing low levels of concentrates may reduce milk taste slightly
E5 protein of human papillomavirus 16 downregulates HLA class I and interacts with the heavy chain via its first hydrophobic domai
Human papillomavirus type 16 E5 protein (HPV-16 E5) is expressed early in papillomavirus infection and is localised primarily in the cell Golgi apparatus (GA) and endoplasmic reticulum. E5 prevents transport of the major histocompatibility class I (MHC I; HLA class I in humans) to the cell surface and retains the complex in the GA. We report that these effects are due, at least in part, to the interaction between E5 and HLA I heavy chain (HC). We also demonstrate that the down-regulation of surface HLA I and interaction with HC are mediated by the first hydrophobic domain of E5. Although E5 downregulates classical HLA selectively as it does not downregulate non-classical HLA, the interaction with the HC of classical HLA I is not specific for a particular haplotype of HLA I. This suggests that E5 can interfere with antigen presentation by most, if not all, classical HLA I haplotypes, with potentially serious consequences as the ability of infected cells to present antigenic peptides to effector T cells would be compromised
Nanoparticles-cell association predicted by protein corona fingerprints
In a physiological environment (e.g., blood and interstitial fluids) nanoparticles (NPs) will bind proteins shaping a "protein corona" layer. The long-lived protein layer tightly bound to the NP surface is referred to as the hard corona (HC) and encodes information that controls NP bioactivity (e.g. cellular association, cellular signaling pathways, biodistribution, and toxicity). Decrypting this complex code has become a priority to predict the NP biological outcomes. Here, we use a library of 16 lipid NPs of varying size (Ø ≈ 100-250 nm) and surface chemistry (unmodified and PEGylated) to investigate the relationships between NP physicochemical properties (nanoparticle size, aggregation state and surface charge), protein corona fingerprints (PCFs), and NP-cell association. We found out that none of the NPs' physicochemical properties alone was exclusively able to account for association with human cervical cancer cell line (HeLa). For the entire library of NPs, a total of 436 distinct serum proteins were detected. We developed a predictive-validation modeling that provides a means of assessing the relative significance of the identified corona proteins. Interestingly, a minor fraction of the HC, which consists of only 8 PCFs were identified as main promoters of NP association with HeLa cells. Remarkably, identified PCFs have several receptors with high level of expression on the plasma membrane of HeLa cells
Potato virus Y HC-Pro reduces the ATPase activity of NtMinD, which results in enlarged chloroplasts in HC-Pro transgenic tobacco
Potato virus Y (PVY) is an important plant virus and causes great losses every year. Viral infection often leads to abnormal chloroplasts. The first step of chloroplast division is the formation of FtsZ ring (Z-ring), and the placement of Z-ring is coordinated by the Min system in both bacteria and plants. In our lab, the helper-component proteinase (HC-Pro) of PVY was previously found to interact with the chloroplast division protein NtMinD through a yeast two-hybrid screening assay and a bimolecular fluorescence complementation (BiFC) assay in vivo. Here, we further investigated the biological significance of the NtMinD/HC-Pro interaction. We purified the NtMinD and HC-Pro proteins using a prokaryotic protein purification system and tested the effect of HC-Pro on the ATPase activity of NtMinD in vitro. We found that the ATPase activity of NtMinD was reduced in the presence of HC-Pro. In addition, another important chloroplast division related protein, NtMinE, was cloned from the cDNA of Nicotiana tabacum. And the NtMinD/NtMinE interaction site was mapped to the C-terminus of NtMinD, which overlaps the NtMinD/HC-Pro interaction site. Yeast three-hybrid assay demonstrated that HC-Pro competes with NtMinE for binding to NtMinD. HC-Pro was previously reported to accumulate in the chloroplasts of PVY-infected tobacco and we confirmed this result in our present work. The NtMinD/NtMinE interaction is very important in the regulation of chloroplast division. To demonstrate the influence of HC-Pro on chloroplast division, we generated HC-Pro transgenic tobacco with a transit peptide to retarget HC-Pro to the chloroplasts. The HC-Pro transgenic plants showed enlarged chloroplasts. Our present study demonstrated that the interaction between HC-Pro and NtMinD interfered with the function of NtMinD in chloroplast division, which results in enlarged chloroplasts in HC-Pro transgenic tobacco. The HC-Pro/NtMinD interaction may cause the formation of abnormal chloroplasts in PVY-infected plants
Liquid meal composition, postprandial satiety hormones, and perceived appetite and satiety in obese women during acute caloric restriction
OBJECTIVE:
The purpose of this study was to compare postprandial satiety regulating hormone responses (pancreatic polypeptide (PP) and peptide tyrosine tyrosine (PYY)) and visual analog scale- (VAS) assessed perceived appetite and satiety between liquid high-protein (HP) and high-carbohydrate (HC) meals in obese women during acute (24-h) caloric restriction.
DESIGN:
Eleven obese premenopausal women completed two conditions in random order in which they consumed 1500 calories as six 250-calorie HP meals or six 250-calorie HC meals over a 12-h period. Blood samples were taken at baseline and every 20 min thereafter and analyzed for PP and PYY concentrations. At these same points, perceived hunger and fullness were assessed with a VAS. The incremental area under the curve (iAUC) was used to compare postprandial responses.
RESULTS:
THE 12-H PP AND PYY IAUC WERE GREATER (P0.05) DURING THE HP CONDITION (PP: 4727±1306 pg/ml×12 h, PYY: 1373±357 pg/ml×12 h) compared with the HC condition (PP: 2300±528 pg/ml×12 h, PYY: 754±246 pg/ml×12 h). Perceived hunger and fullness were not different between conditions (P>0.05). The greatest changes in PYY and perceived fullness occurred after the morning meals during both conditions.
CONCLUSIONS:
These data suggest that in obese women during acute caloric restriction before weight loss, i) liquid HP meals, compared with HC meals, result in greater postprandial PP and PYY concentrations, an effect not associated with differential appetite or satiety responses, and ii) meal-induced changes in PYY and satiety are greatest during the morning period, regardless of dietary macronutrient composition
Dairy foods and dairy protein consumption is inversely related to markers of adiposity in obese men and women
A number of intervention studies have reported that the prevalence of obesity may be in part inversely related to dairy food consumption while others report no association. We sought to examine relationships between energy, protein and calcium consumption from dairy foods (milk, yoghurt, cheese, dairy spreads, ice-cream) and adiposity including body mass index (BMI), waist (WC) and hip circumference (HC), and direct measures of body composition using dual energy X-ray absorptiometry (% body fat and abdominal fat) in an opportunistic sample of 720 overweight/obese Australian men and women. Mean (SD) age, weight and BMI of the population were 51 ± 10 year, 94 ± 18 kg and 32.4 ± 5.7 kg/m2, respectively. Reduced fat milk was the most commonly consumed dairy product (235 ± 200 g/day), followed by whole milk (63 ± 128 g/day) and yoghurt (53 ± 66 g/day). Overall dairy food consumption (g/day) was inversely associated with BMI, % body fat and WC (all p < 0.05). Dairy protein and dairy calcium (g/day) were both inversely associated with all adiposity measures (all p < 0.05). Yoghurt consumption (g/day) was inversely associated with % body fat, abdominal fat, WC and HC (all p < 0.05), while reduced fat milk consumption was inversely associated with BMI, WC, HC and % body fat (all p < 0.05). Within a sample of obese adults, consumption of dairy products, dairy protein, and calcium was associated with more favourable body composition
