Especificación del modelo de habilidades, motivación y oportunidades (AMO) como multiplicativo o aditivo y comprobación de su validez en su aplicación a las prácticas de alta implicación de recursos humanos enfocadas a la mejora continua
Tesis por compendio[ES] El objetivo principal de la presente tesis es dilucidar los aspectos del modelo de habilidades, motivación y oportunidades (modelo AMO) que permitan determinar la relación que existe entre las prácticas de RRHH y la mejora de los resultados a nivel organizacional o individual. El objetivo se vehicula a través de preguntas de investigación específicas. Además, a través de métodos estadísticos evaluamos el alcance y difusión de los sistemas de alta implicación en el contexto laboral español, que posee unas particularidades específicas. Finalmente, investigamos qué modelo de interacción predice mejor la satisfacción laboral a nivel individual, el sumativo o el multiplicativo, lo cual constituye una de las grandes cuestiones no resueltas hasta la fecha.
De nuestra investigación se desprende que el modelo AMO proporciona una estructura apropiada para explorar la relación entre la gestión de RRHH y los resultados a distintos niveles. Muchas investigaciones articulan sus análisis en base a las agrupaciones de prácticas planteadas por el modelo, y numerosos estudios confirman los efectos positivos de los sistemas de alta implicación. Sin embargo, no existe un consenso claro que permita generalizar los resultados y establecer una metodología única. De acuerdo con nuestra investigación, existen dos razones. En primer lugar, hay una gran variedad de enfoques metodológicos, que conducen a resultados muy diversos. Por otro lado, el modelo AMO se integra en un sistema de relaciones complejo, donde además de los mecanismos que moderan las relaciones internas de los sistemas de RRHH y los resultados, operan otros factores organizacionales, personales y externos. Puesto que existen múltiples realidades, definir políticas que garanticen un modelo de gestión único que se adapte a cualquier situación es complejo. Sin embargo, las evidencias demostradas en la literatura juegan sin duda un papel relevante a la hora de tomar decisiones.
Con respecto a la difusión y grado de uso de los sistemas de RRHH en el contexto español, nuestros resultados indican que existe poca influencia de incentivos económicos basados en el rendimiento, quizás debido a la rigidez de la legislación laboral. Los modelos planteados, relacionados con la aplicación formal de sistemas de alta implicación tampoco predicen en gran medida los resultados individuales, aunque sí apuntan a que la categoría de habilidades, y en menor medida la de oportunidades tienen un efecto mayor sobre la satisfacción laboral. Sin embargo, observamos que muchos empleados tienen percepciones positivas en relación con la capacidad de participación, el trabajo en equipo, el intercambio de información, o la formación en el puesto de trabajo, que podrían no estar vinculadas con el diseño y la implantación formal de sistemas de RRHH. En este sentido, algunos estudios evidencian las diferencias entre España y otros países de referencia en relación con la utilización de prácticas de alta implicación.
Esta tesis ofrece diversas contribuciones. En primer lugar, realizamos una síntesis exhaustiva de las características del modelo AMO y sus implicaciones teóricas y prácticas, y señalamos los aspectos que dificultan la generalización de resultados. En segundo lugar, analizamos el grado de uso de los sistemas de alta implicación en el contexto español, señalando las posibles carencias. Nuestras conclusiones pueden fomentar el desarrollo de medidas para incrementar la participación de los empleados, así como para romper las barreras que ralentizan la implantación de los sistemas de alta implicación. Por último, evaluamos la validez de los enfoques sumativo y multiplicativo del modelo para predecir la satisfacción laboral. Contribuimos de este modo al creciente interés por las percepciones de los empleados y los resultados a nivel individual, así como a la conveniencia de explorar enfoques distintos al aditivo para confirmar las supuestas sinergias de los sistemas de alta implicación.[CA] L'objectiu principal de la present tesi és dilucidar els aspectes del model d'habilitats, motivació i oportunitats (model AMO) que permeten determinar la relació que existeix entre les pràctiques de RRHH i la millora dels resultats a nivell organitzacional o individual. Contribuïm d'aquesta manera al creixent interés per les percepcions dels empleats i els resultats a nivell individual, així com a la conveniència d'explorar enfocaments diferents a l'additiu per a confirmar les suposades sinergies dels sistemes d'alta implicació.[EN] The main objective of this thesis is to elucidate the aspects of the abilities, motivation, and opportunities model (AMO model) that allow determining the relationship between HR practices and organizational or individual results improvement. The objective is conveyed through specific research questions. In addition, we assess the scope of high-involvement work systems (HIWS) in the Spanish labour context, which has specific particularities. Finally, we compare the predictive capacity of the summative and multiplicative approaches at the individual level, which is one of the unresolved questions to date.
Our research shows that the AMO model provides an appropriate structure for exploring the relationship between HR management and outcomes at different levels. Many studies articulate their analyses based on the groupings of practices proposed by the model, and numerous studies confirm the positive effects of HIWS. However, there is no clear consensus to generalize the results and establish a single methodology. According to our research, there are two main reasons. First, there is a wide range of methodological approaches, which lead to very mixed results. On the other hand, the AMO model is integrated into a complex system of relationships, where in addition to the mechanisms that moderate the internal relationships of HR systems and the results, other organizational, personal, and external factors that shape these relationships operate. Since there are multiple realities, defining policies guaranteeing a single management model that adapts to any situation is complex. However, the evidence demonstrated in the HR literature undoubtedly plays an important role when making decisions.
We observe several facets regarding the use of HR systems in the Spanish context. Our results indicate little influence of performance-based incentives, perhaps due to the rigidity of labour legislation. Concerning the formal implementation of HIWS, our proposed models do not predict, to a large extent, variations in job satisfaction, although abilities and opportunities categories have greater effects than motivation. Nevertheless, many employees have positive perceptions regarding engagement, teamwork, information sharing, or on-the-job training, which may not be linked to the formal implementation of HR systems. In this sense, some studies show differences between Spain and other reference countries concerning the use of HIWP.
This thesis offers several contributions. First, we offer an exhaustive synthesis of the AMO model features and its theoretical and practical implications, and we point out the aspects that hinder the generalization of results. In-depth knowledge of the model acts as a guide for future research. Secondly, we analyse the degree of use of HIWS in the Spanish context, pointing out the possible shortcomings. Our conclusions may promote the development of policies to increase employee autonomy and participation, job satisfaction, and competitiveness, as well as to break down the institutional and cultural barriers that slow the implementation of HIWS. Finally, we assess the validity of the model's summative and multiplicative approaches to predicting job satisfaction. We thus contribute to the growing interest in employee perceptions and outcomes at the individual level and the desirability of exploring approaches other than additive to confirm the supposed synergies of HIWS.Martínez Tomás, J. COVID-19 Booster Vaccine Acceptance in Ethnic Minority Individuals in the United Kingdom: a mixed-methods study using Protection Motivation Theory
Background: Uptake of the COVID-19 booster vaccine among ethnic minority individuals has been lower than in the general population. However, there is little research examining the psychosocial factors that contribute to COVID-19 booster vaccine hesitancy in this population.Aim: Our study aimed to determine which factors predicted COVID-19 vaccination intention in minority ethnic individuals in Middlesbrough, using Protection Motivation Theory (PMT) and COVID-19 conspiracy beliefs, in addition to demographic variables.Method: We used a mixed-methods approach. Quantitative data were collected using an online survey. Qualitative data were collected using semi-structured interviews. 64 minority ethnic individuals (33 females, 31 males; mage = 31.06, SD = 8.36) completed the survey assessing PMT constructs, COVID-19conspiracy beliefs and demographic factors. 42.2% had received the booster vaccine, 57.6% had not. 16 survey respondents were interviewed online to gain further insight into factors affecting booster vaccineacceptance.Results: Multiple regression analysis showed that perceived susceptibility to COVID-19 was a significant predictor of booster vaccination intention, with higher perceived susceptibility being associated with higher intention to get the booster. Additionally, COVID-19 conspiracy beliefs significantly predictedintention to get the booster vaccine, with higher conspiracy beliefs being associated with lower intention to get the booster dose. Thematic analysis of the interview data showed that barriers to COVID-19 booster vaccination included time constraints and a perceived lack of practical support in the event ofexperiencing side effects. Furthermore, there was a lack of confidence in the vaccine, with individuals seeing it as lacking sufficient research. Participants also spoke of medical mistrust due to historical events involving medical experimentation on minority ethnic individuals.Conclusion: PMT and conspiracy beliefs predict COVID-19 booster vaccination in minority ethnic individuals. To help increase vaccine uptake, community leaders need to be involved in addressing people’s concerns, misassumptions, and lack of confidence in COVID-19 vaccination
Novel Concepts of Mycobacterial Evasion Mechanisms
Mycobacterium tuberculosis -bakteerin aiheuttama tuberkuloosi on tällä hetkellä maailman tappavin infektiotauti. Nykyinen tuberkuloosin antibioottihoito on useamman lääkkeen yhdistelmä ja se kestää useita kuukausia, mutta ei siitäkään huolimatta takaa onnistunutta mykobakteerin häätämistä. Tehottomien antibioottien käyttö sekä pitkät hoitoajat keskeytyksineen edesauttavat antibioottiresistenttien kantojen syntyä. Lisäksi turvallisen ja tehokkaan rokotteen puute tekee tuberkuloosin ehkäisemisestä hankalaa. Yksi syy näihin haasteisiin tuberkuloosin ehkäisemisessä ja hoidoissa on mykobakteerin kyky kehittää krooninen infektio ja suojella itseään ympäristöhaitoilta kuten aktiiviselta immuunivasteelta sekä antimikrobiaalisilta lääkkeiltä.
Tämän tutkimuksen tavoitteena on tutkia mykobakteerin mekanismeja, joiden avulla se pystyy estämään antibioottien ja immuunivasteiden tehokkaan toiminnan, sekä etsiä uusia strategioita näihin mekanismeihin puuttumiseen. Mycobacterium marinum -bakteeria ja seeprakalaa käytettiin tutkimaan isännän ja patogeenin välistä vuorovaikutusta. Mykobakteeri kykenee aktiivisesti vaimentamaan immuunivasteita ja myöhästyttämään hankittujen immuunivasteiden alkamista. Tutkimuksessa testattiin immunomodulaation tehoa näiden mykobakteerin ominaisuuksien estämisessä. Ilman hoitoja noin 10 % aikuisista seeprakaloista pystyy luonnollisesti pitämään mykobakteerimäärän alle qPCR:n detektiorajan. Tätä osuutta voitiin kuitenkin nostaa 25 %:iin aktivoimalla immuunivaste lämpötapetulla Listeria monocytogenes -bakteerilla ennen mykobakteeri-infektiota. Aikaansaatu suojaava immuunivaste vähensi merkitsevästi mykobakteerimääriä mitattuna neljä viikkoa infektion jälkeen ja jopa steriloi mykobakteerin osasta populaatiota sekä villityypin kalassa että rag1-/- -mutanttikaloissa, mikä viittaa synnynnäisten immuunivasteiden tärkeään rooliin mykobakteerin steriloivassa vasteessa. Suojaavassa vasteessa mpeg1, tnf ja nos2b -geenien ilmentyminen oli merkitsevästi lisääntynyt, kun taas sod2 oli vähentynyt verrattuna kontrolliryhmään. Nämä tulokset viittaavat mykobakteerin häätämiseen infektion varhaisessa vaiheessa lisääntyneen tulehdusvasteen ja tehokkaamman mykobakteerin tuhoamisen avulla. Huomattavaa on, että immunolodulaatio lämpötapettua L. monocytogenes -bakteeria hyödyntäen ei saanut aikaan suojaavaa vastetta, jos infektio oli jo olemassa. Vastaavaa immuuniterapiaa voitaisiin kuitenkin käyttää ennaltaehkäisevänä menetelmänä korkean tuberkuloosi- ilmaantuvuuden alueilla.
Toinen tutkittu mykobakteerin suojautumiskeino on biofilmin muodostaminen. Immuunivasteiden heikentämisen ja infektion aikaansaamisen jälkeen mykobakteeri muodostaa bakteeriyhteisöjä granuloomissa ja tuottaa solunulkoista matriksia, joka antaa bakteeripopulaatiolle tukea ja suojaa. Nopeakasvuista bioluminoivaa M. marinum -bakteeria käytettiin biofilmin kypsymisen ja koostumuksen tutkimuksessa in vitro. Tulokset osoittivat, että biofilmin kypsyminen ei muuttanut rifampisiinin MIC-arvoa (minimum inhibitory concentration), mutta nosti MBC-arvoa (minimum bactericidal concentration) 63-kertaisesti verrattuna planktoniseen yksisoluisena kasvavaan mykobakteeriin kahden päivän biofilmin kasvattamisen jälkeen. Toistuvilla antibioottialtistuksilla pystyttiin varmistamaan, että biofilmin lisääntynyt antibioottitoleranssi ei johtunut geneettisestä resistenssistä. Elävien mykobakteerien lukumäärää biofilmissä pystyttiin vähentämään hajottamalla solunulkoisen matriksin tärkeimpiä polymeerejä yhdessä rifampisiini-hoidon kanssa, mikä osoitti biofilmin matriksin tärkeyden antibioottitoleranssissa.
Nopea bioluminesenssimittaukseen perustuva tappokäyräanalyysi validoitiin mittaamaan erityisesti persistoivien mykobakteerien alapopulaatiota. Analyysissa seurataan bakteeripopulaation tappokinetiikkaa korkealla antibioottikonsentraatiolla, ja voidaan mitata persisteribakteerien alapopulaatiota, joka kykenee sietämään antibioottia keskimääräistä pidemmän ajan. Tappokinetiikka oli merkitsevästi hitaampi yhden viikon ikäisellä M. marinum biofilmillä verrattuna planktoniseen yksisoluiseen kasvatukseen. Lisäksi rifampisiinin konsentraatio saturoitui 400 μg/ml:ssa, jonka jälkeen antibioottikonsentraation nostaminen ei nopeuttanut bakteerien tappoa. Tätä menetelmää on mahdollista käyttää seulomaan hoitoja, jotka vaikuttavat erityisesti persistoiviin soluihin.
Lopuksi tutkittiin mykobakteerin biofilmin solunulkoisen matriksin proteiineihin, GroEL1 ja GroEL2, kohdentuvia sybodeja in vitro M. marinum ja M. tuberculosis biofilmeissä sekä ex vivo seeprakalan granuloomissa. Sybodit ovat synteettisiä vasta- aineen sitoutumisosaa matkivia molekyylejä, jonka sitoutumista voidaan helposti muokata ja seuloa halutun kohteen mukaan olemassa olevista sybody-kirjastoista. Seulonnasta saatujen potentiaalisten sybodien sitoutumista mykobakteerin biofilmiin arvioitiin konfokaalimikroskopian avulla. Fluoresenssileimatut anti-GroEL-sybodit tunnistivat GroEL-proteiineja biofilmin päältä sekä in vitro että ex vivo tehden sybodista houkuttelevan vaihtoehdon erilaisten molekyylien kohdentamisessa mykobakteerin biofilmeihin. Nykyisiä antibioottihoitoja voidaan mahdollisesti tehostaa ja hoitojen pituutta lyhentää ottamalla huomioon granuloomien sisällä biofilmeissä olevat mykobakteeripersisterit. Ymmärtämällä paremmin mykobakteerin menetelmiä, joilla se suojelee itseään ympäristön haittavaikutuksilta, tulevaisuuden hoitoja voidaan kohdentaa tehokkaammin Mycobacterium tuberculosis -bakteeria vastaan.Tuberculosis is caused by bacterium Mycobacterium tuberculosis, and it is currently the deadliest infectious disease worldwide. The standard antimicrobial regimen is a cocktail of antimicrobials taken for several months, which still does not guarantee a successful eradication of the bacteria. Treatment dropouts and the use of antimicrobials that are inefficient in eradicating the infection have led to emerged drug-resistance. Also, the lack of safe and effective vaccine makes the prevention of tuberculosis difficult. One reason for the challenges in both prevention and treatment of tuberculosis is the ability of mycobacterium to cause chronic infection and protect itself from environmental hazards such as active immune responses and antimicrobials.
The aim of this study was to examine mycobacterial evasion mechanisms against antimicrobials and immune responses and propose new intervention strategies. Mycobacterium marinum and zebrafish were used to study the interaction of the host and the pathogen. Mycobacterium can actively suppress immune responses and delay the onset of adaptive responses, and to prevent this immune evasion, an immunomodulation was tested. In the adult zebrafish population, around 10% of the population is able to naturally retain the M. marinum load below the detection limit of qPCR. This proportion could be increased up to 25% by priming with heat- killed Listeria monocytogenes prior the mycobacterial infection. The induced protective immune response significantly reduced mycobacterial loads after four weeks of infection and induced clearance in both wildtype and rag1-/- mutant fish showing the important role of innate immune responses in the sterilizing response. The protective immune response was characterized with an increased expression of mpeg1, tnf and nos2b and decreased expression of sod2. These results indicate early clearance mediated via pro-inflammatory responses and enhanced killing of mycobacteria. Importantly, the immunomodulation was ineffective if the infection was already established. However, a similar approach could be used as a prophylactic treatment in high burden areas.
Another mycobacterial evasion strategy is the formation of biofilms. After suppressing immune responses and establishing the infection, mycobacteria form bacterial communities in granulomas and produce extracellular matrix that gives structure and protection for the bacterial population. The fast-growing M. marinum with a bioluminescent cassette was used to study the biofilm maturation and composition in vitro. The results showed that the biofilm maturation did not alter the minimum inhibitory concentration (MIC) but increased minimum bactericidal concentration (MBC) 63 times compared to planktonic cells within two days of biofilm culturing. It was further confirmed with repeated antimicrobial exposures that the increased tolerance of biofilm cultures was not due to genetic resistance. Degrading any of the major extracellular matrix components in combination with rifampicin reduced the number of live bacteria in a biofilm, demonstrating the important role of the biofilm matrix in the antimicrobial tolerance.
Time-kill curve analysis with a quick bioluminescence read-out was established to specifically measure the subpopulation of persister mycobacteria. In the analysis, the killing kinetics of a bacterial population was followed after the exposure to high concentration of a bactericidal antimicrobial, here rifampicin, to measure the persister subpopulation that can tolerate the drug for a prolonged time. For maturing
M. marinum biofilm, the killing kinetics were significantly different after one week of culturing compared to planktonic cells, and the rifampicin concentration was saturated at 400 μg/ml after which increasing the antimicrobial concentration did not alter the killing kinetics. This method has a potential in screening for treatments that specifically target mycobacterial persisters.
Finally, sybodies against biofilm extracellular matrix proteins GroEL1 and GroEL2 were used to specifically target M. marinum and M. tuberculosis biofilms in vitro and in ex vivo zebrafish granulomas. Sybodies are synthetic molecules mimicking the binding domain of antibodies. The binding properties of the sybodies can be easily modified and screened against the wanted target in sybody libraries. The potential sybodies acquired from the screens were assessed by confocal imaging. Fluorescence labelled anti-GroEL sybodies were able to bound GroEL on both in vitro and ex vivo biofilms making sybodies an attractive molecular carrier to target mycobacterial biofilms. A treatment that specifically targets mycobacterial persisters in biofilms inside granulomas could enhance the efficacy of antimicrobial therapy and shorten the current treatment time. Psychiatric sequelae of adolescent cannabis use in the Northern Finland 1986 Birth Cohort
Adolescence is a time of significant brain development, and this maturation process is thought to represent a time of heightened vulnerability to the adverse effects of environmental risk factors such as cannabis. While the association of adolescent cannabis use and subsequent psychosis and depression has received considerable attention in the existing literature, very few prospective studies have focused on the association of early cannabis exposure with severe self-harm or suicide, bipolar disorder, or anxiety disorders. Moreover, though there are reports focusing on the prognostic effect of cannabis use in patients at high risk for psychosis, populationbased studies examining the prognosis of non-treatment-seeking adolescents with both psychotic-like experiences (PLEs) and cannabis exposure are scarce.
The aim of this thesis was to examine the association of early cannabis use with severe self-harm requiring medical attention, bipolar disorder, and depressive or anxiety disorders (studies I–III). Secondly, the prognosis of adolescents with PLEs with or without cannabis exposure was examined with respect to several psychiatric sequelae (study IV). A prospective general population-based Northern Finland 1986 Birth Cohort (N=9432) was utilized. Data on substance use including cannabis use and early psychopathology including PLEs were gathered in a field study in 2001– 02 when the participants were aged 15–16 years. The participants were followed for 18 years (until the year 2018, when they were 33 years). Data on diagnoses made in clinical practice from nationwide registers were utilized as outcome measures.
Early cannabis use was found to be associated with severe self-harm (Study I). While an independent association of this exposure was seen with depressive/anxiety disorders (Study II), the association with a bipolar disorder attenuated to nonsignificance after adjusting for types of other substance use (Study III). In study IV, participants with both PLEs and early cannabis exposure were found to display a greater odds for adverse psychiatric sequelae than participants with only PLEs.
In summary, the findings support the proposal that early cannabis use is an adverse prognostic marker for other psychiatric sequelae in addition to psychosis.Nuorten kannabiksen käytön mielenterveydellinen ennuste Pohjois-Suomen 1986 syntymäkohortissa
Nuoren kehittyvien aivojen arvellaan olevan erityisen haavoittuvia kannabiksen kaltaisille ympäristöriskitekijöille. Kannabiksen käytön ja psykoosin välistä yhteyttä on tutkittu monimuotoisesti, mutta varhaisen kannabikselle altistumisen ja itsensä vahingoittamisen, kaksisuuntaisen mielialahäiriön sekä ahdistuneisuushäiriöiden välistä yhteyttä on tutkittu merkitsevästi tätä vähemmän. Lisäksi siinä missä kannabiksen ennustevaikutusta psykoosiriskissä olevilla potilailla on jonkin verran tietoa, lievempien psykoosin kaltaisten oireiden ja samanaikaisen kannabiksen käytön mielenterveydellistä ennustetta nuorilla ei ole juuri kartoitettu yleisväestöpohjaisissa pitkittäistutkimuksissa.
Väitöskirjatyön tarkoitus oli tutkia varhaisen kannabiksen käytön ja vakavan itsensä vahingoittamiskäyttäytymisen, kaksisuuntaisen mielialahäiriön ja masennusja ahdistuneisuushäiriöiden välistä yhteyttä (tutkimukset I–III). Lisäksi varhaisen kannabiksen käytön vaikutusta mielenterveydelliseen ennusteeseen tutkittiin nuorilla, joilla oli psykoosin kaltaisia kokemuksia ja varhaista kannabiksen käyttöä. Aineistona käytettiin yleisväestöön pohjautuvaa Pohjois-Suomen 1986 syntymäkohorttia (N=9432). Kannabiksen ja muiden päihteidenkäyttöä ja psykoosin kaltaisia kokemuksia kartoitettiin kenttätutkimuksessa vuosina 2001–02 nuorten ollessa 15– 16-vuotiaita. Tutkittavia seurattiin 18 vuoden ajan (vuoteen 2018 ja 33 vuoden ikään asti). Vastemuuttujat muodostettiin kansallisista rekistereistä saatavista diagnoositiedoista.
Nuoruusiän Varhainen kannabiksen käyttö oli yhteydessä vakavaan itsensä vahingoittamiskäyttäytymiseen (tutkimus I). Yhteys nuoruusiän kannabiskäytön ja masennus- ja ahdistuneisuushäiriöiden välillä havaittiin (tutkimus II), mutta yhteys kaksisuuntaiseen mielihäiriöön ei jäänyt tilastollisesti merkitseväksi monimuuttujamalleissa (tutkimus III), kun muu päihdekäyttö otettiin huomioon. Sekä psykoosin kaltaisia kokemuksia että kannabiksen käyttöä raportoineilla nuorilla oli suurempi riski erilaisiin mielenterveyshäiriöihin kuin niillä nuorilla, joilla oli psykoosin kaltaisia kokemuksia ilman varhaista kannabiksen käyttöä.
Tutkimuslöydökset viittaavat siihen, että varhainen kannabiksen käyttö on riskitekijä myöhemmille mielenterveyden häiriöille
