    Evolutionary ecology of obligate fungal and microsporidian invertebrate pathogens

    The interactions between hosts and their parasites and pathogens are omnipresent in the natural world. These symbioses are not only key players in ecosystem functioning, but also drive genetic diversity through co-evolutionary adaptations. Within the speciose invertebrates, a plethora of interactions with obligate fungal and microsporidian pathogens exist, however the known interactions is likely only a fraction of the true diversity. Obligate invertebrate fungal and microsporidian pathogen require a host to continue their life cycle, some of which have specialised in certain host species and require host death to transmit to new hosts. Due to their requirement to kill a host to spread to a new one, obligate fungal and microsporidian pathogens regulate invertebrate host populations. Pathogen specialisation to a single or very few hosts has led to some fungi evolving the ability to manipulate their host’s behaviour to maximise transmission. The entomopathogenic fungus, Entomophthora muscae, infects houseflies (Musca domestica) over a week-long proliferation cycle, resulting in flies climbing to elevated positions, gluing their mouthparts to the substrate surface, and raising their wings to allow for a clear exit from fungal conidia through the host abdomen. These sequential behaviours are all timed to occur within a few hours of sunset. The E. muscae mechanisms used in controlling the mind of the fly remain relatively unknown, and whether other fitness costs ensue from an infection are understudied.European Commissio

    Talking about personal recovery in bipolar disorder: Integrating health research, natural language processing, and corpus linguistics to analyse peer online support forum posts

    Background: Personal recovery, ‘living a satisfying, hopeful and contributing lifeeven with the limitations caused by the illness’ (Anthony, 1993) is of particular value in bipolar disorder where symptoms often persist despite treatment. So far, personal recovery has only been studied in researcher-constructed environments (interviews, focus groups). Support forum posts can serve as a complementary naturalistic data source. Objective: The overarching aim of this thesis was to study personal recovery experiences that people living with bipolar disorder have shared in online support forums through integrating health research, NLP, and corpus linguistics in a mixed methods approach within a pragmatic research paradigm, while considering ethical issues and involving people with lived experience. Methods: This mixed-methods study analysed: 1) previous qualitative evidence on personal recovery in bipolar disorder from interviews and focus groups 2) who self-reports a bipolar disorder diagnosis on the online discussion platform Reddit 3) the relationship of mood and posting in mental health-specific Reddit forums (subreddits) 4) discussions of personal recovery in bipolar disorder subreddits. Results: A systematic review of qualitative evidence resulted in the first framework for personal recovery in bipolar disorder, POETIC (Purpose & meaning, Optimism & hope, Empowerment, Tensions, Identity, Connectedness). Mainly young or middle-aged US-based adults self-report a bipolar disorder diagnosis on Reddit. Of these, those experiencing more intense emotions appear to be more likely to post in mental health support subreddits. Their personal recovery-related discussions in bipolar disorder subreddits primarily focussed on three domains: Purpose & meaning (particularly reproductive decisions, work), Connectedness (romantic relationships, social support), Empowerment (self-management, personal responsibility). Support forum data highlighted personal recovery issues that exclusively or more frequently came up online compared to previous evidence from interviews and focus groups. Conclusion: This project is the first to analyse non-reactive data on personal recovery in bipolar disorder. Indicating the key areas that people focus on in personal recovery when posting freely and the language they use provides a helpful starting point for formal and informal carers to understand the concerns of people diagnosed with bipolar disorder and to consider how best to offer support

    This fifth volume on Advances and Applications of DSmT for Information Fusion collects theoretical and applied contributions of researchers working in different fields of applications and in mathematics, and is available in open-access. The collected contributions of this volume have either been published or presented after disseminating the fourth volume in 2015 in international conferences, seminars, workshops and journals, or they are new. The contributions of each part of this volume are chronologically ordered. First Part of this book presents some theoretical advances on DSmT, dealing mainly with modified Proportional Conflict Redistribution Rules (PCR) of combination with degree of intersection, coarsening techniques, interval calculus for PCR thanks to set inversion via interval analysis (SIVIA), rough set classifiers, canonical decomposition of dichotomous belief functions, fast PCR fusion, fast inter-criteria analysis with PCR, and improved PCR5 and PCR6 rules preserving the (quasi-)neutrality of (quasi-)vacuous belief assignment in the fusion of sources of evidence with their Matlab codes.     This reprint focuses on the application of the combination of synthetic aperture radars and depth learning technology. It aims to further promote the development of SAR image intelligent interpretation technology. A synthetic aperture radar (SAR) is an important active microwave imaging sensor, whose all-day and all-weather working capacity give it an important place in the remote sensing community. Since the United States launched the first SAR satellite, SAR has received much attention in the remote sensing community, e.g., in geological exploration, topographic mapping, disaster forecast, and traffic monitoring. It is valuable and meaningful, therefore, to study SAR-based remote sensing applications. In recent years, deep learning represented by convolution neural networks has promoted significant progress in the computer vision community, e.g., in face recognition, the driverless field and Internet of things (IoT). Deep learning can enable computational models with multiple processing layers to learn data representations with multiple-level abstractions. This can greatly improve the performance of various applications. This reprint provides a platform for researchers to handle the above significant challenges and present their innovative and cutting-edge research results when applying deep learning to SAR in various manuscript types, e.g., articles, letters, reviews and technical reports

    Development of novel antimalarials targeting the plasmodial lactate transporter (PfFNT) through a fluorescence cross-correlation spectroscopy-based approach and functional assay

    Drug resistance is a significant obstacle in the fight against malaria, prompting the exploration of new treatment approaches and drug targets. One potential target is the recently discovered plasmodial lactate transporter, PfFNT (short for Plasmodium falciparum formate-nitrite transporter). Several studies have shown that inhibiting this transporter can cause the accumulation of toxic levels of lactate within the parasite, leading to its death. Two compounds, BH296 and BH267.meta, have been developed to target both the PfFNT wild type (wt) and a relevant mutant, G107S. So far, these compounds have only been tested using a yeast-based functional assay, and biophysical characterization was missing. In this study, fluorescence cross-correlation spectroscopy (FCCS) measurements were performed to determine true Ki-values, as well as kon and koff rate constants for the binding of inhibitors to both the PfFNT wt and G107S mutant. Once the reliability of FCCS measurements had been confirmed, a compound library of 2,000 inhibitors was screened to identify novel scaffolds that have the potential to inhibit PfFNT. FCCS only allows the search for new inhibitors that displace the existing inhibitor, i.e., share the same binding pocket. To identify an alternative binding site, a functional screening assay with AZD3965 and Metformin was developed in this thesis
