Autism spectrum traits in normal individuals : a preliminary VBM analysis
In light of the new DSM-5 autism spectrum disorders diagnosis in which the autism spectrum reflects a group of neurodevelopmental disorders existing on a continuum from mild to severe expression of autistic traits, and recent empirical findings showing a continuous distribution of autistic traits in the general population, our voxel based morphometry study compares normal individuals with high autistic traits to normal individuals with low autistic traits. We hypothesize that normal individuals with high autistic traits in terms of empathizing and systemizing [high systemizing (HS)/low empathizing (LE)] share brain irregularities with individuals that fall within the clinical autism spectrum disorder. We find differences in several social brain network areas between our groups. Specifically, we find increased gray matter (GM) volume in the orbitofrontal cortex, the cuneus, the hippocampus and parahippocampus and reduced GM volume in the inferior temporal cortex, the insula, and the amygdala in our HS/LE individuals relative to our HE/LS (low autistic traits in terms of empathizing and systemizing) individuals
From the emotional integration to the cognitive construction: the developmental approach of Turtle Project in children with autism spectrum disorder
Background: Children with autism spectrum disorder show a deficit in neurobiological processes. This deficit
hinders the development of intentional behavior and appropriate problem-solving, leading the child to implement
repetitive and stereotyped behaviors and to have difficulties in reciprocal interactions, empathy and in the
development of a theory of mind. The objective of this research is to verify the effectiveness of a relationship-based
approach on the positive evolution of autistic symptoms.
Method: A sample of 80 children with autism spectrum disorder was monitored during the first four years of
therapy, through a clinical diagnostic assessment at the time of intake and then in two follow-up.
Results: The results showed that through the Autism Diagnostic Observation Schedule it is possible to
assess the socio-relational key elements on which the therapy is based. There was evidence, in fact, of significant
improvements after two and four years of therapy, both for children with severe autistic symptoms and for those in
autistic spectrum.
Conclusions: Socio-relational aspects represent the primary element on which work in therapy with autistic
children and can be considered as indicators of a positive evolution and prognosis that will produce improvements
even in the cognitive are
"Older Adults with ASD: The Consequences of Aging." Insights from a series of special interest group meetings held at the International Society for Autism Research 2016-2017
A special interest group (SIG) entitled "Older Adults with ASD: The Consequences of Aging" was held at the International Society for Autism Research (INSAR) annual meetings in 2016 and 2017. The SIG and subsequent meetings brought together, for the first time, international delegates who were members of the autistic community, researchers, practitioners and service providers. Based on aging autism research that is already underway in UK, Europe, Australia and North America, discussions focussed on conceptualising the parameters of aging when referring to autism, and the measures that are appropriate to use with older adults when considering diagnostic assessment, cognitive factors and quality of life in older age. Thus, the aim of this SIG was to progress the research agenda on current and future directions for autism research in the context of aging. A global issue on how to define 'aging' when referring to ASD was at the forefront of discussions. The ‘aging’ concept can in principle refer to all developmental transitions. However, in this paper we focus on the cognitive and physical changes that take place from mid-life onwards. Accordingly, it was agreed that aging and ASD research should focus on adults over the age of 50 years, given the high rates of co-occurring physical and mental health concerns and increased risk of premature death in some individuals. Moreover, very little is known about the cognitive change, care needs and outcomes of autistic adults beyond this age. Discussions on the topics of diagnostic and cognitive assessments, and of quality of life and well-being were explored through shared knowledge about which measures are currently being used and which background questions should be asked to obtain comprehensive and informative developmental and medical histories. Accordingly, a survey was completed by SIG delegates who were representatives of international research groups across four continents, and who are currently conducting studies with older autistic adults. Considerable overlap was identified across different research groups in measures of both autism and quality of life, which pointed to combining data and shared learnings as the logical next step. Regarding the background questions that were asked, the different research groups covered similar topics but the groups differed in the way these questions were formulated when working with autistic adults across a range of cognitive abilities. It became clear that continued input from individuals on the autism spectrum is important to ensure that questionnaires used in ongoing and future are accessible and understandable for people across the whole autistic spectrum, including those with limited verbal abilities
Assessment of Metabolic Parameters For Autism Spectrum Disorders
Autism is a brain development disorder that first appears during infancy or childhood, and generally follows a steady course without remission. Impairments result from maturation-related changes in various systems of the brain. Autism is one of the five pervasive developmental disorders (PDD), which are characterized by widespread abnormalities of social interactions and communication, and severely restricted interests and highly repetitive behavior. The reported incidence of autism spectrum disorders (ASDs) has increased markedly over the past decade. The Centre for Disease Control and Prevention has recently estimated the prevalence of ASDs in the United States at approximately 5.6 per 1000 (1 of 155 to 1 of 160) children. Several metabolic defects, such as phenylketonuria, are associated with autistic symptoms. In deciding upon the appropriate evaluation scheme a clinician must consider a host of different factors. The guidelines in this article have been developed to assist the clinician in the consideration of these factors

Impaired neurodevelopmental pathways in autism spectrum disorder: a review of signaling mechanisms and crosstalk.
BackgroundThe development of an autistic brain is a highly complex process as evident from the involvement of various genetic and non-genetic factors in the etiology of the autism spectrum disorder (ASD). Despite being a multifactorial neurodevelopmental disorder, autistic patients display a few key characteristics, such as the impaired social interactions and elevated repetitive behaviors, suggesting the perturbation of specific neuronal circuits resulted from abnormal signaling pathways during brain development in ASD. A comprehensive review for autistic signaling mechanisms and interactions may provide a better understanding of ASD etiology and treatment.Main bodyRecent studies on genetic models and ASD patients with several different mutated genes revealed the dysregulation of several key signaling pathways, such as WNT, BMP, SHH, and retinoic acid (RA) signaling. Although no direct evidence of dysfunctional FGF or TGF-β signaling in ASD has been reported so far, a few examples of indirect evidence can be found. This review article summarizes how various genetic and non-genetic factors which have been reported contributing to ASD interact with WNT, BMP/TGF-β, SHH, FGF, and RA signaling pathways. The autism-associated gene ubiquitin-protein ligase E3A (UBE3A) has been reported to influence WNT, BMP, and RA signaling pathways, suggesting crosstalk between various signaling pathways during autistic brain development. Finally, the article comments on what further studies could be performed to gain deeper insights into the understanding of perturbed signaling pathways in the etiology of ASD.ConclusionThe understanding of mechanisms behind various signaling pathways in the etiology of ASD may help to facilitate the identification of potential therapeutic targets and design of new treatment methods
The emotional contagion in children with autism spectrum disorder
Studies of the last decade have demonstrated that children with Autism
Spectrum Disorder (ASD) showed difficulties in language, social and relational
areas, but they had also impairment in the mechanisms of embodied simulation,
namely the imitative behaviors that allow the body to give an experiential
meaning to own and other’s emotions. The identification of this specific emotional
response in ASD children, also defined as emotional contagion, allows to move
the therapeutic focus from reducing the behavioral symptomatic expressions of
the child to promoting the expression of his ability of emotional regulation. The
aim of this study was to investigate the presence of emotional contagion in 53
ASD children aged between 22 and 66 months, through the Test of emotional
contagion and verify the presence of compromised emotional contagion areas.
Our findings have shown that the severity of the disorder is closely related to
the inability of the child to respond to the emotional stimuli, regardless from
cognitive abilities, and that emotion to which children responded most frequently
was happiness, while the one who responded less was anger
Autism, the Integrations of 'Difference' and the Origins of Modern Human Behaviour
It is proposed here that the archaeological evidence for the emergence of 'modern behaviour' (160,000-40,000 bp) can best be explained as the rise of cognitive variation within populations through social mechanisms for integrating 'different minds', rather than by the development of a single 'modern human mind'. Autism and the autistic spectrum within human populations are used as an example of 'different minds' which when integrated within society can confer various selective benefits. It is proposed that social mechanisms for incorporating autistic difference are visible in the archaeological record and that these develop sporadically from 160,000 years bp in association with evidence for their consequences in terms of technological innovations, improved efficiency in technological and natural spheres and innovative thinking. Whilst other explanations for the emergence Of modern human behaviour may also contribute to observed changes, it is argued that the incorporation of cognitive differences played a significant role in the technological, social and symbolic expression of 'modern' behaviour
