In vitro and in vivo expression of foreign genes by transmissible gastroenteritis coronavirus-derived minigenomes
A helper-dependent expression system based on transmissible gastroenteritis coronavirus (TGEV) has been developed using a minigenome of 3·9 kb (M39). Expression of the reporter gene {beta}-glucuronidase (GUS) (2–8 µg per 106 cells) and the porcine respiratory and reproductive syndrome virus (PRRSV) ORF5 (1–2 µg per 106 cells) has been shown using a TGEV-derived minigenome. GUS expression levels increased about eightfold with the m.o.i. and were maintained for more than eight passages in cell culture. Nevertheless, instability of the GUS and ORF5 subgenomic mRNAs was observed from passages five and four, respectively. About a quarter of the cells in culture expressing the helper virus also produced the reporter gene as determined by studying GUS mRNA production by in situ hybridization or immunodetection to visualize the protein synthesized. Expression of GUS was detected in the lungs, but not in the gut, of swine immunized with the virus vector. Around a quarter of lung cells showing replication of the helper virus were also positive for the reporter gene. Interestingly, strong humoral immune responses to both GUS and PRRSV ORF5 were induced in swine with this virus vector. The large cloning capacity and the tissue specificity of the TGEV-derived minigenomes suggest that these virus vectors are very promising for vaccine development
Positron Escape from Type Ia Supernovae
We generate bolometric light curves for a variety of type Ia supernova models
at late times, simulating gamma-ray and positron transport for various
assumptions about the magnetic field and ionization of the ejecta. These
calculated light curve shapes are compared with light curves of specific
supernovae for which there have been adequate late observations. %The selection
of models is generally not based upon the %ability to fit the late
observations, but rather because the %model has been demonstrated by other
authors to approximate the spectra %and early light curves of that specific SN.
From these comparisons we draw two conclusions: whether a suggested model is
an acceptable approximation of a particular event, and, given that it is, the
magnetic field characteristics and degree of ionization that are most
consistent with the observed light curve shape. For the ten SNe included in
this study, five strongly suggest Co positron escape as would be
permitted by a weak or radially-combed magnetic field. Of the remaining five
SNe, none clearly show the upturned light curve expected for positron trapping
in a strong, tangled magnetic field. Chandrasekhar mass models can explain
normally, sub-, and super- luminous supernova light curves; sub-Chandrasekhar
mass models have difficulties with sub- (and potentially normally) luminous
SNe. An estimate of the galactic positron production rate from type Ia SNe is
compared with gamma-ray observations of Galactic 511 keV annihilation
radiation. Additionally, we emphasize the importance of correctly treating the
positron transport for calculations of spectra, or any properties, of type Ia
SNe at late epochs ( 200 d).Comment: 82 pages including 25 figure

which exhibit the previous best known bound with 100 multiplications
A Highly Ordered Faraday-Rotation Structure in the Interstellar Medium
We describe a Faraday-rotation structure in the Interstellar Medium detected
through polarimetric imaging at 1420 MHz from the Canadian Galactic Plane
Survey (CGPS). The structure, at l=91.8, b=-2.5, has an extent of ~2 degree,
within which polarization angle varies smoothly over a range of ~100 degree.
Polarized intensity also varies smoothly, showing a central peak within an
outer shell. This region is in sharp contrast to its surroundings, where
low-level chaotic polarization structure occurs on arcminute scales. The
Faraday-rotation structure has no counterpart in radio total intensity, and is
unrelated to known objects along the line of sight, which include a Lynds
Bright Nebula, LBN 416, and the star cluster M39 (NGC7092). It is interpreted
as a smooth enhancement of electron density. The absence of a counterpart,
either in optical emission or in total intensity, establishes a lower limit to
its distance. An upper limit is determined by the strong beam depolarization in
this direction. At a probable distance of 350 +/- 50 pc, the size of the object
is 10 pc, the enhancement of electron density is 1.7 cm-3, and the mass of
ionized gas is 23 M_sun. It has a very smooth internal magnetic field of
strength 3 microG, slightly enhanced above the ambient field. G91.8-2.5 is the
second such object to be discovered in the CGPS, and it seems likely that such
structures are common in the Magneto-Ionic Medium.Comment: 16 pages, 5 figures, ApJ accepte
